Single and double layer centrifugation improve the quality of cryopreserved bovine sperm from poor quality ejaculates

Background： density gradient centrifugation was reported as a technique of semen preparation in assisted reproductive techniques in humans and animals. This technique was found to be efficient in improving semen quality after harmful techniques such as cryopreservation. Recently a modified technique, single layer centrifugation,was proposed as a technique providing a large amount of high quality spermatozoa, and this treatment was performed before conservation. Single layer centrifugation has been studied prevalently in stallions and in boars,but limited data were available for bulls. Occasionally bulls are known to experience a transient reduction in semen quality, thus techniques that allow improvement in semen quality could be applied in this context. The aim of this study was the evaluation of single layer and double layer centrifugation by the use of iodixanol, compared with conventional centrifugation and non-centrifuged semen, on the sperm characteristics during the cryopreservation process in bulls with normal and poor semen ***： Single layer centrifugation and double layer centrifugation both significantly increased the percentage of normal spermatozoa and decreased the percentage of non-sperm cells in poor quality samples, while both were ineffective in those of normal quality. Sperm characteristics in poor quality samples increased after single layer centrifugation and double layer centrifugation, reaching values similar to those recorded in normal samples, and this trend is maintained after equilibration and after cryopreservation. Pulse wave doppler ultrasound of testicular arteries and their relationship with semen characteristics in healthy bulls

Background: Semen evaluation is used to estimate the testicular function. In bulls, the spermatozoa present in the ejaculate are the result of a process that begun more than 2 mo earlier, bequeathing a delayed depiction of the actual function of the testis. Since testis vascularization might be critical for the gonad function, selected pulse wave doppler ultrasound parameters were assessed in this study, for instance the peak systolic velocity, the end diastolic velocity and the resistive index of the testicular artery along the spermatic cord, the marginal portion of the testicular artery and the intratesticular branches of the testicular artery both in healthy adult and young *** between these parameters and characteristics of semen that was collected numerous times, before and after the doppler ultrasound ***: The peak systolic velocity and the end diastolic velocity measured in the testicular artery along the spermatic cord(supratesticular artery – SA) were variable among the bulls and within individual bulls, likely due to the convoluted course of the vessel. The resistive index was found highly repeatable in the same bull. A reduction in the resistive index was found between the supratesticular artery and the marginal portion of the testicular artery(P d between the marginal portion of the testicular artery and the intratesticular branches of the testicular artery(P differences were recorded for the pulse wave doppler ultrasound parameters in young bulls compared with adults. On the Short-Term Optimisation of a Hydro Basin with Social Constraints

In this paper, an optimisation problem for calculating the best energy bids of a set of hydro power plants in a basin is proposed. The model is applied to a real Spanish basin for the short-term (24-hour) planning of the operation. The algorithm considers the ecological flows and social consumptions required for the actual operation. One of the hydro plants is fluent, without direct-control abilities. The results show that the fluent plant can be adequately controlled by using the storage capacities of the other plants. In the simulations, the costs related to the social consumptions are more significant than those due to the ecological requirements. An estimate of the cost of providing water for social uses is performed in the study.

在大多数非洲国家都在努力实现经济增长并维持其增长势头的时候,埃塞俄比亚和卢旺达被评为非洲大陆增长最快的经济体。他们还设法在过去10年中保持了这种增长。鉴于这两个国家属于非洲大陆,并且面临着与该大陆其他国家类似的挑战,因此提出了关于这两个国家增长的因素或增长动力以及其他非洲国家可以从中学习哪些知识的经验问题。利用在面板数据分析中GMM估计技术,我们观察到这两个国家的主要增长动力是技术进步、总资本形成、电力消耗、政府规模、外国直接投资（FdI）和本地需求。此外,我们还观察到一些阻碍其增长的因素。我们得出的结论是,其他想要实现经济增长的非洲国家必须紧跟埃塞俄比亚和卢旺达的脚步,同时注意观察到的对其增长不利的因素。我们还建议,不应复制政策的每一个具体措施,而必须根据实施国的需要和要求量身定制政策,以使其生效。

