PGRMC1 Elevation in Multiple Cancers and Essential Role in Stem Cell Survival

Cancer is one of the leading causes of death in America, and there is an urgent need for new therapeutic approaches. The progesterone receptor membrane component 1 (PGRMC1) is a cytochrome b5 related protein that binds heme and is associated with signaling, apoptotic suppression and autophagy. PGRMC1 is essential for tumor formation, invasion and metastasis, and is upregulated in breast, colon, lung and thyroid tumors. In the present study, we have analyzed PGRMC1 levels in over 600 tumor sections, including a larger cohort of lung tumors than in previous studies, and report the first clinical analysis of PGRMC1 levels in human oral cavity and ovarian tumors compared to corresponding nonmalignant tissues. PGRMC1 was highly expressed in lung and ovarian cancers and correlated with patient survival. PGRMC1 has been previously associated with drug resistance, a characteristic of cancer stem cells. The stem cell theory proposes that a subset of cancerous stem cells contribute to drug resistance and tumor maintenance, and PGRMC1 was detected in lung-tumor derived stem cells. Drug treatment with a PGRMC1 inhibitor, AG-205, triggered stem cell death whereas treatment with erlotinib and the ERK inhibitor, PD98059, did not, suggesting a specific role for PGRMC1 in cancer stem cell viability. Together, our data demonstrate PGRMC1 as a potential tumor biomarker across a variety of tumors, as well as a therapeutic target for cancer stem cells.

为快速准确识别出感染黄曲霉毒素的玉米颗粒,基于黄曲霉毒素在365 nm紫外光照射下可发出黄绿色荧光(bright greenish-yellow fluorescence,bGYF)的特性,提出了一种以颜色为特征量的感染毒素颗粒的检出方法。首先针对同一组玉米颗粒样本分别获取其在紫外光和可见光下的2幅图像,对可见光图像进行二值化、颗粒区域填充等预处理操作,得到玉米颗粒的轮廓与位置信息;然后将进行彩色增强后的紫外光图像与预处理后的可见光图像掩膜;最后在掩膜图像中通过RGb模型中G通道下的阈值作为分割参数识别玉米颗粒上的感染区域。结果表明,所提出的识别方法对能够发出荧光的含黄曲霉菌玉米颗粒识别准确率达到84%以上,对含有黄曲霉毒素的正确检出率可达77.8%以上,能够达到区分检测的要求。

目的:探讨信号转导和转录活化因子(signal transducer and activator of transcription,STAT)-1在上皮性卵巢癌组织中的表达及其与预后的关系。方法:应用显微切割法从石蜡切片标本中提取mRNA,实时荧光定量PCR(real-time fluorogentic quantitative PCR,RFQ-PCR)和免疫组织化学方法检测105例原发性卵巢上皮癌和29例正常卵巢组织中STAT-1 mRNA及蛋白的表达,并分析其蛋白表达与预后的相关性。结果:上皮性卵巢癌组织中STAT-1 mRNA的表达高于正常卵巢组织[(0.96±1.05)vs(0.46±0.57),P=0.032],但mRNA表达水平与卵巢癌的手术分期、病理分级以及生存时间无关(P>0.05);上皮性卵巢癌组织中STAT-1和P-STAT-1蛋白的阳性表达率分别为72.4%和71.9%,显著高于正常卵巢组织(P0.05)。在行辅助泰素化疗的病例组中STAT-1和P-STAT-1蛋白高表达患者的预后较好,总的生存时间长于低表达患者(P<0.05);STAT-1蛋白高表达的卵巢癌患者无瘤生存时间明显长于低表达患者(P<0.01)。结论:STAT-1在上皮性卵巢癌中存在异常表达,与紫杉醇类药物化疗后的预后密切相关。STAT-1可能是抑制肿瘤细胞生长的关键因子之一。

The role of the vascularized bone marrow component as a continuous source of donor-derived hematopoietic stem cells that facilitate tolerance induction of vascularized composite allografts is not completely *** this study,vascularized composite tissue allograft transplantation outcomes between recipients receiving either conventional bone marrow transplantation(CbMT)or vascularized bone marrow(VbM)transplantation from balb/c(H2d)to C57bL/6(H2b)mice were *** high-or low-dose CbMT(1.5×10^(8)or 3×10^(7)bone marrow cells,respectively)was *** addition,recipients were treated with costimulation blockade(1 mg anti-CD154 and 0.5 mg CTLA4Ig on postoperative days 0 and 2,respectively)and short-term rapamycin(3 mg/kg/day for the first posttransplant week and then every other day for another 3 weeks).Similar to high-dose conventional bone marrow transplantation,5/6 animals in the vascularized bone marrow group demonstrated long-term allograft survival(>120 days).In contrast,significantly shorter median survival was noted in the low-dose CbMT group(~64 days).Consistently high chimerism levels were observed in the VbM transplantation ***,low levels of circulating CD4^(+)and CD8^(+)T cells and a higher ratio of Treg to Teff cells were maintained in VbM transplantation and high-dose CbMT recipients(>30 days)but not in low-dose VbM transplant ***-specific hyporesponsiveness was shown in tolerant recipients in *** of the vascularized bone marrow component after secondary donor-specific skin transplantation did not affect either primary allograft or secondary skin graft survival.

