Polymorphisms of GSTs in Lung Adenocarcinoma Patients Followed in the Context of a Biobank

Background: Lung Adenocarcinoma (ADC) has been recently associated with distinct molecular changes, leading to the development of molecular-based targeted therapy. The Nancy’s Centre of Biological Resources (“Centre des Ressources Biologiques”, CRB) is an ISO 9001-2000 certified biobank with biological material and follow-up data from lung cancer patients, which collected during the last 20 years. Objective: To estimate and compare the frequency of Glutathionne S-Transferase (GST) polymorphisms in a French population of ADC patients. Methods: A retrospective study was conducted by the CRB between 1988 and 2007: 296 consecutive patients operated upon for ADC and 447 healthy subjects were evaluated. Genomic DNA was obtained from peripheral blood samples collected in EDTA tubes. The DNA was extracted using proteinase K digestion and phenol: chloroform purification. The GST polymorphisms were studied with duplex SYBR Green q PCR using specific primers and results being read on melt curves. Results: Two GST classes were monitored during this research. The Mu class GST (GSTM) and the Theta class GST (GSTT) members. We studied the incidence of each genotype, as well as the GSTMT (combined Mu and Theta class) and null genotype in ADC and control patients. ADC patients had a higher incidence of the GSTM polymorphism (p 0.0001, 95%CI 1.63 - 3.24) and a lower incidence of the GSTT polymorphism (p 0.0001, 95%CI 0.31 - 0.66) comparing to control. The null and GSTMT genotype had no significant statistical differences between the two groups. Conclusion: ADC patients were found to have a higher incidence of the GSTM genotype and a lower incidence of the GSTT genotype, compared to controls. Future studies may help elucidate the possible contribution of these genotypic differences in lung adenocarcinoma carcinogenesis or regarding the response to chemotherapy.

AIM: To evaluate the response of hepatocellular carcinoma (HCC) to transarterial chemoembolization (TACE) using a simplified protocol of parametric contrast-enhanced ultrasound (pCEUS). METHODS: Eighteen patients with HCC (18 target tumors, diameter: 2.8-12 cm) were evaluated before, and 20 d after TACE. The distribution and morphology of TACE-induced necrosis in these tumors precluded accurate evaluation by visual assessment or by simple measurements. For pCEUS, a 4.8 mL bolus of SonoVue (Bracco, Milan, Italy) was intravenously administered and analysis of tumor perfusion during the initial phase of enhancement (0-30 s post injection) was performed with dedicated software (Qontrast, Bracco, Milan, Italy). Time-intensity curves were plotted and three parameters were calculated: peak intensity (PI, in percentage %), time to peak (TTP in seconds, s) and area under the curve during wash-in (AUC-WI, in arbitrary units, a.u). Magnetic resonance imaging was the standard imaging modality for post-treatment evaluation. Changes in tumor size were recorded and response was assessed according to response evaluation criteria in solid tumors criteria. RESULTS: A statistically significant decrease in PI and AUC-WI was observed in the treated tumors post TACE; PIpre: 21.5% ± 8.7% (mean ± SD), PIpost: 12.7% ± 6.7%, P < 0.001, AUC-WI pre: 17493 ± 9563 a.u, AUC-WI post: 9585 ± 5494 a.u, P < 0.001. A slight increase in TTP was noted post TACE, but this was not statistically significant; TTP pre: 13.1 ± 4.3 s, TTP post: 13.6 ± 4.2 s , P = 0.058). The changes in the aforementioned parameters were not accompanied by significant tumor shrinkage. CONCLUSION: pCEUS, even when limited to the study of the arterial phase of tumoral enhancement, can detect and quantify early perfusional changes in HCC post TACE.

