Hepatocyte nuclear factor 1 alpha(HNF1A),hepatocyte nuclear factor 4 alpha(HNF4A),and forkhead box protein A2(FOXA2) are key transcription factors that regulate a complex gene network in the liver,creating a regulatory transcriptional *** Encode and ChIP-Atlas databases identify the recognition sites of these transcription factors in many glycosyltransferase *** in silico analysis of HNF1A,HNF4A,and FOXA2 binding to the ten candidate glyco-genes studied in this work confirms a significant enrichment of these transcription factors specifically in the *** previous studies identified HNF1A as a master regulator of fucosylation,glycan branching,and galactosylation of plasma ***,we aimed to functionally validate the role of the three transcription factors on downstream glyco-gene transcriptional expression and the possible effect on glycan *** used the state-of-the-art clustered regularly interspaced short palindromic repeats/dead Cas9(CRISPR/dCas9) molecular tool for the downregulation of the HNF1A,HNF4A,and FOXA2 genes in HepG2 cells-a human liver cancer cell *** results show that the downregulation of all three genes individually and in pairs affects the transcriptional activity of many gly co-genes,although downregulation of glyco-genes was not always followed by an unambiguous change in the corresponding glycan *** effect is better seen as an overall change in the total HepG2 N-glycome,primarily due to the extension of biantenna ry *** propose an alternative way to evaluate the N-glycome composition via estimating the overall complexity of the glycome by quantifying the number of monomers in each glycan *** also propose a model showing feedback loops with the mutual activation of HNF1A-FOXA2 and HNF4A-FOXA2 affecting glyco-genes and protein glycosylation in HepG2 cells.

Preparation of microparticles and nanoparticles using membrane-assisted dispersion,micromixing,and evaporation processes

Synthetic microporous membranes are increasingly used for energy-efficient and controlled production of micro-and nanoparticles and micro-and nanoemulsions with tuneable morphology and physico-chemical properties through various micromixing,emulsification,and evaporation *** emul-sification processes,the membrane pores are used for dispersed phase injection and size-controlled generation of droplets and droplet-templated *** micromixing processes,membrane is utilised as a micromixer for mixing two miscible liquids,usually solvent and antisolvent-rich solutions,which leads to the creation of supersaturation and subsequent nanoprecipitation or *** mem-brane evaporation processes,membrane is used to prevent phase dispersion while allowing efficient molecular diffusion of solvent and/or antisolvent vapour through gas-filled *** dispersion processes can be operated continuously by decoupling shear stress on the membrane surface from cross flow using tube insets,flow pulsations,swirling flow,membrane oscillations or membrane *** generation and solidification can be performed continuously in a single pass by connecting membrane module with a downstream *** dispersion processes can be used for pro-duction of nanoparticles such as nanovesicles(liposomes,micelles,ethosomes,and niosomes),nanogels,polymeric,lipid and metallic nanoparticles,and *** main advantages of membrane-assisted particle generation are in low energy consumption,controlled geometry and hydrodynamic conditions at the microscale level,flexible throughput due to modular and scalable design of membrane devices,and a wide choice of available microporous membranes with various wall porosities,wetta-bilities,pore sizes,and pore morphologies to suit different applications.

Optimal Offering of Energy Storage in UK Day-Ahead Energy and Frequency Response Markets

The offering strategy of energy storage in energy and frequency response(FR) markets needs to account for country-specific market regulations around FR products as well as FR utilization factors, which are highly uncertain. To this end, a novel optimal offering model is proposed for stand-alone price-taking storage participants, which accounts for recent FR market design developments in the UK, namely the trade of FR products in time blocks, and the mutual exclusivity among the multiple FR products. The model consists of a day-ahead stage, devising optimal offers under uncertainty, and a real-time stage, representing the storage operation after uncertainty is materialized. Furthermore, a concrete methodological framework is developed for comparing different approaches around the anticipation of uncertain FR utilization factors(deterministic one based on expected values, deterministic one based on worst-case values, stochastic one, and robust one), by providing four alternative formulations for the real-time stage of the proposed offering model, and carrying out an out-of-sample validation of the four model instances. Finally, case studies employing real data from UK energy and FR markets compare these four instances against achieved profits, FR delivery violations, and computational scalability.

