We present an analysis of 288 young stellar objects(YSOs)in the Perseus molecular cloud that have well defined g and r-band lightcurves from the Zwicky Transient *** the 288 YSOs,238 sources(83%of our working sample)are identified as variables based on the normalized peak-to-peak variability metric,with variability fraction of 92%for stars with disks and 77%for the diskless *** variables are classified into different categories using the quasiperiodicity(Q)and flux asymmetry(M)***-three variables are classified as strictly periodic objects that are well phased and can be attributed to spot modulated stellar *** also identify 22 bursters and 25 dippers,which can be attributed to accretion burst and variable extinction,*** with disks tend to have asymmetric and non-repeatable lightcurves,while the YSOs without disks tend to have(quasi)periodic *** periodic variables have the steepest change in g versus g-r,while bursters have much flatter changes than dippers in g versus *** and quasiperiodic variables display the lowest variability *** models suggest that the variability amplitudes of periodic variables correspond to changes of the spot coverage of 30%-40%,burster variables are attributed to accretion luminosity changes in the range of l_(acc)/l_(*)=0.1-0.3,and dippers are due to variable extinction with A_(V)changes in the range of 0.5-1.3 mag.

Bacterial adherence to the acquired dental pellicle, important in dental caries (caries), is mediated by receptor-adhesins such as salivary agglutinin binding to Streptococcus mutans antigen I/II (I/II). Ten selected I/II epitopes were chosen to determine their reactivity to human salivary IgA. Previous studies suggested that a specific HlA biomarker group (HlA-DRB1*04) may have differential influence of immune responses to I/II. However, it was not known whether secretory IgA (SIgA) responses to the selected epitopes from HlA-DRB1*04 positive subjects were different compared to controls, or across other caries-related factors such as total IgA (TIgA). Thirty-two total subjects were matched according to HlA type, gender, ethnicity and age. HlA genotyping, oral bacterial, immunoglobulin and antibody analyses were performed. A large observed difference emerged with regard to the natural immune reservoir of TIgA in HlA-DRB1*04 positive subjects, specifically, a 27.6% reduction compared to controls. In contrast to all other epitopes studied, HlA-DRB1*04 positive subjects also exhibited reduced reactivity to I/II epitope 834-853. HlA-DRB1*04 positive subjects exhibited lower specific SIgA activity/TIgA to 834-853 and also a lower specific reactivity to 834-853/whole cell S. mutans UA159. Furthermore, HlA-DRB1*04 positive subjects exhibited lower responses to I/II in its entirety. The large observed difference in TIgA and the 834-853 reactivity pattern across multiple measures suggest potentially important connections pertaining to the link between HlA-DRB1*04 and caries.

Muscle RING-finger protein-1 (MuRFl) functions and cellular localization are regulated by SUMOl post-translational modification

The muscle RING-finger protein-1 (MuRFl) is an E3 ubiquitin ligase expressed in skeletal and cardiac muscle tissues and it plays important roles in muscle remodeling. Upregulation of MuRFl gene transcription participates in skeletal muscle atrophy, on contrary downregulation of protein expression leads to cardiac hypertrophy. MuRFl gene point mutations have been found to generate protein aggregate myopathies defined as muscle disorder characterized by protein accumulation in muscle fibers. We have discovered that MuRFl turned out to be also a target for a new post-translational modification arbitrated by conjugation of SUM01 and it is mediated by the SUMO ligases E2 UBC9 and the E3 PIASy/4. SUMOylation takes place at lysine 238 localized at the second coiled-coil protein domain that is required for efficient substrate interaction for polyubiquitination. We provided evidence that SUMOylation is essential for MuRFl nuclear translocation and its mitochondria accumulation is enhanced in hyper? glycemic conditions delivering a stabilization of the overall SUMOylated proteins in cultured myocytes. Thus, our findings add this SUM01 post-translational modification as a new concept to understand muscle disorders related to the defect in MuRFl activity.

