Developmental and Feedforward Control of the Expression of Folate Biosynthesis Genes in Tomato Fruit

Little is known about how plants regulate their folate content, including whether the expression of folate biosynthesis genes is orchestrated during development or modulated by folate levels. Nor is much known about how folate levels impact the expression of other genes. These points were addressed using wild-type tomato fruit and fruit engineered for high folate content. In wild-type fruit, the expression of genes specifying early steps in folate biosynthesis declined during development but that of other genes did not. In engineered fruit overexpressing foreign GTP cyclohy- drolase I and aminodeoxychorismate synthase genes, the expression of the respective endogenous genes did not change, but that of three downstream pathway genes--aminodeoxychorismate lyase, dihydroneopterin aldolase, and mitochondrial folylpolyglutamate synthase--respectively increased by up to 7.8-, 2.8-, and 1.7-fold, apparently in response to the build-up of specific folate pathway metabolites. These results indicate that, in fruit, certain folate pathway genes are developmentally regulated and that certain others are subject to feedforward control by pathway intermediates. Micro- array analysis showed that only 14 other transcripts （of 11 000 surveyed） increased in abundance by two-fold or more in high-folate fruit, demonstrating that the induction of folate pathway genes is relatively specific.

Dietary protein(P)and carbohydrate(C)have a major impact on the sweet taste ***,it remains unclear whether the balance of P and C influences the sweet taste ***,we use the nutritional geometry framework(NGF)to address the interaction of protein and carbohydrates on sweet taste using Drosophila as a *** results reveal that high-protein,low-carbohydrate(HPLC)diets sensitize to sweet taste and low-protein,high-carbohydrate(LPHC)diets desensitize sweet taste in both male and female *** further investigate the underlying mechanisms of the effects of two diets on sweet taste using RNA *** compared to the LPHC diet,the m RNA expression of genes involved in the metabolism of glycine,serine,and threonine is significantly upregulated in the HPLC diet group,suggesting these amino acids may mediate sweet taste *** further find that sweet sensitization occurs in flies fed with the LPHC diet supplemented with serine and *** study demonstrates that sucrose taste sensitivity is affected by the balance of dietary protein and carbohydrates possibly through changes in serine and threonine.

NARCCAP Model Skill and Bias for the Southeast United States

This paper investigates dynamically downscaled regional climate model (RCM) output from the North American Regional Climate Change Assessment Program (NARCCAP) for two sub-regions of the Southeast United States. A suite of four statistical measures were used to assess model skill and biases were presented in hindcasting daily minimum and maximum temperature and mean precipitation during a historical reference period, 1970-1999. Most models demonstrated high skill for temperature during the historical period. Two outliers included two RCMs run using the Geophysical Fluids Dynamics Lab (GFDL) model as their lateral boundary conditions;these models suffered from a cold maximum temperature bias. Improvement with GFDL-based projections of maximum temperature was noted from May through November when they ran with observed seasurface conditions (GFDL-timeslice), particularly for the east sub-region. Precipitation skill proved mixed-relatively high when measured using a probability density function overlap measurement or the index of agreement, but relatively low when measured with root-mean square error or mean absolute error, because several models overestimated the frequency of extreme precipitation events.

The zoonotic cryptosporidiosis is globally distributed,one of the major diarrheal diseases in humans and *** oocysts are also one of the major environmental concerns,making it a pathogen that fits well into the One Health *** its importance,fully effective drugs are not yet ***-cryptosporidial drug discovery has historically faced many unusual challenges attributed to unique parasite biology and technical *** significant progresses have been made recently,anti-cryptosporidial drug discovery still faces a major obstacle:identification of systemic drugs that can be absorbed by patients experiencing watery diarrhea and effectively pass through electron-dense(ED)band at the parasite-host cell interface to act on the epicellular *** may be a need to develop an in vitro assay to effectively screen hits/leads for their capability to cross ED *** the meantime,non-systemic drugs with strong mucoadhesive properties for extended gastrointestinal exposure may represent another direction in developing anti-cryptosporidial *** developing both systemic and non-systemic drugs,a non-ruminant animal model exhibiting diarrheal symptoms suitable for routine evaluation of drug absorption and anti-cryptosporidial efficacy may be very helpful.

对于外文化者而言,中国的内文化是极其复杂且难于理解的。中国是有史以来人类最早的文明发源地之一;然而自19世纪以来,中国也在发生了着深刻的文化变化。因此,当前文化研究对如何准确地描绘当今中国文化现状,描绘中国文化的变化方向,阐释变化产生的原因,仍存在不小的困难。很多学者认为,日益增多的西方文化产品和文化信息输入导致中国的年轻一代正朝着个人主义价值取向转变。但是,此类观点恰好与同样简单化标签化地归纳中国文化为“集体主义”的论断是相互冲突的。因此,基于一系列扎根理论的研究,本文采用了主位研究的方法,聚焦于在中国90后群体中极为盛行的概念“梦想”,分析其背后所蕴藏的文化价值观变化。具体而言,本文旨在探索目前在中国年轻一代流行的“梦想”是否是全然来自于西方的概念;进而探讨中国90后的文化价值观发生了何种变化,而这些文化价值观变化是否可以映射“梦想”这一概念的渗透。研究发现,尽管大多数受访者的梦想的确是是纯粹地源于个人兴趣爱好,但他们对“梦想”地追逐与个人主义价值观的增强之间并没有必然联系。确切地说,通过对中国当代青年对“梦想”的描述进行分析,研究揭示了四个理想结果价值观与三个过程价值观,一共七种价值观;他们部分与中国传统价值观吻合,而部分又与现代价值观或西方价值观契合。其中最引人注意的发现在于,在中国90后眼中,“梦想”是一个纯粹的本土概念。通过大量量化研究,多维度以及价值观研究,本文为了解中国年轻一代的多重价值观提供了丰富而深刻的见解,为学者向世界阐述“新”中国提供了全新的视角。

Site-controlled formation of InGaAs quantum nanostructures——Tailoring the dimensionality and the quantum confinement

we report on InGaAs quantum disks （QDks） controllably formed on the top （001） facet of nano-patterned GaAs pyramidal platforms. The QDks exhibit pyramidal shape with special facets and varied dimensions, depending on the GaAs pyramidal buffer and the amount of InGaAs deposited. The formation of QDks is explained by the overgrowth of an InGaAs layer and thereafter coalescence of small InGaAs islands. Photoluminescence （PL） characteristics of ensemble QDks and exciton features of individual QDks together demonstrate that we may achieve a transition from zero-dimensional （0D） to two-dimensional （2D） quantum structure with increasing QDk size. This transition provides the flexibility to continuously tailor the dimensionality and subsequently the quantum confinement of semiconductor nanostructures via site-controlled self-assembled epitaxy for device applications based on single quantum structures.
