The Effects of Several Types of Induced Abiotic Stress on Cephalaria joppensisGermination under Controlled Conditions

The recently domesticated species, Cephalaria joppensis (CJ), is emerging as a new alternative forage crop in Israel. It has high biomass potential and nutritional values that are comparable to forage wheat. However, CJ emerges slowly under cold conditions, which hinders its development as a major winter crop. Additional tolerance for abiotic stress would improve its performance as a forage crop. We examined the effects of several abiotic factors (i.e., cold, salinity, drought and pH) on CJ germination under controlled conditions. The effect of temperature was studied by incubating seeds at different temperatures between 7°C and 35°C. The effects of salinity, osmotic potential and pH were tested by incubating seeds at different NaCl, PEG and pH levels, respectively. Temperature, salinity and osmotic potential significantly affected germination;whereas pH did not. Temperature did not affect the final proportion of germinated seeds, but did affect other germination-rate variables, indicating that germination rate might be the limiting factor under field conditions. Salinity also affected germination-rate variables, but not the proportion of seeds that germinated. Notably, CJ was found to be relatively resistant to high salt concentrations, with a 273 mM NaCl threshold for germination, indicating its potential as a relatively salt-tolerant forage crop. Both the proportion of germinated seeds and the germination rate were highly sensitive to the osmotic-potential treatments, indicating that drought resistance will remain the biggest challenge for CJ. This study provides baseline data for a rapid and efficient system for further screening for abiotic-stress tolerance among wild and cultivated lines of CJ.

Computer analysis of N-(β-D-galactopyranosyl)-thiosemicarbazide compounds by in silico method revealed high probability of antibacterial (antimycobacterial), anti-tuberculosis (antituberculosic), antiviral (Influenza), antitumor (antineoplastic) 9 > Pa > 0.5 and with a low probability of cytotoxic/cytostatic (cytostatic/cytotoxic) activities. An experimental study by in vitro and in vivo methods allowed us to conclude that studied new synthetic compound N-(β-D-galactopyranosyl)-thiosemicarbazide in the studied concentrations has a pronounced bactericidal and bacteriostatic effects.

Effect of the Technological Status of Small Cow-Calf Farm Producers on the Induction to Resumption of Ovarian Activity of Dual-Purpose Cattle Raised under Topical Conditions

The aim of the present study was to evaluate the impact of the technological level in small-scale dual-purpose cow-calf Bos indicus female operations on the resumption of the ovarian activity post-partum. A total of 13 small scale dual-purpose Bos indicus × Bos taurus operations were included in this experiment and they were classified according to their technological status as: high technological status (HT), medium technological status (MT) and low technological status (LT). Ninety-three mature cows were treated with either a CIDR (controlled internal drug release device) with estradiol benzoate (CIDR+EB) or alone (CIDR). At day 9 all animals received intravaginally a CIDR for nine days. At day 0, the CIDR was withdrawn and 24 hours later one dose of estradiol benzoate was administered to 51 cows (17 in HT, 17 in MT and 17 in LT), the remaining animals (n = 42) did not receive the estradiol benzoate administration (14 in HT, 13 in MT and 15 in LT). Comparisons were performed on serum progesterone concentrations after CIDR treatment. By day 7 after implant withdrawal, the proportion of cows that resume ovarian activity detected by progesterone concentration increased in all technological level farms, no statistical main effect was observed within technological level. By day 17 there was a significant main effect of technological level (P = 0.05) on the proportion of animal that resumed the estrous cycles, but neither a main effect of treatment (P = 0.97) nor an interaction between technological level and treatment (P = 0.98). Furthermore, technological level of the farm showed a tendency (P = 0.07) to affect that resume the estrous activity, and a significant effect of treatment was observed (P = 0.005) but no interaction between technological level and treatment. A Comparative Study on Pharmacokinetics of Tricin, a Flavone from Gramineous Plants with Antiviral Activity

Tricin (and tricin containing plant extracts) has been shown to exert a pronounced antiviral activity, high radical scavenging activity and is favored for its safety profile. In the present study we have analyzed the pharmacokinetics of tricin after a single intravenous and oral administration to Wistar rats of an ethanol extract of Calamagrostis Adans and Deschampsia Beauv plants (test agent) at different doses. Tricin concentrations in blood plasma and blood cells were measured at different time points. Two-compartment (for intravenous injection) and one compartment (for oral administration) models were used for the analysis of tricin pharmacokinetics. The results showed that the pharmacokinetics of tricin after intravenous injection of test agent has a pronounced biphasic character, is well described by a two-compartment pharmacokinetic model, and is characterized by non-linear dose-dependence. The pharmacokinetics of tricin administered orally is characterized by a high rate of absorption from the gastrointestinal tract into the blood and rather slow elimination, which leads to a large volume of distribution in the body and a fairly high bioavailability. The obtained results indicate the advantages of the oral route of administration over the intravenous route.