Buried bumper syndrome(BBS)is the situation in which the internal bumper of the gastrostomy tube,due to prolonged compression of the tissues between the external and the internal bumper,migrates from the gastric lumen into the gastric wall or further,into the tract outside the gastric lumen,ending up anywhere between the stomach mucosa and the surface of the *** restricts liquid food from entering the stomach,since the internal opening is obstructed by gastric mucosal *** performed a comprehensive search of the PubMed literature to retrieve all the case-reports and case-series referring to BBS and its management,after which we focused on the endoscopic techniques for releasing the internal bumper to re-establish the functionality of the *** the“push”and the“push and pull T”techniques to the most sophisticated-using high tech instruments,all 10 published techniques have been critically analysed and the pros and cons presented,in an effort to optimize the criteria of choice based on maximum efficacy and safety.

Stroboscopic Evaluation of the Vocal Folds’ Morphometric Characteristics, between the Different Voice Categories on Professional Singers

Objective(s): The aim of this study is to explore if there is a correlation between the typical voice classification and the morphometric characteristics of the vocal folds, using video laryngeal stroboscopy, on professional singers in Greece. Methods: 70 professional singers, 50% men (35 men) and 50% women (35 women), were recruited for this study. A stroboscopic evaluation was prepared for every participant. Additionally, the voice classification and features of individuals were correlated statistically. Results: Statistically significant difference were observed between the voice categories and the size of stomatopharynx, the shape of the hard palate, the length of the vocal folds, the width and thickness of the vocal folds, as well as the tone of speech, in male singers. Statistically significant differences were also observed between the voice categories and all the variables except the shape of epiglottis, the color of speech and the shape of the vocal folds, in female singers. Conclusions: There is correlation between the morphometric characteristics of the vocal folds and the voice categories.

Comparison of the S-, N- or P-Deprivations’ Impacts on Stomatal Conductance, Transpiration and Photosynthetic Rate of Young Maize Leaves

Seven-day-old maize (Zea mays) plants were grown hydroponically for ten days in deprived nutrient solutions against the corresponding control grown under full nutrition;the effects of S-, N- or P-deprivation on laminas’ mean stomatal conductance (gs), transpiration rate (E) and photosynthetic rate (A) were monitored, along with the impact on the laminas’ total dry mass (DM), water amount (W), length and surface area (Sa). Furthermore, a time series analysis of each parameter’s response ratios (Rr), i.e. the treatment’s value divided by the corresponding control’s one, was performed. Under S-deprivation, the Rr of laminas’ mean gs, E, and A presented oscillations within a ±15% fluctuation zone, notably the “control” zone, whilst those of laminas’ total DM, water amount, surface area, and length included oscillation during the first days and deviation later on, presenting deviation during d10. Under the N-deprivation conditions all Rr time courses except the A one, included early deviations from the control zone without recovering. The deviation from the control zone appeared at d4. Under P-deprivation, all Rr time courses represented oscillations within the control zone. P-deprivation’s patterns resembled those of S-deprivation. Compared to the one of the S-deprivation, the P-one’s oscillations took place within a broader zone. Linear relationships among the various Rr patterns were found between gs-E, gs-A, E-A, DM-W and DM-Sa. In conclusion, the impact of P-deprivation appeared in an early stage and included an alleviation action, the one of N-deprivation appeared early with no alleviation action, whilst that of S-deprivation appeared later, being rather weaker when compared to the impact of the P-deprivation’s impact.

A Connectivity-Based Legalization Scheme for Standard Cell Placement

Standard cell placement algorithms have been at the forefront of academic research concerning the physical design stages of VLSI design flows. The penultimate step of a standard cell placement procedure is legalization. In this step the manufacturability of the design is directly settled, and the quality of the solution, in terms of wirelength, congestion, timing and power consumption is indirectly defined. Since the heavy lifting regarding processing is performed by global placers, fast legalization solutions are protruded in state-of-the-art design flows. In this paper we propose and evaluate a legalization scheme that surpasses in execution speed two of the most widely used legalizers, without not only corrupting the quality of the final solution in terms of interconnection wirelength but improving it in the process.
