PsRNA:A Computing Engine for the Comparative Identification of Putative Small RNA Locations within Intergenic Regions

Small RNAs （sRNAs） are non-coding transcripts exerting their functions in the cells directly. Identification of sRNAs is a difficult task due to the lack of clear sequence and structural biases. Most sRNAs are identified within genus specific intergenic regions in related genomes. However, several of these regions remain un-annotated due to lack of sequence homology and/or potent statistical identification tools. A computational engine has been built to search within the intergenic regions to identify and roughly annotate new putative sRNA regions in Enterobacteriaceae genomes. It utilizes experimentally known sRNA data and their flanking genes/KEGG Orthology （KO） numbers as templates to identify similar sRNA regions in related query genomes. The search engine not only has the capability to locate putative intergenic regions for specific sRNAs, but also has the potency to locate conserved, shuffled or deleted gene clusters in query genomes. Because it uses the KO terms for locating functionally important regions such as sRNAs, any further KO number assignment to additional genes will increase the sensitivity. The PsRNA server is used for the identification of putative sRNA regions through the information retrieved from the sRNA of interest. Objective:To investigate the analgesic and anti-inflammatory activities of ethanol extract of Passiflora foetida(***) ***:Ethanol extract of *** leaf was evaluated for analgesic action by acetic acid-induced writhing and hot plate method in albino *** anti-inflammatory property of ethanolic leaf extract was tested by carrageenan induced acute paw edema and histamine induced acute paw edema in ***:The dose 200 mg/kg of P. foetida leaf extract exhibited highest significant analgesic activity[(13.50±0.43) min]at a reaction time of 20 min in hot plate method in *** ethanol extract of leaf dose 100 mg/kg produced a highly significant anti inflammatory effect[(1.302±0.079) mL]in ***:It is very clear that *** also has analgesic and anti-inflammatory activities for the pharmaceuticals.

In the present study,anti-radical,anti-diabetic and antimicrobial activities of different solvent extracts of Passiflora ligularis fruits were *** the various solvents,acetone extract displayed maximum total phenolic(640.70 mg GAE/g extract),tannin(214.30 mg GAE/g extract)and flavonoid contents(387.33 mg RE/g extract).Results of antioxidant studies revealed that the acetone extract of fruits possessed an efficient 2,2-diphenyl-1-picryl-hydrazyl(DPPH•)(IC5019.13g/mL),2,2-azinobis(3-ethylbenzothiozoline-6-sulfonic acid)(ABTS•+)(9800.94mol/L trolox equi/g extract),superoxide(78.27%)and nitric oxide(79.95%)radical scavenging activities,ferric reducing antioxidant power(43.06 mmol Fe(II)/mg extract),metal chelating(134.53 mg EDTA/g extract)*** acetone extract of *** fruits also exhibited significant(P<0.005)inhibition activities onα-amylase(82.56%)andα-glucosidase(75.36%)***,the fruits of *** also demonstrated antibacterial activities against Gram(+)and Gram(−)bacteria and inhibited the fungal strains Candida albicans(14.85 mm)and Aspergillus niger(13.91 mm)in the disc diffusion *** of polyphenolics by HPLC showed the presence of ellagic acid,gallic acid,and ***,the results indicate that the fruit pulp of *** can serve as a potential antioxidative and antimicrobial agent in food and pharmaceutical industries.

Nature and source of the ore-forming fluids associated with orogenic gold deposits in the Dharwar Craton

