Mass Spectrometric Evidence of Heparin Disaccharides for the Catalytic Characterization of A Novel Endolytic Heparinase

Heparinase from different sources can eliminate heparin or/and heparan sulfate into various low-molecular weight heparins with different characteristics. Porcine intestinal mucosa heparin was de- graded into a series of oligosaccharides by a novel heparinase from the species Sphingobacterium. Disaccha- ride components from the digests were separated and purified by ultrafiltration and HPLC. Five major peaks appeared as three types according to their retention time. The mass spectrometry of peak I mainly gave the non-sulfated disaccharide with the mass of 379 Da. Peak II and III were indicated as two major monosulfated disaccharides with molecular mass of 417 and 459 Da respectively. Moreover, the peak III represented an N- acetyl disaccharide. Both peak IV and V showed the same mass of 496 Da, hinting that they were disulfate- substituted disaccharides. No trisulfate-substituted disaccharides were detected in the mixture of the heparin digest though they were abundant in the heparin structure. The results revealed that the heparinase might specifically cut the sites with low sulfated domain in heparin.

Possible Energy Network with polygeneration system and CCS for china

On basis of adopting polygeneration systems for power and alternative fuels,capturing CO2.with near zero energy penalties,and storing CO2.on sites,a new kind of Energy Network can integrate energy utilization,CO2.capture,transportation and storage ***-economic analysis of this solution focusing on Inner Mongolia and the Yangtze River Delta districts had been carried with comparison to the chain method for energy utilization and CO2.*** solution can save 2..5% of energy,and reduce 35% of total *** adoption of advanced polygeneration systems contributes 52.2. of the total saved costs,and the integration of energy utilization and CO2.sequestration in the Energy Network contributes 47.8%.From the aspect of CCS,the CO2.sequestration cost in the Energy Network can be as low as 12.$/t due to the lower energy penalties of capture in polygeneration systems and the combination of CO2.source and *** Energy Network exhibits attractive performance on energy saving,costs reduction for CCS,which may be a promising solution for sustainable development of china.

It is always difficult to extract population in-formation of small watershed during the region environment assessment. The report adopted compound areal interpola-tion to study the population in Chaohu watershed with the GIS technique and landuse interpretation data from remote sensing. The result indicated that the method is effective to extract population information of small watershed.

Characterization of a sodium-regulated glutaminase from cyanobacterium Synechocystis sp. PCC 6803

Glutaminase is widely distributed among microorganisms and mammals with important functions. Lit-tle is known regarding the biochemical properties and functions of the deamidating enzyme glutami-nase in cyanobacteria. In this study a putative glutaminase encoded by gene slr2.79 in Synechocystis sp. PCC 6803 was investigated. The slr2.79 was expressed as histidine-tagged fusion protein in Es-cherichia coli. The purified protein possessed glutaminase activity, validating the functional assign-ment of the genomic annotation. The apparent Km value of the recombinant protein for glutamine was 2..6 ± 0.9 mmol/L, which was comparable to that for some of other microbial glutaminases. Analysis of the purified protein revealed a two-fold increase in catalytic activity in the presence of 1 mol/L Na+. Moreover, the Km value was decreased to 12.2.± 1.9 mmol/L in the presence of Na+. These data demon-strate that the recombinant protein Slr2.79 is a glutaminase which is regulated by Na+ through in-creasing its affinity for substrate glutamine. The slr2.79 gene was successfully disrupted in Synecho-cystis by targeted mutagenesis and the △slr2.79 mutant strain was analyzed. No differences in cell growth and oxygen evolution rate were observed between △slr2.79 and the wild type under standard growth conditions, demonstrating slr2.79 is not essential in Synechocystis. Under high salt stress condition, however, △slr2.79 cells grew 1.2.-fold faster than wild-type cells. Moreover, the photosyn-thetic oxygen evolution rate of △slr2.79 cells was higher than that of the wild-type. To further charac-terize this phenotype, a number of salt stress-related genes were analyzed by semi-quantitative RT-PCR. Expression of gdhB and prc was enhanced and expression of desD and guaA was repressed in △slr2.79 compared to the wild type. Identification of the Angel -related elements in cyprinid fishes and their phylogenetic implications

Angel-related element belongs to the family of miniature inverted-repeat transposable elements (MITEs). In this paper we report the identification of an Angel- related element in the series Leuciscini of cyprinid fishes, which is located in the second intron of the growth hormone (GH) gene. We have also found that this element is absent in orthologous locus in the series Barbini of cyprinid fishes, that provides new evidence for the monophyly of the series Leuciscini. The insertion of Angel -related element into the GH gene might take place in the common ancestor of the series Leuciscini after its divergence from the series Barbini. The high sequence divergence and relatively broad species distribution of Angel-related elements implies that they might be ancient transposons which appeared about 2. million years ago.

