MicroRNA (miR396) negatively regulates expression of ceramidase-like genes in Arabidopsis

MicroRNAs (miRNAs) are 21-23 nucleotide (nt), endogenous RNAs that regulate gene expression by targeting mRNAs for direct cleavage or translational repression in plants. In Arabidopsis, miR396 is encoded by two different loci (MIR396a and MIR396b) and sequence analysis suggests it may target three ceramidase-like genes (Atceramidase-like 1, Atceramidase-like 2 and Atceramidase-like 3). To demonstrate the biological function of miR396, we inserted the synthetic precursors, MIR396a or MIR396b, under the control of the enhanced cauliflower mosaic virus (CaMV) 35S promoter, into a plant transformation vector (pOCA30) and transformed the constructs into Arabidopsis. The promoter increased miR396 levels by more than 2-fold, indicating appropriate maturation of the synthetic precursor MIR396a or MIR396b transcript in transgenic plants. Microarray analysis showed that the transcript levels of two ceramidase-like genes (Atceramidase-like 1, Atceramidase-like 2) were decreased by more than 2-fold and lactosylceramide 4-a-galactosyltransferase increased by more than 2-fold in transgenic plants compared with the empty vector-transformed plants. Northern blot analysis showed that the mRNA levels of the two ceramidase-like genes were significantly reduced in transgenic plants. These results indicated that miR396 probably plays a crucial role in the ceramide metabolism pathway by negatively regulating the expression of ceramidase-like genes in Arabidopsis.

Self-assembly of peptides into fibrils and other morphologies has attracted much attention inmany fields,especially in nanofabrication,pathology and *** this paper,self-assembly of GAV-9peptide in organic solvents,ethanol and acetone,was investigated using atomic force microscopy(AFM)and attenuated total reflectance-Fourier transform infrared spectroscopy(ATR-FTIR).The results indicatedthat GAV-9 self-assembled into various nanostructures in both solvents after deposited and evaporated *** with β-sheet conformation were observed in both solvents when the peptide concentrationwas higher than 280 μ***,ordered fibrils with β-sheet conformation were formed in ethanol,butnot in acetone,with a peptide concentration ranging from 7 μM to 28 μ*** attribute the formation ofvarious nanostructures to the different physicochemical properties of the polar organic solvents on the self-assembly of GAV-9 peptide.

Phylogeny of fireflies (Coleoptera: Lampyridae) inferred from mitochondrial 16S ribosomal DNA, with references to morphological and ethological traits

We sequenced partial mitochondrial 16S ribosomal DNA (16S rDNA) of 18 firefly species from Southwest of china. Combined with homologous sequences previously reported, phylogenetic trees including Japanese, Korean and chinese species were reconstructed by neighbor-joining, maximum parsimony and Bayesian methods. All reconstructions agree on most nodes of the trees. Monophyly of Lampyridae is not supported because Rhagophthalmus ohbai in Rhagophthalmidae is included within it. Lamprigera, a genus placed unreliably in Lampyrinae, shows a close relationship to Amydetinae. Monophyly of Luciolinae is not supported because Pristolycus sagulatus (Lampyrinae) is included within it. In the Luciolinae, monophyly of Curtos and Hotaria is well established, respectively, but both morphological and molecular data continue to indicate that Luciola is not monophyletic and its subdivision is indeed necessary. Within the Lampyrinae, both Pyrocoelia and Diaphanes are not monophyletic, but monophyly of Pyrocoelia + Diaphanes is well supported. Phylogeny of Diaphanes is discussed for the first time. Generic placement of a newly discovered species (Diaphanes pectinealis Li et Liang) sharing some characters of Pyrocoelia and Diaphanes challenges the delimitation of these two similar genera. With references to the firefly mating systems, we suggest that more emphases should be placed on those sexually selected characters such as antennal structure in taxonomy of Lampyridae.

