Salivary glands(SG)are exocrine organs with secretory units commonly injured by ***-engineered organoids and extracellular vesicles(EV)are currently under investigation as potential strategies for SG ***,three-dimensional(3D)cultures of SG functional organoids(SGo)and human dental pulp stem cells(hDPSC)were generated by magnetic 3D bioassembly(M3DB)*** growth factor 10(FGF10)was used to enrich the SGo in secretory epithelial *** 11 culture days via M3DB,SGo displayed SG-specific acinar epithelial units with functional properties upon *** consistently develop 3D hDPSC in vitro,3 culture days were sufficient to maintain hDPSC undifferentiated genotype and phenotype for EV *** isolation was performed via sequential centrifugation of the conditioned media of hDPSC and SGo *** were characterized by nanoparticle tracking analysis,electron microscopy and *** were in the exosome range for hDPSC(diameter:88.03±15.60 nm)and for SGo(123.15±63.06 nm).Upon ex vivo administration,exosomes derived from SGo significantly stimulated epithelial growth(up to 60%),mitosis,epithelial progenitors and neuronal growth in injured SG;however,such biological effects were less distinctive with the ones derived from ***,these exosome biological effects were investigated by proteomic *** spectrometry profiling of SGo exosomes predicted that cellular growth,development and signaling was due to known and undocumented molecular targets downstream of *** were identified as one of the novel targets requiring further ***,M3DB platforms can generate exosomes with potential to ameliorate SG epithelial damage.

目的:近年来,阴茎海绵体注射（ICI）干细胞治疗勃起功能障碍（ED）引起广泛的关注。但是,目前的研究发现绝大多数注射的干细胞很快被血流冲出海绵体。这成为干细胞海绵体注射治疗ED的主要技术障碍。为此,我们设计这个实验,旨在应用新的纳米技术使ICI的干细胞停留在海绵体里,从而有效的发挥干细胞的局部治疗的多种特性治疗ED。方法:体外实验:同种异体提取并培养脂肪干细胞（ADSCs）,并用CD3 1,CD34,CD45和CD90进行细胞表型鉴定。ADSCs与磁化纳米粒子（NanoShuttle）绑定生成Nano-ADSCs。体外观察Magnet对Nano-ADSCs的影响。体内细胞追踪实验:建立双侧海绵体神经损伤（BCNC）的大鼠ED模型。细胞在ICI前用CellTracker标记。24只BCNC大鼠随机分成3组（n=8）:ADSCs ICI组,Nano-ADSCs ICI组和Nano-ADSCs ICI+magnet组。在ICI后的第1,3,5和9天分别取2只大鼠的阴茎海绵体组织做免疫荧光进行体内细胞跟踪。疗效评估实验:40只BCNC大鼠随机分成4组（n=10）:BCNC组,BCNC+ADSCs ICI组,BCNC+Nano-ADSCs ICI组和BCNC+Nano-ADSCs ICI+Magnet组。在ICI后第28天,电刺激下测量海绵体内压和平均动脉压（ICP和MAP）。随后收取阴茎组织做免疫组化和蛋白免疫印迹,测指标PECAM-1和α-SMA。数据统计分析采用One-way ANOVA with post-hoc test。结果:体外实验:***可以与ADSCs成功绑定形成Nano-ADSCs。2.经过25天的观察,Nano-ADSCs无显著形态学上的变化,生长无明显异常。***-ADSCs的移动受Magnet的影响,朝Magnet方向呈聚集生长。4.流式细胞术显示:CD3 1-,CD34+,CD45-,CD90+。体内细胞追踪实验:***后,Nano-ADSCs+Magnet组多数Nano-ADSCs被保留在阴茎海绵体里面达三天以上,其他两个对照组只有极少数ADSCs停留在阴茎海绵体里面。2.从第5天开始CellTracker信号开始减弱,第9天信号几乎消失。疗效评估实验:ICI后第28天Nano-ADSCs仍然可通过NanoShuttle定位在阴茎海绵体内,部分能被定位在α-SMA和PECAM-1阳性染色区域。Nano-ADSCs+Magnet组的ICP/MAP比值与α-SMA和PECAM-1的表达显著高于其他两组（P<0.05）。结论:这种新的纳米技术的方法能有效的使ICI后ADSCs停留在海绵体里面生长,从而发挥局部治疗作用。它克服了先前干细胞ICI治疗的主要技术问题。我们进一步证实这种新方法比传统的ADCSs ICI方法更能显著改善神经损伤性大鼠的勃起功能。

Historical Perspective of Synthetic Biology in Food Production

Due to projections of population increase, which suggest that the world population will reach 8.6 billion people by 2030, as well as the reduction of existing natural resources, studies on food production technologies that minimize the impacts on the environment become relevant. In this context, with the creation of the first living organism controlled by a synthetic genome, since 2010, scientists from several countries study alternatives to produce energetic material and food through synthetic biology. Therefore, the present study sought to identify which the contributions, reflexes, trends and/or challenges of synthetic biology are in food production through a review of the literature. As results, it was noticed that there were significant advances in studies that seek forms of food production with the use of synthetic biology, as well as with technologies that reduce the use of natural resources and the impact of agricultural production on the environment. The topics of food safety, ethics and consumer perception, regarding the use of such technologies, are also emphasized. However, there is much to be studied on the subject, in particular, the need for experimental testing in different crops and processes due to food safety, and feasibility of the industrialization of such technologies.

Effective calcium(Ca)management is crucial for optimizing oil palm cultivation and enhancing crop *** study aimed to gain insights into the dynamics of Ca concentration,accumulation,exportation,immobilization,and recycling in various oil palm organs relative to plant *** experiment was conducted at the Agropalma enterprise site in the northeastern region of Para State,Brazil,evaluating seven plant age treatments:2,3,4,5,6,7,and 8 years *** a completely randomized design with four *** results demonstrated an age-related increase in Ca concentration in petioles,rachis,arrows,male inflorescences,peduncles,and ***,Ca accumulation exhibited an upward trend in all organs with progressing plant ***,the study revealed an enhanced Ca use efficiency across all plant organs in correlation with the age of oil palm *** findings underscore the dynamic nutritional demands of oil palm,influencing Ca immobilization,cycling,and export throughout its developmental stages.

Mechanical Circulatory Support in Patients with Sepsis： A Case Series

MCS （mechanical circulatory support） has shown great efficacy in treatment of respiratory mid cardiovascular diseases when conventional therapy is insufficient. The use of ECMO （extracolporeal membrane oxygenation） for the treatment of cardiogenic shock secondary to sepsis is still controversial. Studies have shown that ECMO should be considered as a valuable therapeutic option in cases of refractory cardiovascular dysfunction in severe septic patients, although more studies ale needed to consolidate its efficacy. We report the case of two patients, without previous history of cardiopathy, subjected to temporary MCS, with use of ECMO for the treatment of refractory cardiogenic shock secondary to sepsis.
