Epstein-Barr virus infection and persistence in nasopharyngeal epithelial cells

Epstein-Barr virus(EBV) infection is closely associated with undifferentiated nasopharyngeal carcinoma(NPC), strongly implicating a role for EBV in NPC pathogenesis; conversely, EBV infection is rarely detected in normal nasopharyngeal epithelial tissues. In general, EBV does not show a strong tropism for infecting human epithelial cells, and EBV infection in oropharyngeal epithelial cells is believed to be lytic in nature. To establish life-long infection in humans, EBV has evolved efficient strategies to infect B cells and hijack their cellular machinery for latent infection. Lytic EBV infection in oropharyngeal epithelial cells, though an infrequent event, is believed to be a major source of infectious EBV particles for salivary transmission. The biological events associated with nasopharyngeal epithelial cells are only beginning to be understood with the advancement of EBV infection methods and the availability of nasopharyngeal epithelial cell models for EBV infection studies. EBV infection in human epithelial cells is a highly inefficient process compared to that in B cells, which express the complement receptor type 2(CR2) to mediate EBV infection. Although receptor(s) on the epithelial cell surface for EBV infection remain(s) to be identified, EBV infection in epithelial cells could be achieved via the interaction of glycoproteins on the viral envelope with surface integrins on epithelial cells, which might trigger membrane fusion to internalize EBV in cells. Normal nasopharyngeal epithelial cells are not permissive for latent EBV infection, and EBV infection in normal nasopharyngeal epithelial cells usually results in growth arrest. However, genetic alterations in premalignant nasopharyngeal epithelial cells, including p16 deletion and cyclin D1 overexpression, could override the growth inhibitory effect of EBV infection to support stable and latent EBV infection in nasopharyngeal epithelial cells. Understanding the role of PIN1 in hepatocellular carcinoma

PIN1 is a peptidyl-prolyl cis/trans isomerase that binds and catalyses isomerization of the specific motif comprising a phosphorylated serine or threonine residue preceding a proline(p Ser/Thr-Pro) in proteins. PIN1 can therefore induce conformational and functional changes of its interacting proteins that are regulated by proline-directed serine/threonine phosphorylation. Through this phosphorylation-dependent prolyl isomerization, PIN1 fine-tunes the functions of key phosphoproteins(e.g., cyclin D1, survivin, β-catenin and x-protein of hepatitis B virus) that are involved in the regulation of cell cycle progression, apoptosis, proliferation and oncogenic transformation. PIN1 has been found to be over-expressed in many cancers, including human hepatocellular carcinoma(HCC). It has been shown previously that overexpression of PIN1 contributes to the development of HCC invitro and in xenograft mouse model. In this review, we first discussed the aberrant transcription factor expression, miR NAs dysregulation, PIN1 gene promoter polymorphisms and phosphorylation of PIN1 as potential mechanisms underlying PIN1 overexpression in cancers. Furthermore, we also examined the role of PIN1 in HCC tumourigenesis by reviewing the interactions between PIN1 and various cellular and viral proteins that are involved in β-catenin, NOTCH, and PI3K/Akt/mT OR pathways, apoptosis, angiogenesis and epithelial-mesenchymal transition. Finally, the potential of PIN1 inhibitors as an anti-cancer therapy was explored and discussed.

The objective of this study was to characterize sympathetic nervous system （SNS） activity in girls with adolescent idiopathic scoliosis （AIS） and investigate the correlation of such activity with body weight and bone mass. Fifty-eight girls with AIS and 21 healthy controls were recruited. Abnormal growth pattern （lower weight, lower body mass index （BMI）, and longer arm span） and significantly increased adrenaline （AD） level, norepinephrine （NE） level, and low-frequency （LF） were found in girls with AIS. Circulating AD level, NE level, and LF power were negatively correlated with body weight and BMI but significantly correlated with bone quality.

Current fire retardants are known to be toxic to humans and our *** environmental-friendly flame retardants(FRs),protein-based flame retardants have been studied extensively recently,even though they are not *** this study,we designed,synthesized and tested a durable protein-based FR through the fusion of the adhesion domain from either mussel foot protein-5(mfp-5)or cellulose-binding domain(CBD)with flame retardant protein(SR protein and alpha casein).We first verified the expression of the recombinant proteins in Escherichia coli using Western ***,we coated the fusion protein(carrying cell lysates)to cotton fabrics and wood and verified with Infrared(IR)*** a vertical burning test and wood flammability test,we confirmed the flame retardancy of the materials after the protein *** the vertical burning test,the SR protein and alpha casein flame retardant proteins with the CBD adhesion domain showed a 50.0%and 43.3%increase in flame *** data is also consistent in the wood flame retardancy *** imaging experiments also suggested these new fire retardants can be preserved on the materials well even after ***,our results showed that flame-retardant proteins with adhesion domains are high potential candidates of green alternative flame retardants.

Vitreous haemorrhage:a population-based study of the incidence and risk factors in Taiwan

AIM：To report the epidemiology and incidence of vitreous hemorrhage and to evaluate risk factors for patients with vitreous hemorrhage（VH）in ***：A retrospective population-based *** a sample of one million subjects from all enrollees of the Taiwan Health Insurance programme. All data were obtained from the Taiwan Health Insurance Research Database,which contained patient sex,date of birth,all records of clinical visits and hospitalizations,and diagnosis codes as included in the International Classification of Diseases,Ninth Revision,Clinical Modification（ICD-9-CM）. The main outcome measures were the incidence and risk factors of ***：From 2001 to 2010,the database claim 4379 newly diagnosed cases were identified with VH. The average incidence of VH in Taiwan was 4.8 cases per ten-thousand person-years generally and increased with time especially in subjects who aged between 40 and 59 y and when the VH was associated with proliferative diabetic retinopathies or retinal vein occlusions. A de-finitely upward trends in the incidence of VH from 2001 to 2010 were noted（P-value for increasing trend 〈0.001）. Univariate Cox＇s regression analysis pointed out that older age（for 40-59,P〈0.001,HR=9.39; for ≥60,P〈0.001,HR=11.39）,male gender（P=0.03,HR=1.07）and subjects who had been prescribed anti-coagulation drug included aspirin,warfarinand clopidogrel（P〈0.001,HR=2.20）were significant risk factors for suffering from ***：The incidence of VH is estimated being 4.8 cases per 10 000 person-years in Taiwan. Age,male gender and having been prescribed anti-coagulation drugs are associated with the incidence of VH.

