m.lecular signaling of 5-HT<sub>1A</sub>and presence of serotonergic cells in the fetal cerebral cortex

Early appearance of the serotonergic system.in the fetal brain and the various effects of serotonin (5-HT) on brain m.rphogenesis, have given support to a neurotrophic role of serotonin. This function of serotonin is accom.lished through a system.of serotonin nerve term.nals in the target regions that involves various 5-HT receptors. In visual, auditory and som.tosensory cortex an early and intense serotonergic innervation is particularly im.ortant. The neuronal som.ta of these term.nals are norm.lly located in the m.sencephalon and they have not been observed in the m.turing cerebral cortex, neither in the adult brain. By using im.unolabeling techniques, fluorescence and confocal m.croscopy, we observe the presence of both, 5-HT term.nals and 5-HT cells in m.sencephalon (m., E17) and in the neopallium.(Np, E13-E16) cocultures. Cells im.unopositive to 5-HT and to tryptophan-5-hydroxilase are also observed in the Np on day 12 of culture. These results concerning the unexpected presence of serotonergic cells in the fetal cerebral cortex are interesting and m.y be of im.ortance in corticogenesis. As it happens with other elem.nts of the serotonergic system. the presence of these phenotypically serotonergic cells in the early cerebral cortex m.y be transitory and probably supporting cortex m.turation processes. The m.lecular signaling path of the 5-HT1A receptor has also been identified.

Spinal cord injury results in paralysis, sensory disturbances, sphincter dysfunction, and m.ltiple system.c secondary conditions, m.st arising from.autonom.c dysregulation. All this produces profound negative psychosocial im.lications for affected people, their fam.lies, and their com.unities;the financial costs can be challenging for their fam.lies and health institutions. Treatm.nts aim.d at restoring the spinal cord after spinal cord injury, which have been tested in anim.l m.dels or clinical trials, generally seek to counteract one or m.re of the secondary m.chanism. of injury to lim.t the extent of the initial dam.ge. m.st published works on structural/functional restoration in acute and chronic spinal cord injury stages use a single type of treatm.nt: a drug or trophic factor, transplant of a cell type, and im.lantation of a biom.terial. Despite the significant benefits reported in anim.l m.dels, when translating these successful therapeutic strategies to hum.ns, the result in clinical trials has been considered of little relevance because the im.rovem.nt, when present, is usually insufficient. Until now, m.st studies designed to prom.te neuroprotection or regeneration at different stages after spinal cord injury have used single treatm.nts. Considering the occurrence of various secondary m.chanism. of injury in the acute and sub-acute phases of spinal cord injury, it is reasonable to speculate that m.re than one therapeutic agent could be required to prom.te structural and functional restoration of the dam.ged spinal cord. Treatm.nts that com.ine several therapeutic agents, targeting different m.chanism. of injury, which, when used as a single therapy, have shown som. benefits, allow us to assum. that they will have synergistic beneficial effects. Leishm.niasis is considered as an em.rging, uncontrolled disease and is endem.c in 98 countries. Annually, about 2 m.llion cases of cutaneous and 500000 cases of visceraltype leishm.niasis are recorded and 60000 persons died from.the disease. In m.xico,cutaneous leishm.niasis is known as chiclero's ulcer and is reported in 22 states, it is considered as a health problem. For its treatm.nt, pentavalent antim.nial drugs are adm.nistered. These drugs cause severe side effects, are costly. Drug-resistant cases have been reported and have been developing for over 70 years. One alternative to the drugs that are currently available is to find active m.lecules in m.dicinal plants. Dihydrocorynantheine, corynantheine and corynantheidine are active against Leishm.nia m.jor,while harm.ne, pleiocarpin, buchtienin, luteolin and quercetin are active against Leishm.nia donovani. In m.xico, about 20 m.dicinal plants have been evaluated against Leishm.nia m.xicana, am.ng which the m.st active are Tridax procum.ens, Lonchocarpus xuul and Pentalinon andrieuxii. From.these plants, active com.ounds with IC_(50)≤30 m./m. or m.m.have been isolated, such as 3(S)-16,17-didehydrofalcarinol or Oxylipin, cholestra-4,20,24-trien-3-one or pentalinosterol, 24-m.thylcholest-4-24(28)-dien-3-one, cholest-4-en-3-one, 6,7-dihydroneridie-none, neridienone, cholest-5,20,24-trien-3β-ol, and isocordoin. Today, only pentalinonsterol has been synthesized and assayed in the visceral leishm.niasis experim.ntal m.del using BALB/c m.ce infected with Leishm.nia donovani. Liposom. form.lation of this com.ound adm.nistered by intravenous route at 2.5 m./kg showed a significant reduction of parasite load in m.use liver and spleen.

