From single to combinatorial therapies in spinal cord injuries for structural and functional restoration

Spinal cord injury results in paralysis, sensory disturbances, sphincter dysfunction, and multiple systemic secondary conditions, most arising from autonomic dysregulation. All this produces profound negative psychosocial implications for affected people, their families, and their communities;the financial costs can be challenging for their families and health institutions. Treatments aimed at restoring the spinal cord after spinal cord injury, which have been tested in animal models or clinical trials, generally seek to counteract one or more of the secondary mechanisms of injury to limit the extent of the initial damage. Most published works on structural/functional restoration in acute and chronic spinal cord injury stages use a single type of treatment: a drug or trophic factor, transplant of a cell type, and implantation of a biomaterial. Despite the significant benefits reported in animal models, when translating these successful therapeutic strategies to humans, the result in clinical trials has been considered of little relevance because the improvement, when present, is usually insufficient. Until now, most studies designed to promote neuroprotection or regeneration at different stages after spinal cord injury have used single treatments. Considering the occurrence of various secondary mechanisms of injury in the acute and sub-acute phases of spinal cord injury, it is reasonable to speculate that more than one therapeutic agent could be required to promote structural and functional restoration of the damaged spinal cord. Treatments that combine several therapeutic agents, targeting different mechanisms of injury, which, when used as a single therapy, have shown some benefits, allow us to assume that they will have synergistic beneficial effects. Leishmaniasis is considered as an emerging, uncontrolled disease and is endemic in 98 countries. Annually, about 2 million cases of cutaneous and 500000 cases of visceraltype leishmaniasis are recorded and 60000 persons died from the disease. In Mexico,cutaneous leishmaniasis is known as chiclero's ulcer and is reported in 22 states, it is considered as a health problem. For its treatment, pentavalent antimonial drugs are administered. These drugs cause severe side effects, are costly. Drug-resistant cases have been reported and have been developing for over 70 years. One alternative to the drugs that are currently available is to find active molecules in medicinal plants. Dihydrocorynantheine, corynantheine and corynantheidine are active against Leishmania major,while harmane, pleiocarpin, buchtienin, luteolin and quercetin are active against Leishmania donovani. In Mexico, about 20 medicinal plants have been evaluated against Leishmania mexicana, among which the most active are Tridax procumbens, Lonchocarpus xuul and Pentalinon andrieuxii. From these plants, active compounds with IC_(50)≤30 mg/mL or m M have been isolated, such as 3(S)-16,17-didehydrofalcarinol or Oxylipin, cholestra-4,20,24-trien-3-one or pentalinosterol, 24-methylcholest-4-24(28)-dien-3-one, cholest-4-en-3-one, 6,7-dihydroneridie-none, neridienone, cholest-5,20,24-trien-3β-ol, and isocordoin. Today, only pentalinonsterol has been synthesized and assayed in the visceral leishmaniasis experimental model using BALB/c mice infected with Leishmania donovani. Liposome formulation of this compound administered by intravenous route at 2.5 mg/kg showed a significant reduction of parasite load in mouse liver and spleen.

Objective:To estimate to what extent the mixture of ursolic acid and oleanolic acid,in addition to the antitubercular standard regime,affects the hepatotoxicity ***:Liver injury was induced in male BALB/c mice by administering,per os and daily for 11 weeks,a combination of anti-Tubercular(anti-TB) agents Rifampicin(10 mg/kg),Isoniazid(10 mg/kg),and Pyrazinamide(30 mg/kg).The ursolic acid and oleanolic acid mixture at doses of 100 or 200 μg/mouse/day was subcutaneously injected throughout the entire study period(11 weeks).Biochemical and hematological analysis was supplemented by liver histological ***:Animals treated with the mixture of triterpenic acids exhibited significantly decreased aspartate transaminase and alanine aminotransferase levels and amelioration of the histopathological alterations produced by the anti-TB ***:The triterpene mixture is able to prevent the steatosis induced by the anti-TB drugs.

