Relationship between seminal plasma zinc concentration and spermatozoa-zona pellucida binding and the ZP-induced acrosome reaction in subfertile men

The aim of this study was to determine the relationship between seminal zinc concentration and spermatozoazona pellucida （ZP） binding and the ZP-induced acrosome reaction （ZPIAR） in subfertile men. Semen analyses and seminal zinc concentration assessments were carried out according to the World Health Organization manual for 458 subfertile men. A spermatozoa-ZP interaction test was carried out by incubating 2 × 10^6 motile spermatozoa with a group of four unfertilized oocytes obtained from a clinical in vitro fertilization programme. After 2 h of incubation, the number of spermatozoa bound per ZP and the ZPIAR of ZP-bound spermatozoa were examined. The effect of adding 0.5 mmol L^-1 zinc to the media on the ZPIAR of spermatozoa from normozoospermic men was also tested in vitro. Seminal zinc concentration positively correlated with sperm count and duration of abstinence, but negatively correlated with semen volume. On analysis of data from all participants, both spermatozoa-ZP binding and the ZPI- AR were significantly correlated with sperm motility and normal morphology, but not with seminal zinc concentration. However, in men with normozoospermic semen, the seminal zinc concentration was significantly higher in men with defective ZPIAR （〈 16%） than in those with normal ZPIAR （ ≥ 16% ）（P 〈 0.01）. The addition of 0.5 mmol L^-1 zinc to the culture media had no effect on spermatozoa-ZP binding, but significantly reduced the ZPIAR in vitro （P 〈 0. 001）. In conclusion, seminal zinc concentration is correlated with sperm count and the duration of abstinence in subfertile men. In men with normozoospermic semen, high seminal zinc concentration may have an adverse effect on the ZPIAR.

Mannose-binding lectin (MBL) is a pattern-recognition molecule that binds to characteristic carbohydrate mo-tifs present on the surface of many different pathogens. MBL binding stimulates the immune system via the lectin pathway of complement activation. In certain clinical situations, often characterized by pre-existing immune compromise, MBL deficiency increases the risk of infec-tious and other disease-specific complications. Many of the key pathogenic processes inherent to common gastroenterological diseases, such as infection, immuno-logical damage, and carcinogenesis, have been linked to MBL. This editorial reviews the biology of MBL, outlines key disease associations to document the breadth of influence of MBL, and finally, highlights the relevance of MBL to both gastroenterological health and disease.

Impact of geography on organ allocation:Beyond the distance to the transplantation center

Ozone and ozone/hydrogen peroxide treatment to remove gemfibrozil and ibuprofen from treated sewage effluent: Factors influencing bromate formation

Reuse of treated sewage effluent is an important option to overcome water *** wastewater treatment methods are inadequate for the removal of several persistent contaminants such as pharmaceuticals and personal care products(PPCPs).In this study the removal of ibuprofen and gemfibrozil by ozonation and ozone/hydrogen peroxide(O3/H2O2)advanced oxidation process(AOP),as well as the formation of bromate were investigated at different temperatures and pH *** removal of gemfibrozil and a maximum of 80%ibuprofen removal were achieved using ozone dosage of 1.5 mg/L with O3:H2O2 ratio of 1:0.25 in the O3/H2O2 *** effect of temperature on the removal efficiency of these two compounds was found to be negligible from 25 to 40ᵒC for both *** effect from 6 to 9 was also found to be negligible for gemfibrozil removal,while ibuprofen had relatively lower removal by ozonation at pH 6 compared to higher pH *** formation was decreased to 0.012 mg/L when the pH was increased to *** the temperature to 40ᵒC also resulted in less bromate formation which was the lowest value obtained in this study at 0.0102 mg/*** of H2O2 did not affect the formation of bromate and in some cases it was found to be slightly higher compared with ozonation treatment.

Non-alcoholic fatty liver disease: a narrative review of genetics

Non-alcoholic fatty liver disease(NAFLD) is now the most common cause of chronic liver diseases worldwide. It encompasses a spectrum of disorders ranging from isolated hepatic steatosis to nonalcoholic steatohepatitis(NASH),fibrosis, cirrhosis, and hepatocellular carcinoma. One of the key challenges in NAFLD is identifying which patients will progress. Epidemiological and genetic studies indicate a strong pattern of heritability that may explain some of the variability in NAFLD phenotype and risk of progression. To date, at least three common genetic variants in the PNPLA3, TM6 SF2, and GCKR genes have been robustly linked to NAFLD in the population. The function of these genes revealed novel pathways implicated in both the development and progression of NAFLD. In addition,candidate genes previously implicated in NAFLD pathogenesis have also been identified as determinants or modulators of NAFLD phenotype including genes involved in hepatocellular lipid handling, insulin resistance,inflammation, and fibrogenesis. This article will review the current understanding of the genetics underpinning the development of hepatic steatosis and the progression of NASH. These newly acquired insights may transform our strategy to risk-stratify patients with NAFLD and to identify new potential therapeutic targets.