Molecular and in vitro Characterization of Field Isolates of Bovine Herpesvirus-1

Bovine Herpesvirus-1 (BoHV-1) is distributed worldwide and is a major pathogen in cattle, being the causal agent of a variety of clinical syndromes. The aim of this study was to isolate and to characterize (molecular and biological characterization) BoHV-1 from 29 immunosuppressed animals. It was possible to obtain 18 isolates, each from a different animal, such as from the respiratory and reproductive tracts. In some cases the cytopathic effect was visible 12 hours post-inoculation, and became characteristic after 36-48 hours. Biological characteristics were evaluated and compared with Iowa and Colorado-1 reference strains, and differences were found in plaque size, virus titer measured by TCId50 and PFU/mL, and one step virus curves. These results showed that some isolates had a highly virulent-like behavior in vitro, compared to the reference strains, with shorter eclipse periods, faster release of virus into the supernatants, and higher burst size and viral titer. There were no differences in glycoprotein expression of BoHV-1 isolates, measured by Western blot on monolayers. Moreover, using restriction endonucleases analysis, most of the viruses were confirmed as BoHV-1.1 and just one of them was confirmed as BoHV-1.2a subtype. These findings suggest that some wild-type BoHV-1 isolates could be useful as seeds to develop new monovalent vaccines.

Analysis of growth and death kinetics at single-cell resolution is a key step in understanding the complexity of the nonreplicating growth phenotype of the bacterial pathogen Mycobacterium ***,we developed a single-cell-resolution microfluidic mycobacterial culture device that allows time-lapse microscopy-based Iong-term phenotypic visualization of the live replication dynamics of *** technology was successfully applied to monitor the real-time growth dynamics of the fast-growing model strain Mycobacterium smegmatis(***)while subjected to drug treatment regimens during continuous culture for 48 h inside the microfluidic device.A clear morphological change leading to significant swelling at the poles of the bacterial membrane was observed during drug *** addition,a small subpopulation of cells surviving treatment by frontline antibiotics was observed to recover and achieve robust replicative growth once regular culture media was provided,suggesting the possibility of identifying and isolating nonreplicative *** device is a simple,easy-to-use,and low-cost solution for studying the single-cell phenotype and growth dynamics of mycobacteria,especially during drug treatment.

In recent years,wire-arc directed energy deposition(wa dEd),which is also commonly known as wire-arc additive manufacturing(WAAM),has emerged as a promising new fabrication technique for magnesium *** major reason for this is the possibility of producing parts with a complex geometry as well as a fine-grained *** the process has been shown to be applicable for Mg-Al-Zn alloys,there is still a lack of knowledge in terms of the influence of the WAAM process on the age-hardening ***,this study deals with the aging response of a WAAM AZ91 *** order to fully understand the mechanisms during aging,first,the as-built condition was analyzed by means of high-energy X-ray diffraction(HEXRd)and scanning electron *** investigations revealed a finegrained,equiaxed microstructure with adjacent areas of alternating Al ***,the difference between single-and double-step aging as well as conventional and direct aging was studied on the as-built WAAM AZ91 alloy for the first *** aging response during the various heat treatments was monitored via in situ HEXRd *** electron microscopy and hardness studies were *** results showed that the application of a double-step aging heat treatment at 325℃with pre-aging at 250℃slightly improves the mechanical properties when compared to the single-step heat treatment at 325℃.However,the hardness decreases considerably after the pre-aging ***,aging at lower temperatures is preferable within the investigated temperature range of 250-325℃.Moreover,no significant difference between the conventionally aged and directly aged samples was ***,the specimens showed enhanced precipitation kinetics during aging as compared to cast *** could be attributed to a higher amount of nucleation sites and the particular temperature profile of the solution heat treatment.