Communication Resources Allocation for Time Delay Reduction of Frequency Regulation Service in High Renewable Penetrated Power System

Neoadjuvant treatment of pancreatic ductal adenocarcinoma:Whom,when and how

Pancreatic ductal adenocarcinoma(PDAC),which is notorious for its aggressiveness and poor prognosis,remains an area of great unmet medical need,with a 5-year survival rate of 10%-the lowest of all solid *** diagnosis,only 20%of patients have resectable pancreatic cancer(RPC)or borderline RPC(BRPC)disease,while 80%of patients have unresectable tumours that are locally advanced pancreatic cancer(LAPC)or have distant *** 60%of patients who undergo upfront surgery for RPC are unable to receive adequate adjuvant chemotherapy(CHT)because of postoperative complications and early cancer *** important paradigm shift to achieve better outcomes has been the sequence of therapy,with neoadjuvant CHT preceding *** surgical stages have emerged for the preoperative assessment of nonmetastatic pancreatic cancers:RPC,BRPC,and *** main goal of neoadjuvant treatment(NAT)is to improve postoperative outcomes through enhanced selection of candidates for curative-intent surgery by identifying patients with aggressive or metastatic disease during initial CHT,reducing tumour volume before surgery to improve the rate of margin-negative resection(R0 resection,a microscopic margin-negative resection),reducing the rate of positive lymph node occurrence at surgery,providing early treatment of occult micrometastatic disease,and assessing tumour chemosensitivity and tolerance to treatment as potential surgical *** this editorial,we summarize evidence concerning NAT of PDAC,providing insights into future practice and study *** research is needed to establish predictive biomarkers,measures of therapeutic response,and multidisciplinary stra tegies to improve patient-centered outcomes.

随着分布式资源在配电网中的比例不断提高,如何在市场化交易机制下实现配用电系统安全经济运行成为当下的研究热点。在市场环境下,配用电系统各层的运行管理面临着不确定性逐层加剧、市场规模快速扩展、市场交易与系统安全运行难以有效衔接等多重挑战。该文首先梳理市场环境下配用电系统运行优化的关键问题;其次,对传统解析优化方法的研究成果与研究中仍待解决的问题进行总结;然后,针对配用电系统市场交易、运行优化等问题特点,系统性地介绍深度强化学习技术,分析归纳深度强化学习在配用电系统中的研究现状。最后,提炼出贯穿配用电系统多层多主体协同优化问题中的三重研究需求,并对深度强化学习技术未来的应用路径与发展趋势进行展望。

Why ecosystem characteristics predicted from remotely sensed data are unbiased and biased at the same time-and how this affects applications

Remotely sensed data are frequently used for predicting and mapping ecosystem characteristics,and spatially explicit wall-to-wall information is sometimes proposed as the best possible source of information for ***,wall-to-wall information typically relies on model-based prediction,and several features of model-based prediction should be understood before extensively relying on this type of *** such feature is that model-based predictors can be considered both unbiased and biased at the same time,which has important implications in several areas of *** this discussion paper,we first describe the conventional model-unbiasedness paradigm that underpins most prediction techniques using remotely sensed(or other)auxiliary *** this point of view,model-based predictors are typically ***,we show that for specific domains,identified based on their true values,the same model-based predictors can be considered biased,and sometimes severely *** suggest distinguishing between conventional model-bias,defined in the statistical literature as the difference between the expected value of a predictor and the expected value of the quantity being predicted,and design-bias of model-based estimators,defined as the difference between the expected value of a model-based estimator and the true value of the quantity being *** show that model-based estimators(or predictors)are typically design-biased,and that there is a trend in the design-bias from overestimating small true values to underestimating large true ***,we give examples of applications where this is important to acknowledge and to potentially make adjustments to correct for the design-bias *** argue that relying entirely on conventional model-unbiasedness may lead to mistakes in several areas of application that use predictions from remotely sensed data.

BACKGROUND The present study investigated perfectionism from the bioecological model perspective as a multidimensional construct manifested in forms of excessively high personal standards,exaggerated worries about personal mistakes,doubt in one’s performance,oversized order and organization emphasis,and the importance of parental valuations and *** To investigate the relation between perfectionism;the quality of family,peer,and college relationships;and media usage and content *** The research was implemented in 2020 with 203 students(134 female,66%)aged 18-25 years,enrolled at the University of Osijek in *** questionnaire had five parts:A)sociodemographic data;B)the Multidimensional Perfectionism Scale;C)the“general life satisfaction”and“current situational life satisfaction”scale;D)family,peer,and college relationships quality scale;and E)media usage and content interests *** of the implemented instruments showed satisfactory reliability.A hierarchical regression analysis was implemented with the aim of establishing significant perfectionism *** Age and gender were significant predictors of *** with lower family relationship quality reported higher parental expectations and complaining as well as significantly higher doubts in personal performance and concern about ***,a lower peer relationship quality predicted doubts in personal performance and stronger concerns about *** quality of college relationships positively predicted higher perfectionist personal standards and *** life satisfaction predicted higher concerns about mistakes,while current situational life satisfaction predicted higher levels of perfectionist *** usage intensity had no significant *** interest in information-educational media predicted higher personal standards as well as concern about mistakes and *** interests in entertainment