Two decades ago,the term“infodemic”was coined right after the Severe Acute Respiratory Syndrome(SARS)emerged(1).The World Health Organization(WHO)defined infodemic as“too much information including false or misleading information in digital and physical environments during a disease outbreak.”While fully realizing the negative impact of misinformation and disinformation during an epidemic,we argue that most epidemic related information was generated not as a result of bad intention,but as a result of haste,confusion and the lack of reliable *** is especially true for coronavirus disease 2019(COVID-19),the emergence of a novel virus and uncertainties surrounding variants and *** a lack of timely scientific evidence,it is reasonable and even recommended for global citizens to debate and voice skepticism on certain *** the past 3 years of the COVID-19 pandemic,there was some degree of infodemic during the first year.

Physician approaches to drug shortages: Results of a national survey of pediatric hematologist/oncologists

AIM To evaluate personnel involved in scarce drug prioritization and distribution and the criteria used to inform drug distribution during times of shortage among pediatric hematologists/oncologists. METHODS Using the American Society of Pediatric Hematology/Oncology(ASPHO) membership list, a 20 question survey of pediatric hematologists/oncologists was conducted via email to evaluate personnel involved in scarce drug prioritization and distribution and criteria used to inform scarce drug distribution. RESULTS Nearly 65% of the 191 study respondents had patients directly affected by drug shortages. Most physicians find out about shortages from the pharmacist(n = 179, 98%) or other doctors(n = 75, 41%). One third of respondents do not know if there is a program or policy for handling drug shortages at their institution. The pharmacist was the most commonly cited decision maker for shortage drug distribution(n = 128, 70%), followed by physicians(n = 109, 60%). One fourth of respondents did not know who makes decisions about shortage drug distribution at their institution. The highest priority criterion among respondents was use of the shortage drug for curative, rather than palliative intent and lowest priority criterion was order of arrival or first-come *** Despite pediatric hematology/oncology physicians and patients being heavily impacted by drug shortages, institutional processes for handling shortages are lacking. There is significant disparity between how decisions for distribution of shortage drugs are currently made and how study respondents felt those decisions should be made. An institution-based, and more importantly, a societalapproach to drug shortages is necessary to reconcile these disparities.

Conventional electronic stability control(ESC) system uses differential braking to stabilize the yaw rate and side slip of the vehicle *** the roll stability is critically important,it is absent from the current ESC *** this paper,an model predictive control(MPC) scheme is proposed that potentially can achieve both yaw stability and roll *** objective of the proposed MPC algorithm is to track a reference yaw rate and body side slip angle,which serves the purpose of yaw stability ***,a rollover prevention constraint is imposed to achieve roll *** MPC optimizes the vehicle dynamics by stabilizing the yaw motion while observing the rollover prevention constraint,hence the yaw and roll stability can be achieved ***,it is found that the optimization results from MPC intentionally introducing larger slip angles into the system to avoid rollover,which is a somewhat counter-intuitive action.A high fidelity heavy-duty truck model from TruckSim is used for simulations and the effectiveness of the proposed approach is demonstrated by the TruckSim and Simulink joint simulations.

Genomic regions underlying agronomic traits in linseed(Linum usitatissimum L.) as revealed by association mapping

The extreme climate of the Canadian Prairies poses a major chal enge to improve yield. Although it is possible to breed for yield per se, focusing on yield-related traits could be advantageous because of their simpler genetic architecture. The Canadian flax core col ection of 390 accessions was genotyped with 464 simple sequence repeat markers, and phenotypic data for nine agronomic traits including yield, bol s per area, 1,000 seed weight, seeds per bol , start of flowering, end of flowering, plant height, plant branching, and lodging col ected from up to eight environments was used for association mapping. Based on a mixed model （principal component analysis （PCA） t kinship matrix （K））, 12 significant marker-trait associations for six agronomic traits were identi-fied. Most of the associations were stable across environments as revealed by multivariate analyses. Statistical simulation for five markers associated with 1000 seed weight indicated that the favorable al eles have additive effects. None of the modern cultivars carried the five favorable al eles and the maximum number of four observed in any accessions was mostly in＆amp;nbsp;breeding lines. Our results confirmed the complex genetic architecture of yield-related traits and the inherent difficulties associated with their identification while il ustrating the potential for improvement through marker-assisted selection.