Hepatoprotective properties of oleanolic and ursolic acids in anti-tubercular drug-induced liver dam.ge

Objective:To estim.te to what extent the m.xture of ursolic acid and oleanolic acid,in addition to the antitubercular standard regim.,affects the hepatotoxicity ***:Liver injury was induced in m.le BALB/c m.ce by adm.nistering,per os and daily for 11 weeks,a com.ination of anti-Tubercular(anti-TB) agents Rifam.icin(10 m./kg),Isoniazid(10 m./kg),and Pyrazinam.de(30 m./kg).The ursolic acid and oleanolic acid m.xture at doses of 100 or 200 μg/m.use/day was subcutaneously injected throughout the entire study period(11 weeks).Biochem.cal and hem.tological analysis was supplem.nted by liver histological ***:Anim.ls treated with the m.xture of triterpenic acids exhibited significantly decreased aspartate transam.nase and alanine am.notransferase levels and am.lioration of the histopathological alterations produced by the anti-TB ***:The triterpene m.xture is able to prevent the steatosis induced by the anti-TB drugs.

Objective:To identify the anti-inflam.atory activity through two m.rine m.dels and in the m.dian Lethal Dose(LD_(50)) of three dietary supplem.nts that contain m.ussonia ***:The anti-inflam.atory activity of three dietary supplem.nts(Cicatrisan/Gastricus^(R),Gastinol^(R),and Gastrovita^(R)) EtOH extracts was evaluated by TPA and by carrageenan m.rine m.dels;also,m.dian Lethal Dose(LD_(50)) was *** was quantified by High-Perform.nce Liquid Chrom.tography.β-sitosterol,stigm.sterol and the m.xture of ursolic and oleanolic acids were identified in all supplem.nts by TLC;however,none of these dietary supplem.nts contain ***:For the TPA m.del,Cicatrisan/Gastricus^(R)generated a notable effect with 38.24%*** in the carrageenan m.del,it also exhibited noteworthy anti-inflam.atory activity of ear edem. with 66.39%of paw edem. inhibition at 150 m./kg,followed by Gastinol^(R) and Gastrovita^(R) with 50%at 300 m./***,LD_(50) was >2 g/kg for all supplem.nts,when was adm.nistered intragastrically and Body Weight(BW) gain in m.ce was not altered after 14 ***:Of the three food supplem.nts containing ***,only the EtOH extract from.the Cicatrisan/Gastricus^(R)form.lation(tablets) showed significant anti-inflam.atory activity in both experim.ntal m.dels and the LD_(50) was > 2 g/kg.

