Our previous studies have demonstrated that Fam20 C promotes differentiation and mineralization of odontoblasts,ameloblasts,osteoblasts and osteocytes during tooth and bone *** of the Fam20 C gene inhibits bone and tooth growth by increasing fibroblast growth factor 23 in serum and causing hypophosphatemia in conditional knockout ***,control and regulation of the expression of Fam20 C are still *** this study,we generated a transgenic reporter model which expresses green fluorescence protein（GFP） driven by the Fam20 C *** was used to insert a 16 kb fragment of the mouse Fam20 C gene（containing the 15 kb promoter and 1.1 kb of exon 1） intoa pBluescript SK vector with the topaz variant of GFP and a bovine growth hormone polyadenylation *** expression was subsequently evaluated by histomorphometry on cryosections from E14 to adult *** was evident in the bone and teeth as early as *** GFP signal was maintained stably in odontoblasts and osteoblasts until 4 weeks after *** expression of GFP was significantly reduced in teeth,alveolar bone and muscle by 8 weeks of *** also observed colocalization of the GFP signal with the Fam20 C antibody in postnatal 1- and 7-day-old *** generation of Fam20C-GFP transgenic mice will provide a unique model for studying Fam20 C gene expression and the biological function of this gene during odontogenesis and osteogenesis.

LIN28 is an RNA binding protein with important roles in early embryo development,stem cell differentiation/re-programming,tumorigenesis and *** studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the ***,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo *** LIN28 expression drops upon exit from the 2-cell stage,and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after *** cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus ***,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity,and its loss leads to nucleolar phase separation defects,ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor ***28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci,and thus de-repressed Dux and reduced rRNA ***28 knockout cells with nucleolar stress are more likely to assume a slowly cycling,translationally inert and anabolically inactive state,which is a part of previously unappreciated 2C-like transcriptional *** findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development.

Because the class of wideband noise is quite realistic in applications and is a good approximation to white noise, this paper focuses on the asymptotic properties of integro-differential systems with wideband noise perturbations. Using perturbed test function methods combined with the martingale averaging techniques, and weak convergence methods, it is shown that when the small parameter tends to zero, the underlying process converges weakly to a stochastic process satisfying a stochastic integro-differential equation. Inspired by the developments from [3] and [9], the novelty of our paper lies in the treatment of functional differential equations in which differential-integral equations are dealt with. Because of the features of the systems involved, different methods for estimates and proofs from the aforementioned references are developed in this paper. Not only are the results interesting from a mathematical point of view, but also they will be of great utility to a wide range of problems in control and optimization problems.

Ensemble Kalman filter-based targeted observation is one of the best methods for determining the optimal observational array for oceanic buoy deployment. This study proposes a new algorithm suitable for a‘cross-region and cross-variable’ approach by introducing a projection operator into the optimization process. A targeted observational analysis was conducted for El Ni?o–Southern Oscillation(ENSO) events in the tropical western Pacific for the Tropical Pacific Observation System(TPOS) 2020. The prediction target was at the Ni?o 3.4 region and the first 10 optimal observational sites detected reduced initial uncertainties by 70%, with the best observational array located where the Rossby wave signal dominates. At the vertical level, the most significant contribution was derived from observations near the *** study provides insights into understanding ENSO-related variability and offers a practical approach to designing an optimal mooring array. It serves as a scientific guidance for designing a TPOS observation network.

The dynamic force load in grinding process is considered as a crucial factor affecting the quality of parts,and a better understanding of the mechanism of force generation is conducive to revealing the evolution of material microstructure more *** this study,an iterative blending integrating grinding force model that comprehensively considers the impact of grain size and dislocation density evolution of the material is *** to the grinding kinematics,the interaction of grit-workpiece is divided into rubbing,plowing,and chip formation stages in each grinding *** this basis,the evolution of material microstructure in the current chip formation stage will affect the rubbing force in the next grinding arc through flow stresses,which in turn will influence the total grinding ***,the flow stress models in rubbing and chip formation stages are firstly established,and then the dislocation density prediction model is established experimentally based on the characteristics of grain *** effects of the evolution of grain size and dislocation density on the grinding forces during the grinding process are studied by means of iterative *** results indicate that the implementation of an iterative blending scheme is instrumental in obtaining a higher accurate prediction of the grinding force and a deeper insight of the influence mechanisms of materials microstructure on grinding process.

Objective： To examine the apoptotic effect of ent-llα-hydroxy-15-oxo-kaur-16-en-19-oic-acid （5F）, a compound isolated from Pteris semipinnata L （PsL）, in human lung cancer A549 cells. Methods： A549 cells were treated with 5F （0-80 lag/ml） for different time periods. Cytotoxicity was examined using a Ml-I- method. Cell cycle was examined using propidium iodide staining. Apoptosis was examined using Hoechst 33258 staining, enzyme-linked immunosorbent assay （ELISA） and caspase-3 activity analysis. Expression of representative apoptosis-related proteins was evaluated by Western blot analysis. Reactive oxygen species （ROS） level was measured using standard protocols. Potential interaction of 5F with cisplatin was also examined. Results： 5F inhibited the proliferation of A549 cells in a concentration- and time-dependent manner. 5F increased the accumulation of cells in sub-G1 phase and arrested the cells in the G2 phase. Exposure to 5F induced morphological changes and DNA fragmentation that are characteristic of apoptosis. The expression of p21 was increased. 5F exposure also increased Bax expression, release of cytochrome c and apoptosis inducing factor （AIF）, and activation of caspase-3. 5F significantly sensitized the cells to cisplatin toxicity. Interestingly, treatment with 5F did not increase ROS, but reduced ROS production induced by cisplatin. Conclusion： 5F could inhibit the proliferation of A549 cells by arresting the cells in G2 phase and by inducing mitochondrial-mediated apoptosis.