Purpose: To report the use of commercially available preservative-free intrav itreal triamcinolone acetonide for the treatment of macular oedema due to retina l vascular diseases. Design: Retrospective interventional case series. Methods: Charts of eyes that received 4 mg preservative-free intravitreal triamcinolone acetonide for the treatment of persistent macular oedema due to retinal vascular diseases were reviewed. Patients were included if they had a follow-up of at least 3 months. Visual acuity, intraocular pressure, presence of an anterior cha mber reaction, and mean macular thickness on optical coherence tomography (OCT) were recorded. Results: A total of 10 eyes of 10 patients were identified. Visua l acuity improved by a mean of 1.1 Suellen lines at 1 month and 1.3 lines at 3 m onths. Macular thickness on OCT decreased by a mean of 183.5 μm at 1 month (Plar pressure increased from a mean of 13.5 mmHg at baseline to 15.3 at 1 month, and 14.5 at 3 months. Only the 1-month change in intraocular p ressure was statistically significant (P=0.0274). There were no cases of endopht halmitis, anterior chamber reaction, or retinal detachment. Conclusion: In this small retrospective, noncomparative series, commercially available preservative -free intravitreal triamcinolone acetonide had no adverse outcomes. Macular oed ema was noted to decrease following treatment.

This review considers available evidence for mechanisms of conferred adaptive advantages in the face of specific infectious *** short,we explore a number of genetic conditions,which carry some benefits in adverse circumstances including exposure to infectious *** examples discussed are conditions known to result in resistance to a specific infectious disease,or have been proposed as being associated with resistance to various infectious *** infectious diseasedgenetic disorder pairings include malaria and hemoglobinopathies,cholera and cystic fibrosis,tuberculosis and Tay-Sachs disease,mycotic abortions and phenylketonuria,infection by enveloped viruses and disorders of glycosylation,infection by filoviruses and NiemannePick C1 disease,as well as rabies and myasthenia *** also discuss two genetic conditions that lead to infectious disease hypersusceptibility,although we did not cover the large number of immunologic defects leading to infectious disease *** of the resistance-associated pairings(malaria/hemogloginopathies,cholera/cystic fibrosis,tuberculosis/Tay-Sachs,and mycotic abortions/phenylketonuria)appear to be a result of selection pressures in geographic regions in which the specific infectious agent is *** other pairings do not appear to be based on selection pressure and instead may be ***,research investigating these relationships may lead to treatment options for the aforementioned diseases by exploiting established mechanisms between genetically affected cells and infectious *** may prove invaluable as a starting point for research in the case of diseases that currently have no reliably curative treatments,e.g.,HIV,rabies,and Ebola.

Chronic intestinal failure(CIF)is a rare but feared complication of Crohn’s *** on the remaining length of the small intestine,the affected intestinal segment,and the residual bowel function,CIF can result in a wide spectrum of symptoms,from single micronutrient malabsorption to complete intestinal *** of CIF has improved significantly in recent *** in home-based parenteral nutrition,in particular,have translated into increased survival and improved quality of ***,60%of patients are permanently reliant on parenteral *** results with new drugs such as teduglutide have added a new dimension to CIF *** outcomes of patients with CIF could be greatly improved by more effective prevention,understanding,and *** complex cases,the care of patients with CIF requires a multidisciplinary approach involving not only physicians but also dietitians and nurses to provide optimal intestinal rehabilitation,nutritional support,and an improved quality of ***,we summarize current literature on CIF and short bowel syndrome,encompassing epidemiology,pathophysiology,and advances in surgical and medical management,and elucidate advances in the understanding and therapy of CIF-related complications such as catheter-related bloodstream infections and intestinal failure-associated liver disease.

Here we discuss recent progress in understanding tropical cyclone(TC)subseasonal variability and its *** has been a concerted effort to understand the sources of predictability at subseasonal time-scales,and this effort has continued to make progress in recent *** the Madden-Julian Oscillation(MJO),other modes of variability affect TCs at these time-scales,in particular various equatorial ***,TC activity is also modulated by extratropical processes via Rossby wave *** has also been progress in the ability of models to simulate the MJO and its modulation of TC *** efforts have created multi-model ensemble datasets,which have made it possible to evaluate the forecast skill of the MJO and TCs on subseasonal time-scales in multiple forecasting *** there is positive skill in some cases,there is strong dependence on the ensemble system considered,the basin examined,and whether the storms have extratropical influences or ***,the definition of skill differs among *** centers are currently issuing subseasonal TC forecasts using various techniques(statistical,statistical-dynamical and dynamical).There is also a strong interest in the private sector for forecasts with 3-4 weeks lead time.