Neoarchean orogenic gold deposits, associated with the greenstone-granite milieus in the Dharwar Craton include(1) the famous Kolar mine and the world class Hutti deposit;(2) small mines at HiraBuddini, Uti, Ajjanahalli, and Guddadarangavanahalli;(3) prospects at Jonnagiri; and(4) old mining camps in the Gadag and Ramagiri-Penakacherla belts. The existing diametric views on the source of ore fluid for formation of these deposits include fluids exsolved from granitic melts and extracted by metamorphic devolatilization of the greenstone sequences. Lode gold mineralization occurs in structurally controlled higher order splays in variety of host rocks such as mafic/felsic greenstones, banded iron formations, volcaniclastic rocks and granitoids. Estimated metamorphic conditions of the greenstones vary from lower greenschist facies to mid-amphibolite facies and mineralizations in all the camps are associated with distinct hydrothermal alterations. Fluid inclusion microthermometric and Raman spectroscopic studies document low salinity aqueous-gaseous(H_2O + CO_2 ± CH_4 + NaCl) ore fluids,which precipitated gold and altered the host rocks in a narrow P-T window of 0.7-2.5 kbar and 215-320℃. While the calculated fluid O-and C-isotopic values are ambiguous, S-isotopic compositions of pyrite-precipitating fluid show distinct craton-scale uniformity in terms of its reduced nature and a suggested crustal sulfur *** ages on greenstone metamorphism, granitoid plutonism and mineralization in the Hutti Belt are tantamount, making a geochronology-based resolution of the existing debate on the metamorphic *** fluid source impossible. In contrast, tourmaline geochemistry suggests involvement of single fluid in formation of gold mineralization, primarily derived by metamorphic devolatilization of mafic greenstones and interlayered sedimentary rocks, with minor magmatic contributions. Groundwater is generally presumed to be good for human consumption and is used as a main source of drinking water. Although there are numerous reasons for groundwater pollution, anthropogenic sources are consid-ered as the prime ones. In this study, twenty-two groundwater samples were collected from the flood plains of upper Palar River during the pre- and post-monsoon seasons to assess the extent of pollution and effects on human health. Physico-chemical characteristics of groundwater were analyzed and compared with those of drinking water standards recommended by the World Health Organization (WHO) and the Bureau of Indian Standards (BIS). The Piper's tri-linear diagram shows the nature of alkali earth with the high contents of alkalies and prevailing sulphate in the pre-sent samples. The univariate statistics and correlation analysis were performed to find out the relationships between the variables. The tannery effluents, solid wastes and sewage were suspected to be the predominant sources of pollu-tion in the area.

The study was to find out the effect of cadmium and mercury on levels of hemolymph glucose, crustacean hyperglycemic hormone（CHH） and hepatopancreas glycogen in the blue swimmer crab Portunus pelagicus. The experiments were performed in both intact and eyestalk ablated crabs. Quantification of CHH was done by the indirect ELISA with the aid of primary anti-Carcinus maenas-CHH antibody. Higher glucose concentration was observed on exposure to 8×10^–6 of cadmium（（825.6±5.42） μg/m L） and 6×10^–6 of mercury（（90.5±6.25） μg/m L）after 48 h and 24 h respectively. Higher level of hemolymph glucose was observed in eyestalk intact crabs on exposure to cadmium and mercury than eyestalk ablated crabs. Decrease in the levels of CHH was observed in both eyestalk intact and ablated crabs on heavy metal exposure. Decline of the hepatopancreas glycogen level was also witnessed with the exposure to heavy metal, which validated its utilization in the production of glucose. Thus this study brings to light, the variations in hemolymph glucose, CHH and hepatopancreas glycogen on heavy metal stress. These carbohydrate metabolites can be used as biomarkers in assessing heavy metal contamination in water bodies.

Molecular dynamics of amorphous pharmaceutical fenofibrate studied by broadband dielectric spectroscopy

Fenofibrate is mainly used to reduce cholesterol level in patients at risk of cardiovascular disease. Thermal transition study with the help of differential scanning calorimetry （DSC） shows that the aforesaid active pharmaceutical ingredient （API） is a good glass former. Based on our DSC study, the molecular dynamics of this API has been carried out by broadband dielectric spectroscopy （BDS） covering wide temperature and frequency ranges. Dielectric measurements of amorphous fenofibrate were per- formed after its vitrification by fast cooling from a few degrees above the melting point （Tm=354.11 K） to deep glassy state. The sample does not show any crystallization tendency during cooling and reaches the glassy state. The temperature dependence of the structural relaxation has been fitted by single Vogel- Fulcher-Tamman （VFT） equation. From VFT fit, glass transition temperature （Tg） was estimated as 250.56 K and fragility （m） was determined as 94.02. This drug is classified as a fragile glass former. Deviations of experimental data from Kohlrausch-Williams-Watts （KWW） fits on high-frequency flank of α-peak indicate the presence of an excess wing in fenofibrate. Based on Ngai＇s coupling model, we identified the excess wing as true Johari-Goldstein （JG） process. Below the glass transition temperature one can clearly see a secondary relaxation （γ） with an activation energy of 32.67 kJ/mol.