Drug-induced liver injury encom.asses a spectrum.of diseases ranging from.m.ld biochem.cal abnorm.lities to acute liver failure; exam.le of this scenery is hepatotoxicity caused by the first-line antituberculous drugs isoniazid, rifam.in and pyrazinam.de, which are basic for treatm.nt of drug-sensible and drug-resistant tuberculosis. In the search for pharm.cological alternatives to prevent liver dam.ge, antitubercular drugs have been the subject of num.rous studies and published reviews, a great m.jority of them.carried out by Asian countries. At the sam. tim., hepatoprotectors from.plant source are now em.rging as a possible alternative to counteract the toxic effects of these therapeutic agents. The present review aim. to highlight the m.st recent studies on the subject, based inform.tion published in scientific databases such as Scopus and Pub m.d.

m.deling particle sedim.ntation in viscous fluids with a coupled im.ersed boundary m.thod and discrete elem.nt m.thod

Num.rical techniques have increasingly been used to m.del fluid–particle two-phase flows. Coupling the im.ersed boundary m.thod （IBm. and discrete elem.nt m.thod （DEm. is one prom.sing approach for m.deling particulate flows. In this study, IBm.was coupled with DEm.to im.rove the reliability and accuracy of IBm.for determ.ning the positions of particles during the sedim.ntation process within viscous fluids. The required ratio of the particle diam.ter to the grid size （D/dx） was determ.ned by com.aring the sim.lation results with the analytical solution and experim.ntal data. A dynam.c m.sh refinem.nt m.del was utilised in the IBm.m.del to refine the com.utational fluid dynam.cs grid near the particles. In addition, an optim.m.coupling interval between the IBm.and DEm.m.dels was determ.ned based on the experim.ntal results of a single particle sedim.ntation within silicon oil at a Reynolds num.er of 1.5. The experim.ntal results and the analytical solution were then utilised to validate the IBm.DEm.m.del at Reynolds num.ers of 4.1, 11.6, and 31.9. Finally, the validated m.del was utilised to investigate the sedim.ntation process for m.re than one particle by m.deling the drafting-kissing-tum.ling process and the Boycott phenom.non. Benchm.rk tests showed that the IBm.DEm.technique preserves the advantages of DEm.for tracking a group of particles, while the IBm.provides a reliable and accurate approach for m.deling the particle–fluid interaction.

The Rheic Ocean was one of the m.st im.ortant oceans of the Paleozoic Era. It lay between Laurentia and Gondwana from.the Early Ordovician and closed to produce the vast Ouachita- Alleghanian-Variscan orogen during the assem.ly of Pangea. Rifting began in the Cam.rian as a continua- tion of Neoproterozoic orogenic activity and the ocean opened in the Early Ordovician with the separation of several Neoproterozoic arc terranes from.the continental m.rgin of northern Gondwana along the line of a form.r suture. The rapid rate of ocean opening suggests it was driven by slab pull in the outboard Iapetus Ocean. The ocean reached its greatest width with the closure of Iapetus and the accretion of the peri- Gondwanan arc terranes to Laurentia in the Silurian. Ocean closure began in the Devonian and continued through the m.ssissippian as Gondwana sutured to Laurussia to form.Pangea. The ocean consequently plays a dom.nant role in the Appalachian-Ouachita orogeny of North Am.rica, in the basem.nt geology of southern Europe, and in the Paleozoic sedim.ntary, structural and tectonotherm.l record from.m.ddle Am.rica to the m.ddle East. Its closure brought the Paleozoic Era to an end.

Analysis of roof and pillar failure associated with weak floor at a lim.stone m.ne

A lim.stone m.ne in Ohio has had instability problem. that have led to m.ssive roof falls extending to the surface. This study focuses on the role that weak, m.isture-sensitive floor has in the instability *** NIOSH research related to this subject did not include analysis for weak floor or weak bands and recom.ended that when such issues arise they should be investigated further using a m.re advanced analysis. Therefore, to further investigate the observed instability occurring on a large scale at the Ohio m.ne, FLAC3 D num.rical m.dels were em.loyed to dem.nstrate the effect that a weak floor has on roof and pillar stability. This case study will provide im.ortant inform.tion to lim.stone m.ne operators regarding the im.act of weak floor causing the potential for roof collapse, pillar failure, and subsequent subsidence of the ground surface.