Gastroesophageal reflux disease after diagnostic endoscopy in the clinical setting

AIM: To investigate the outcome of patients with symptoms of gastroesophageal reflux disease (GERD) referred for endoscopy at 2 and 6 mo post endoscopy. METHODS: Consecutive patients referred for upper endoscopy for assessment of GERD symptoms at two large metropolitan hospitals were invited to participate in a 6-mo non-interventional (observational) *** two institutions are situated in geographically and socially disparate areas. Data collection was by selfcompletion of questionnaires including the patient assessment of upper gastrointestinal disorders symptoms severity and from hospital records. Endoscopic finding using the Los-Angeles classification, symptom severity and it's clinically relevant improvement as change of at least 25%, therapy and socio-demographic factors were assessed. RESULTS: baseline data were available for 266 patients and 2-mo and 6-mo follow-up data for 128 and 108 patients respectively. At baseline, 128 patients had erosive and 138 non-erosive reflux disease. Allmost all patient had proton pump inhibitor (PPI) therapy in the past. Overall, patients with non-erosive GERD at the index endoscopy had significantly more severe symptoms as compared to patients with erosive or even complicated GERD while there was no difference with regard to medication. After 2 and 6 mo there was a small, but statistically significant improvement in symptom severity (7.02 ± 5.5 vs 5.9 ± 5.4 and 5.5 ± 5.4 respectively); however, the majority of patients continued to have symptoms (i.e. , after 6 mo 81% with GERD symptoms). Advantaged socioeconomic status as well as being unemployed was associated with greater improvement. CONCLUSION: The majority of GORD patients receive PPI therapy before being referred for endoscopy even though many have symptoms that do not sufficiently respond to PPI therapy.

5-HTr Expression in Primary Trigeminal Neuron Following Injury

Trigeminal ganglia neurons significantly affect the amplitude and type of 5-HT receptor gene expression following activation of their axon terminals and sensitisation by painful stimuli. Moreover, these neurons significantly alter gene expression in cytoskeletal proteins following injury. The aim of the present study was to determine whether peripheral and/or central deafferenting lesions affect gene expression in serotonergic receptors that are involved in pain transmission. Adult rats were subjected to unilateral ablation of the facial sensory and motor cortices. Fifteen days after the surgery, degeneration of the cortico-trigeminal pathway was observed. Presynaptic deafferentation of the primary trigeminal neurons and central afferents of the contralateral ganglia was conducted. As a consequence of the excision of the meninges covering the ablated cortices, the peripheral axotomy of the trigeminal-vascular primary neurons of the ipsi-lateral side was induced. Serotonergic receptor (5-HT5A/5b/1b/1D/1F) gene expression was analysed in both sides of the trigeminal ganglia neurons. The results of the present study showed a significant increase in 5-HT5A/5b/1b/1D receptor gene expression in the primary sensory neurons of both ganglia, with the highest levels of expression noted in the ganglia contralateral to the lesion. 5-HT1F receptor expression, however, was more strongly expressed in the ganglia ipsilateral to the lesion. Our results also confirm that the adaptive response of primary trigeminal neurons to injury involves anatomical remodelling, as well as changes in receptor gene expression involved in sensory transmission. This may explain the distortion of sensory signals observed in trigeminal neuropathic states, and may lead to the development of novel pharmacological interventions.

background The receptor tyrosine kinase-like orphan receptor 2 （ROR2） gene has been recently shown to play important roles in palatal development in animal models and resides in the chromosomal region linked to non syndromic cleft lip with or without cleft palate in *** aim of this study was to investigate the possible association between ROR2 gene and non-syndromic oral *** Here we tested 38 eligible single-nucleotide polymorphisms （SNPs） in ROR2 gene in 297 non-syndromic cleft lip with or without cleft palate and in 82 non-syndromic cleft palate case parent trios recruited from Asia and *** based Association Test was used to test for deviation from Mendelian *** software was used to test potential parent of origin *** maternally mediated in utero effects were assessed using the TRlad Multi-Marker approach under an assumption of mating symmetry in the *** Significant evidence of linkage and association was shown for 3 SNPs （rs7858435,rs10820914 and rs3905385） among 57 Asian non-syndromic cleft palate trios in Family based Association Tests.P values for these 3 SNPs equaled to 0.000068,0.000115 and 0.000464 respectively which were all less than the significance level （0.05/38=0.0013） adjusted by strict bonferroni *** odds ratios for the risk allele were 3.42 （1.80-6.50）,3.45 （1.75-6.67） and 2.94 （1.56-5.56）,*** evidence of linkage and association was not shown for study groups other than non-syndromic cleft *** evidence for parent-of-origin nor maternal genotypic effect was shown for any of the ROR2 markers in our analysis for all study *** Our results provided evidence of linkage and association between the ROR2 gene and a gene controlling risk to non-syndromic cleft palate.