Effect of sediment resuspension on underwater light field in shallow lakes in the middle and lower reaches of the Yangtze River:A case study in Longgan Lake and Taihu Lake

Based on three continuous in situ underwater light field measurement under different wind waves conditions in Longgan Lake, Meiliang Bay of Taihu Lake in July 2003 and littoral zone near TLLER in July 2004, respectively, the effects of sediment resuspension caused by wind waves on PAR diffuse attenuation, absorption coefficients and euphotic depths are analyzed. In Longgan Lake, PAR diffuse attenuation coefficients during small, middle and large wind waves were 1.74, 2.02 and 2.45 m-1, respectively, and the corresponding PAR spectral diffuse attenuations ranged from 0.98 to 2.97, 1.34 to 3.95 and 1.80 to 5.40 m-1, respectively. In Meiliang Bay, PAR diffuse attenuation coefficients were 2.63, 3.72, 4.37 m-1 during small, middle and large wind waves. PAR diffuse attenuation coefficients increased by 41% and 66% from small to middle, large wind waves, *** coefficients integrated over the range of PAR of CDOM, phytoplankton were 0.26, 0.28 m-1;0.76, 0.49 m-1, respectively during middle and large wind waves. Absorption coefficients integrated over the range of PAR of non-algal particulate matter and total suspended particulate matter increased from 0.94 to 1.73 m-1, and from 1.70 to 2.22 m-1, respectively during middle and large wind waves. Relative contributions of absorption coefficients of non-algal particulate matter to total absorption coefficient integrated over the range of PAR were 44.14%, 65.05%, respectively, during middle and large wind waves. PAR euphotic depths decreased by 0.40, 0.19, 0.20 m from middle to large wind waves in Longganhu Lake, Meliang Bay and littoral zone near TLLER. Significant correlations were found between transparency, PAR diffuse attenuation coefficients, euphotic depths and total suspended particulate matter, wind velocity, wave height. A formula on linear complexity of highest coordinate sequences from maximal periodic sequences over Galois rings

Using a polynomial expression of the highest coordinate map, we deduce an exact formula on the linear complexity of the highest coordinate sequence derived from a maximal periodic sequence over an arbitrary Galois ring of characteristic p2 , where p is a prime. This generalizes the known result of Udaya and Siddiqi for the case that the Galois ring is Z4.

The deep thermal characteristic of continental margin of the northern South china Sea

Heat flow plays an important role in the study ot thermal structure and thermal evolutionof continental margin of the northern South china Sea. The analysis of heat flow value shows that margin heat flow in the northern South china Sea is relatively high setting, but the percentage of crustal heat flow is lower than 35% in terrestrial heat flow. The terrestrial heat flow exhibited a current of rise from the Northern Continental Margin to the Southern Central Basin. However, the proportion of crustal heat flow in terrestrial heat flow slowly dropped down in the same direction. It is suggested that the main factor causing high heat flow setting is the moving up of hot material from asthenosphere.

Molecular identification of species in Prunus sect. Persica (Rosaceae), with emphasis on evaluation of candidate barcodes for plants

Species of Prunus L. sect. Persica are not only important fruit trees, but also popular ornamental and medicinal plants. Correct identification of seedlings, barks, or fruit kernels is sometimes required, but no reliable morphological characters are available. Nowadays, the technique of DNA barcoding has the potential to meet such requirements. In this study, we evaluated the suitability of 11 DNA loci (atpB-rbcL, trnH-psbA, trnL-F, trnS-G, atpF-H, rbcL, matK, rpoB, rpoC1, nad1, and internal transcribed spacer [ITS]) as candidate DNA barcodes for peaches, using samples from 38 populations, covering all the species in sect. Persica. On the whole, the primers worked well in this group and sequencing difficulties were met only in the case of ITS locus. Five loci (rbcL, matK, rpoB, rpoC, and nad1) have very low variation rates, whereas atpB-rbcL, atpF-H, trnH-psbA, trnL-F and trnS-G show more variability. The most variable loci, atpB-rbcL and trnH-psbA, can distinguish three of the five species. Two two-locus combinations, atpB-rbcL+trnL-F and atpB-rbcL+atpF-H, can resolve all five species. We also find that identification powers of the loci are method-dependent. The NeighborNet method shows higher species identification power than maximum parsimony, neighbor joining, and unweighted pair group method with arithmetic mean methods.

Discovery of immune related factors in Fenneropenaeus chinensis by annotation of ESTs

A total of 10446 expressed sequence tags (ESTs) are obtained by a large scale sequencing of a cDNA library from cephalothorax of adult Fenneropenaeus chinensis . An EST analysis platform was built up based on local computers and bioinformatic techniques were used to annotate these ESTs in order to promptly find possible functional genes, especially for immune related factors. About 4% of the ESTs show similarity to the coding sequences of such factors, including lectin, serine protease, serpin, lysozyme, etc. These ESTs provide a partial profile of the immune system in F. chinensis and useful information for further study on these genes.