Objective:To identify the anti-inflammatory activity through two murine models and in the median Lethal Dose(LD_(50)) of three dietary supplements that contain Moussonia ***:The anti-inflammatory activity of three dietary supplements(Cicatrisan/Gastricus^(R),Gastinol^(R),and Gastrovita^(R)) EtOH extracts was evaluated by TPA and by carrageenan murine models;also,median Lethal Dose(LD_(50)) was *** was quantified by High-Performance Liquid Chromatography.β-sitosterol,stigmasterol and the mixture of ursolic and oleanolic acids were identified in all supplements by TLC;however,none of these dietary supplements contain ***:For the TPA model,Cicatrisan/Gastricus^(R)generated a notable effect with 38.24%*** in the carrageenan model,it also exhibited noteworthy anti-inflammatory activity of ear edema with 66.39%of paw edema inhibition at 150 mg/kg,followed by Gastinol^(R) and Gastrovita^(R) with 50%at 300 mg/***,LD_(50) was >2 g/kg for all supplements,when was administered intragastrically and Body Weight(BW) gain in mice was not altered after 14 ***:Of the three food supplements containing ***,only the EtOH extract from the Cicatrisan/Gastricus^(R)formulation(tablets) showed significant anti-inflammatory activity in both experimental models and the LD_(50) was > 2 g/kg.

Drug-induced liver injury encompasses a spectrum of diseases ranging from mild biochemical abnormalities to acute liver failure; example of this scenery is hepatotoxicity caused by the first-line antituberculous drugs isoniazid, rifampin and pyrazinamide, which are basic for treatment of drug-sensible and drug-resistant tuberculosis. In the search for pharmacological alternatives to prevent liver damage, antitubercular drugs have been the subject of numerous studies and published reviews, a great majority of them carried out by Asian countries. At the same time, hepatoprotectors from plant source are now emerging as a possible alternative to counteract the toxic effects of these therapeutic agents. The present review aims to highlight the most recent studies on the subject, based information published in scientific databases such as Scopus and Pub Med.

instability associated Land cover change can lead to slope by accelerating erosive processes with agriculture, forestry, and infrastructure. The Rio Chiquito-Barranca del Muerto subbasin has experienced an increase in land cover change due to government programs and the establishment of agricultural and urban areas. The aim of this study was to provide a model to map the susceptibility to gravitational processes along sites where anthropogenic land cover change has occurred. The method was based on the stratification of the subbasin according to landforms and cartographic variables. These variables were used in a multi- criteria assessment to assign weights according to their contribution to the onset of new gravitationalprocesses. Those weights were used to create a susceptibility map based on a weighted linear sum. The accuracy of the resulting map was validated in an error matrix with a random stratified design based on susceptibility classes per landform. The results produced a map of areas with susceptibility to gravitational processes due to land cover change; this susceptibility is very high in the undifferentiated pyroclastic slope and limestone mountain, where it derives not only from anthropogenie effects on natural vegetation cover, but also from steep slopes, weathered materials, low apparent density, high erosivity, and previous gravitational processes. The results support other studies that concluded that loss of vegetation is a triggering factor in the formation of gravitational processes, but also show that excessive reforestation can increase gravitational processes.

The Rheic Ocean was one of the most important oceans of the Paleozoic Era. It lay between Laurentia and Gondwana from the Early Ordovician and closed to produce the vast Ouachita- Alleghanian-Variscan orogen during the assembly of Pangea. Rifting began in the Cambrian as a continua- tion of Neoproterozoic orogenic activity and the ocean opened in the Early Ordovician with the separation of several Neoproterozoic arc terranes from the continental margin of northern Gondwana along the line of a former suture. The rapid rate of ocean opening suggests it was driven by slab pull in the outboard Iapetus Ocean. The ocean reached its greatest width with the closure of Iapetus and the accretion of the peri- Gondwanan arc terranes to Laurentia in the Silurian. Ocean closure began in the Devonian and continued through the Mississippian as Gondwana sutured to Laurussia to form Pangea. The ocean consequently plays a dominant role in the Appalachian-Ouachita orogeny of North America, in the basement geology of southern Europe, and in the Paleozoic sedimentary, structural and tectonothermal record from Middle America to the Middle East. Its closure brought the Paleozoic Era to an end.

We describe the valued gabriel quiver of a wedge product of coalgebras and study the category of comodules of a semiprime coalgebra. In particular, we prove that any monomial semiprime k-tame fc-tame coalgebra is string. We also prove a version of Eisenbud-Griffith theorem for coalgebras, namely, any hereditary semiprime strictly quasi-finite coalgebra is serial.
