Pre-ischemia electro-acupuncture potentiates the expression of Bcl-2 and transforming growth factor-beta 1 in rat brains

The expression of the anti-apoptotic molecules Bcl-2 and transforming growth factor-beta 1 is known to confer protective effects on the cerebral ischemia-reperfusion *** current study investigated the expression levels of Bcl-2 and transforming growth factor-beta 1 in response to multiple pre-ischemia electro-acupuncture at acupoints Zusanli（ST36）and Fengchi（gB20） *** were divided into five groups：uninjured,control,non-acupoint,gB20 and ST36. Rats in the non-acupoint,gB20 and ST36 groups received 30 minutes（3 times or 18 times）of electro-acupuncture stimulation before experimental cerebral ischemia was ***-2 and transforming growth factor-beta 1 were found to be significantly increased in the ST36 groups with either 3 or 18 electro-acupuncture treatments（P〈0.05）.The production was higher with 18 electro-acupuncture treatments in the ST36 groups（P〈0.05）.In the gB20 groups,significant increase was only observed in transforming growth factor-beta 1 with 18 electro-acupuncture treatments（P〈0.05）.No significant elevation of the level of transforming growth factor-beta 1 was observed in the non-acupoint ***,the production of Bcl-2 increased with 18 treatments in the non-acupoint groups（P〈0.05）.The data suggest that multiple pre-ischemia electro-acupuncture at ST36 was effective in conferring neuroprotective effect on the brain by means of upregulation of Bcl-2 and transforming growth factor-beta 1 and the effect was increase with the number of treatment.

Use of the DSI/Ponemah jacketed external telemetry system for ECg assessments in dog telemetry

OBJECTIVE To demonstrate that the DSI /Ponemah Jet System(v5.00) can generate high quality ECg data with current methods,enabling cardiovascular function to be assessed in free moving dogs in their home *** Four male beagle dogs were used on this *** electrodes were placed on each animal and were connected to each JET *** JET device was then placed into a pocket of an externally worn animal jacket for each *** JET device transmits to a Bluetooth? *** Bluetooth? receiver can receive signals from up to 6 JET devices depending upon the device *** JET device can transmit data up to 10 meters with negligible interference and no cross-talk exists between *** animals underwent acclimatization to jackets prior to commencement of *** dog was placed into a pen and ECg measurements taken simultaneously using the DSI /Ponemah Jet *** were recorded for 1 hour pre-dose and continually for the following 23 hours under control conditions,then dosed with ascending doses of Moxifloxacin at intervals of 48 hours between each *** AND CONClUSION Prolonged QT and QTcV(Van de Water's correction) interval was observed for animals dosed at 100 mg·kg-1 from 4-23 h *** QTcV effects(19% increase with 100 mg·kg-1 Moxifloxacin vs control).The DSI telemetry system performed well when testing the dog to the known pharmacological profile of *** sensitively and efficiently acquired ECg data from the beagle dogs and demonstrated the sensitivity of the system to detect test article induced QT *** allows a colony of dogs to be dedicated for toxicology and safety pharmacology *** DSI /ponemah JET system enables flexibility in study design and can be utilized on a range of study types to facilitate high quality ECgs and reduce the requirement for repeat dosing safety pharmacology *** data generated from this investigation demonstrate the DSI /ponemah JET system(v5.

Investigation of fertility and early embryonic toxicity of caffeine in rats

Haplotype resolved chromosome level genome assembly of Citrus australis reveals disease resistance and other citrus specific genes

Mice with a heterozygous lrp6 deletion have impaired fracture healing

Bone fracture non-unions, the failure of a fracture to heal, occur in 10%-20% of fractures and are a costly and debilitating clinical problem. The Wnt/fl-catenin pathway is critical in bone development and fracture healing. Polymorphisms of linking low-density lipoprotein receptor-related protein 6 （lRP6）, a Wnt-binding receptor, have been associated with decreased bone mineral density and fragility fractures, although this remains controversial. Mice with a homozygous deletion of lrp6 have severe skeletal abnormalities and are not viable, whereas mice with a heterozygous deletion have a combinatory effect with lrp5 to decrease bone mineral density. As fracture healing closely models embryonic skeletal development, we investigated the process of fracture healing in mice heterozygous for lrp6 （lrp6~/-） and hypothesized that the heterozygous deletion of lrp6 would impair fracture healing. Mid-diaphyseal femur fractures were induced in lrp6~^- mice and wild-type controls （lrp6~/~）. Fractures were analyzed using micro-computed tomography （~CT） scans, biomechanical testing, and histological analysis. lrp6~/- mice had significantly decreased stiffness and strength at 28 days post fracture （PF） and significantly decreased BV/TV, total density, immature bone density, and mature area within the callus on day-14 and -21 PF; they had significantly increased empty callus area at days 14 and 21 PF. Our results demonstrate that the heterozygous deletion of lrp6 impairs fracture healing, which suggests that lrp6 has a role in fracture healing.

Combination antiretroviral studies for patients with primary biliary cirrhosis

Following the characterization of a human betaretrovirus in patients with primary biliary cirrhosis(PBC),pilot studies using antiretroviral therapy have been conducted as proof of principal to establish a link of virus with disease and with the eventual aim to find better adjunct therapies for patients unresponsive to ursodeoxycholic *** the first open label pilot study,the reverse transcriptase inhibitor lamivudine had little demonstrable biochemical or histological effect after 1 ***,lamivudine in combination with zidovudine was associated with a significant reduction in alkaline phosphatase as well as improvement in necroinflammatory score,cholangitis and ductopenia over a 12 mo period.A double blind,multi-center randomized controlled trial using lamivudine with zidovudine for 6 mo confirmed a significant reduction in alkaline phosphatase,AlT and AST in patients on antiviral ***,none of the patients achieved the stringent endpoint criteria for normalization of alkaline ***,some patients developed biochemical rebound consistent with drug resistance.A major fault of these studies has been the inability to measure the viral load in peripheral blood and therefore,provide a direct correlation between improvement of hepatic biochemistry and reduction in viral ***,viral mutants to lamivudine with zidovudine were later characterized in the NOD.c3c4 mouse model of PBC that has been used to test other antiretroviral regimens to *** combination of tenofovir and emtricitabine reverse transcriptase inhibitors and the HIV protease inhibitor,lopinavir were found to abrogate cholangitis in the NOD.c3c4 mouse model and the same regimen normalized the liver tests in a PBC patient with HIV and human betaretrovirus *** combination antiretroviral therapy has now been used in a double blind randomized controlled crossover study for patients with PBC followed by an open label extension *** a thi