Novel 3-phenyl/pyridinyl-trans-2-(aryl/heteryl)vinyl-3H-quinazolin-4-ones 3a,b, 4a, 5a, 7a and their 6,7-difluoro de- rivatives 3c,d, 4b, 5b, 7b have been obtained by condensation of the correspondent 2-methylquinazolin-4-ones 1, 6 with aromatic (heterocyclic) aldehydes in the presence of ZnCl2 (AcONa) or by the reaction of 2-methyl-3,1-benzoxazin-4- ones 2 with the Shiff bases. Effects of aryl(heteryl) substituents on photophysical properties of (aryl/heteryl) quinazolinylethenes have been studied.

Spin-Labelled 1-Ethyl-1-Nitrosourea Prevents Doxorubicin and Bleomycin-Induced Oxidative Stress in Lungs, Hearts and Kidneys of Tumour-Bearing Mice

This study was carried out to determine the possible protective effect of 1-ethyl-3-[4-(2, 2, 6, 6-tetramethylpiperidine-1-oxyl)]-1-nitrosourea (SLENU), recently synthesised in our laboratory on doxorubicin and bleomycin-induced oxidative toxicity in C57 black tumour-bearing mice. Specifically, alterations in some biomarkers of oxidative stress, such as lipid peroxidation products measured as malondialdehyde (MDA) levels and activities of the antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT), were studied in lung, heart and kidney homogenates isolated from C57 black tumor-bearing mice after i.p. treatment with solutions of DOX (60 mg/kg) and BLM (60 mg/kg). The same biomarkers were also measured after i.p. pretreatment of mice with SLENU (100 mg/kg). After treatment with doxorubicin, heart and kidney homogenates of mice had significantly higher productions of lipid peroxidation compared to lung homogenates. It was accompanied by increased activity of the antioxidant defence enzyme superoxide dismutase and decreased activity of catalase. Bleomycin-induced oxidative stress was confirmed by significantly higher production of lipid peroxidation in lungs compared to heart homogenates, elevation of the anti-oxidant activity of superoxide dismutase and decreased activity of catalase enzymes. After pre-treatment of the mice with SLENU, the levels of all studied oxidative stress biomarkers were significantly improved in comparison with those of the mice treated alone with either bleomycin, or doxorubicin. The present results and those from a previously demonstrated superoxide scavenging activities (SSA) of the nitrosourea SLENU have enabled us to explain the protective effect of the spin-labelled nitrosourea on doxorubicin and bleomycin-induced oxidative stress by scavenging of??O2- and increased NO release.

将吡咯衍生物单体(碘化N,N,N-三甲基-(2-吡咯-1-乙基))进行氧化聚合,合成水溶性聚吡咯衍生物,将该产物与V2O5溶胶混合,进行聚合物溶液插层反应,合成聚(N,N,N-三甲基-(2-吡咯-1-乙基))(PTPAI)/VXG复合薄膜。采用电化学阻抗谱(EIS)和循环伏安(CV)对其电学性能进行了测试。结果表明:该复合薄膜的等效电路为R1(Q1[R2W1]),Rs=160.215Ω,Rct=6278.86Ω,双电层电容CPE=1.73216×10-4F,离子迁移系数4.37×10-12cm2/s(-0.5V)。PTPAI提高了VXG的氧化还原电位,且出现另一个较小的氧化还原峰电位。

Photodynamic therapy(PDT)is one of the most appealing photonic modalities for cancer treatment based on anticancer activity of light-induced photosensitizer-mediated reactive oxygen species(ROS),but a limited depth of light penetration into tissues does not make possible the treatment of deep-seated neoplasms and thus complicates its widespread clinical ***,we introduce the concept of genetically encoded bioluminescence resonance energy transfer(BRET)-activated PDT,which combines an internal light source and a photosensitizer(PS)in a single-genetic construct,which can be delivered to tumors seated at virtually unlimited depth and then triggered by the injection of a substrate to initiate their *** illustrate the concept,we engineered genetic NanoLuc-miniSOG BRET pair,combining NanoLuc luciferase flashlight and phototoxic flavoprotein miniSOG,which generates ROS under luciferase-substrate *** prove the concept feasibility in mice bearing NanoLuc-miniSOG expressing tumor,followed by its elimination under the luciferase-substrate ***,we demonstrate a targeted delivery of NanoLuc-miniSOG gene,via tumor-specific lentiviral particles,into a tumor,followed by its successful elimination,with tumor-growth inhibition(TGI)coefficient exceeding 67%,which confirms a great therapeutic potential of the proposed *** conclusion,this study provides proof-of-concept for deep-tissue“photodynamic”therapy without external light source that can be considered as an alternative for traditional PDT.