Pediatric Application of Dynamic Contrast-Enhanced MR Imaging (DCE-MR) in the management of Extra-Cranial Tumor: Experience in Routine Clinical Practice

Background: Dynamic contrast-enhanced MR imaging (DCE-MR) is becoming a widely accepted complementary method for diagnosing breast cancer and other cancers in adults. It is useful to predict tumor response to anticancer therapy and monitor the tumor response to the therapy. This form of imaging techniques has not been adequately explored in pediatric oncology patients. Objective: To determine the potential role of dynamic contrast-enhanced MR imaging (DCE-MR) in the diagnosis and treatment response monitoring of childhood and young adult extra-cranial tumors in routine clinical setting. Methods: children with suspected extra-cranial solid tumors, including newly diagnosed or follow-up cases of confirmed tumors, were recruited. DCE-MR was performed with intravenous injection of 0.1 mmol/kg contrast. The enhancement time curves were plotted and the enhancement patterns were categorized into type 1, 2 and 3 curves. Enhancement curve patterns and maximal enhancement intensity were compared with types of tumor in newly diagnosed cases. The preoperative percentiles of inactive area on the colour map were compared with the necrotic areas on histologic sections of the resected specimens in follow-up cases. Pearson chi-square test and Unpaired two-sample t-test were used for statistical analysis. Results: There were 36 patients, involving 28 malignant and 8 benign cases. There were 14 type 3 curves, (all of them were malignant tumors), 6 type 2 curves and 16 type 1 curves. All the benign cases (n = 8) demonstrated type 1 curve (accuracy & negative predictive value = 100%). All the malignant cases after treatment showed type 2 or 1 curve. For those cases with operation done afterwards, the extent of tumor necrosis was correlated closely with pathology findings (accuracy = 93.3%). Somaclonal variation in tissue culture is a common phenomenon induced by various external or internal environmental conditions,resulting in heritable or non-heritable alterations in gene expression. One crucial mechanism involved in plant growth and development is epigenetic regulation. A highly dynamic epigenome can respond to environmental changes by regulating gene expression. DNA methylation is one of these epigenetic modifications that can alter gene expression in tissue-cultured pineapple plants. The underlying mechanism of such somaclonal variations in pineapple and the epigenetic regulation involvement in somaclonal variations has not been studied. This study performed DNA methylome and transcriptome sequencing of wild-type(WT) and mutant pineapple plants(WS, HW, and TW). We observed altered DNA methylation patterns in chlorophyll development in the mutants. Specifically, we noticed that the methylation levels in the CHG and CHH contexts were lower in the gene body regions compared to the upstream and downstream regions. We identified several thousand differentially methylated regions(DMRs) located at the gene body regions, some of which overlapped with the differentially expressed genes(DEGs). Functional enrichment analyses suggested that these genes were involved in chlorophyll metabolism. Thus, our results revealed that the transcriptional regulation of many chlorophyll metabolic essential genes could be regulated by DNA methylation caused by somaclonal variations and provided insights into epigenetic mechanisms underlying the regulation of chlorosis in pineapple plants.

Second malignant neoplasms in childhood cancer survivors in a tertiary paediatric oncology centre in Hong Kong, china

Background childhood cancer survivors were at risk of development of second malignant neoplasms. The aim of this study is to evaluate the incidence, risk factors and outcome of second malignant neoplasms in childhood cancer survivors in a tertiary paediatric oncology centre in Hong Kong, china. Methods We performed a retrospective review of patients with childhood cancer treated in children＇s Cancer Centre in Prince of Wales Hospital, Hong Kong, china between May 1984 and June 2009. Case records of patients who developed second malignant neoplasms were reviewed. Results Totally 1374 new cases aged less than 21-year old were treated in our centre in this 25-year study period. Twelve cases developed second malignant neoplasms with 10-year and 20-year cumulative incidence of 1.3% （95% confidence interval 0.3%-2.3%） and 2.9% （95% confidence interval 1.1%-4.7%） respectively. Another 4 cases were referred to us from other centres for the management of second malignant neoplasms. In this cohort of 16 children with second malignant neoplasms, the most frequent second malignant neoplasms were acute leukemia or myelodysplastic syndrome （n--6） and central nervous system tumor （n=4）. Median interval between diagnosis of primary and second malignant neoplasms was 7.4 years （range 2.1-13.3 years）. Eight patients developed second solid tumor within the previous irradiated field. Radiotherapy significantly increased the risk of development of second solid tumor in patients with acute lymphoblastic leukemia （P=0.027）. Seven out of 16 patients who developed second malignant neoplasms had a family histo＇ry of cancer among the first or second-degree relatives. Nine patients died of progression of second malignant neoplasms, mainly resulted from second central nervous system tumor and osteosarcoma. Conclusions Cumulative incidence of second cancer in our centre was comparable to western countries. Radiotherapy was associated with second solid tumour among patients with acute lymphoblast