Relationships between tree-ring width index and NDVI of grassland in Delingha

Using five well-replicated Qilian juniper (Sabina przewalskii Kom.) tree-ring width index se- ries, monthly normalized difference vegetation index (NDVI) of grassland, and climatic data from 1982.to 2.01, the relationships between tree-ring width index, NDVI of grassland, and climatic data were analyzed firstly. Then, the relationship between tree-ring width index and NDVI of grassland was explored. The re- sults showed that: (1) Temperature and precipitation in June influenced tree-ring width index and NDVI of grassland deeply in Delingha. (2. There were sig- nificant relationships between five tree-ring width index series (DLH1-DLH5) and monthly NDVI of grassland from June to September, with the most significant relationship being between tree-ring width index series and NDVI of grassland in August. (3) The PC1 (the first principal component derived from DLH1-DLH5 series) exhibited good agreement with monthly NDVI of grassland in the grass growth sea- son (from June to September) and the averaged NDVI in the growth season, which was attributed to their common responses to water-supply limit in Delingha. This study may allow an increase in studying the past dynamics of grassland in Delingha in that the variation of grassland NDVI during the last millennium has been reconstructed from PC1.

Artificial coordinating field and its application to motion planning of robots in uncertain dynamic environments

Artificial coordinating fields (ACF) are proposed to deal with the motion planning problems of mobile robots in uncertain dynamic environments. An ACF around an obstacle can generate two orthogonal force vectors to a robot: one is called the coordinating force vector which is purposively designed in this paper, and the other is the repulsive force vector which is the same as that in a conventional artificial potential field. The ACF is designed according to the updated motion purpose and the relative states of the robot with respect to its local environment, and it also satisfies the robot’s dynamic constraints. The direction of the coordinating force can be determiend on line according to an optimal evaluation function. The ACF can effectively remove the local minima, and reduce the oscillation of the planned trajectory between multiple obstacles. Only local knowledge of the environments is needed in the ACF-based motion planning. The properties of the ACF such as controllability, adaptability, safety and reachability are studied and discussed in detail in this paper. Theoretical analysis and simulations are given to illustrate our main results.

Experimental study on phosphorus release from sediments of shallow lake in wave flume

Influence of wave on sediment resuspension and nutrients release from sediments, collected from Lake Taihu and Lake Chaohu, was studied in flume experiments. Under strong-wave conditions, concentrations of suspended solids (SS), total phosphorus (TP) and dissolved total phosphorus (DTP) in overlying water were increased significantly following the sediments re-suspension. During the experiments on sediments of Lake Taihu and Lake Chaohu, TP concentrations increased 6 times and 3 times, and DTP concentration increased 100% and 70% more than it in presuspension, respectively. Concentration of soluble reactive phosphorus (SRP) of experiment on sediment of Lake Taihu increased 2.%. During the massive sediment suspension, the dissolved phosphorus in pore water and much of the phosphorus adsorbed by the sediment particles were released into overlying water. The phenomena in this wave flume experiment are quite similar to the situation observed in situ of Lake Taihu. The critical wave stresses of sediment re-suspension are nearly equal. The change of concentrations of SS, TP, and SRP was the same as that in situ *** study showed that concentrations of TP and SRP in lake water could be increased significantly by wave disturbance. Phosphorus release was significantly enhanced by wave disturbance at the beginning of massive sediment re-suspension, but decreased later.

A new class of symmetric periodic solutions of the spatial elliptic restricted three-body problem

We show that there exists a new class of symmetric periodic solutions of the spatial elliptic restricted three-body problem. In such a solution, the infinitesimal body is confined to the vicinity of a primary and moves on a nearly circular orbit. This orbit is almost perpendicular to the orbital plane of the primaries, where the line of symmetry of the orbit lies. The existence is shown by applying a corollary of Arenstorf's fixed point theorem to a periodicity equation system of the problem. And this existence doesn't require any restriction on the mass ratio of the primaries, nor on the eccentricity of their relative elliptic orbit. Potential relevance of this new class of periodic solutions to real celestial body systems and the follow-up studies in this respect are also discussed.

Geographical Agglomeration of chinese Manufacturing Industries

Based on the plant level data from the First National Census of Basic Units, the Second National Census of Basic Units, and the First National Economic Census of china, this paper ex- amines the geographical agglomeration of chinese manufacturing industries at a more disaggregated level both by industrial classi-fication and spatial unit analysis. Applying the agglomeration index proposed by Maurel and Sedillot, studies show that the most agglomerated industries are resource-intensive, and capital and technology intensive while industries demanding localized inputs or serving localized markets or favored by local governments are fairly dispersed. The more disaggregated industries are more spa- tially agglomerated. At the finer spatial scale, industries are more dispersed. Results indicate that within-county spillovers are stronger than nearby-county spillovers, and within-prefecture spillovers are stronger than nearby prefecture spillovers. Local- ized spillovers are still quite substantial at a range beyond that of counties. Comparing the agglomeration index of chinese manu- facturing industries in 1996, 2.01 and 2.04, agglomeration seems to be a general tendency. Results also indicate that some industries have experienced remarkable changes in their levels of agglom- eration in the period 1996–2.04.
