An efficient and safe strategy for germ cell-specific automatic excision of foreign DNA in F1 hybrid transgenic silkworms

The safety of transgenic technology is a major obstacle in the popularization and use of transgenic silkworms and their *** sericulture,only the first filial generation(F1)hybrid eggs produced by cross-breeding Japanese and Chinese original strains are usually used for the large-scale breeding of silkworms,but this may result in uncontrolled transgene dispersal during the popularization and application of the F1 hybrid transgenic *** address this issue,we developed a safe and efficient strategy using the GAL4/Upstream activating sequence(UAS)system,the FLP/flippase recognition target(FRT)system,and the gonad-specific expression gene promoters(RSHP1p and Nanosp)for the germ cell-specific automatic excision of foreign DNA in the F1 hybrid transgenic *** established 2 types of activator strains,R1p::GAL4-Gr and Nsp::GAL4-Gr,containing the testis-specific GAL4 gene expression cassettes driven by RSHP1p or Nanosp,respectively,and 1 type of effector strain,UAS::FLP-Rg,containing the UAS-linked FLP gene expression *** FLP recombinase-mediated sperm-specific complete excision of FRT-flanked target DNA in the F1 double-transgenic silkworms resulting from the hybridization of R1p::GAL4-Gr and UAS::FLP-Rg was 100%,whereas the complete excision efficiency resulting from the hybridization of Nsp::GAL4-Gr and UAS::FLP-Rg ranged from 13.73%to 80.3%.Additionally,we identified a gene,sw11114,that is expressed in both testis and ovary of Bombyx mori,and can be used to establish novel gonad-specific expression systems in transgenic *** strategy has the potential to fundamentally solve the safety issue in the production of F1 transgenic silkworm eggs and provides an important reference for the safety of transgenic technology in other insect species.

Atomically precise Ru-O-Ru clusters for enhanced water dissociation in alkaline hydrogen evolution

Atomic clusters typically exhibit distinctive electronic structures and physicochemical ***,as the size decreases,their ability to adsorb and dissociate water also diminishes,thereby affecting chemical reactions involving water *** the adsorption and dissociation capabilities of atomic clusters towards water molecules and elucidating the mechanisms underlying their performance enhancement have become important research ***,employing the carrier-anchored strategy,Ru-O-Ru atomic clusters were prepared and displayed excellent activity and durability in the hydrogen evolution ***,the Ru-O-Ru atomic clusters exhibited only 86 mV overpotential at 100 mA·cm−^(2) and superior membrane-electrode-assembly activity than commercial Ru/C *** radiation-based Fourier transform infrared spectroscopic measurements revealed that the modification of oxygen in Ru-O-Ru units promoted the reorientation of water molecules from a H-up orientation to H-down,therefore,enhanced the formation of strong hydrogen-bond network of interfacial water on the surface of Ru-O-Ru clusters,leading to enhanced adsorption and dissociation of water and accelerated Volmer *** findings provide a potential strategy and deep insights for the development of atomic clusters in electrocatalysts.

Protective Effects of Danmu Extract Syrup on Acute Lung Injury Induced by Lipopolysaccharide in Mice through Endothelial Barrier Repair

Objective:To investigate the effects of Danmu Extract Syrup(DMS) on lipopolysaccharide(LPS)-induced acute lung injury(ALI) in mice and explore the ***:Seventy-two male Balb/C mice were randomly divided into 6 groups according to a random number table(n=12),including control(normal saline),LPS(5 mg/kg),LPS+DMS 2.5 mL/kg,LPS+DMS 5 mL/kg,LPS+DMS 10 mL/kg,and LPS+Dexamethasone(DXM,5 mg/kg) *** pretreatment with DMS and DXM,the ALI mice model was induced by LPS,and the bronchoalveolar lavage fluid(BALF) were collected to determine protein concentration,cell counts and inflammatory *** lung tissues of mice were stained with hematoxylin-eosin,and the wet/dry weight ratio(W/D) of lung tissue was *** levels of tumor necrosis factor-α(TNF-α),interleukin(IL)-6 and IL-1βin BALF of mice were detected by enzyme linked immunosorbent *** expression levels of Claudin-5,vascular endothelial(VE)-cadherin,vascular endothelial growth factor(VEGF),phospho-protein kinase B(p-Akt) and Akt were detected by Western blot ***:DMS pre-treatment significantly ameliorated lung histopathological *** with the LPS group,the W/D ratio and protein contents in BALF were obviously reduced after DMS pretreatment(P<0.05 or P<0.01).The number of cells in BALF and myeloperoxidase(MPO) activity decreased significantly after DMS pretreatment(P<0.05 or P<0.01).DMS pre-treatment decreased the levels of TNF-α,IL-6 and IL-1β(P<0.01).Meanwhile,DMS activated the phosphoinositide 3-kinase/protein kinase B(PI3K/Akt) pathway and reversed the expressions of Claudin-5,VE-cadherin and VEGF(P<0.01).Conclusions:DMS attenuated LPS-induced ALI in mice through repairing endothelial *** might be a potential therapeutic drug for LPS-induced lung injury.

Quantum dots have been widely applied in biosensing due to their outstanding optical *** emissions of quantum dots are mainly determined by their composition and size,as described by the Brus’s ***,in this case,their emissions are hardly regulated reversibly and responsively,which are unsuitable for biosensing and *** the last decade,capping ligands have been used for designing biosensors because of their responsive regulation on the photoluminescence of quantum ***,we first summarize the advances in characterization and calculation specific for ligands,which have helped to provide insights into the photoluminescence process and energy band theory of quantum *** then review two ways of ligand design that influence the optical properties of quantum dots:affecting the process of photoluminescence,or the orbital/electronic *** the latter case,the atoms on both the ligand and the surface of the quantum dot interact to affect the energy band structure of the quantum dot *** are presented of how these quantum dots that possess responsive properties due to the design of the ligands have been applied to *** further exploration,we hope to see advances in the fundamental understanding of the energy band structures and practical applications of these quantum dots.

A dynamic soil freezing characteristic curve model for frozen soil

The soil freezing characteristic curve(SFCC)plays a fundamental role in comprehending thermohydraulic behavior and numerical simulation of frozen *** study proposes a dynamic model to uniformly express SFCCs amidst varying total water contents throughout the freezing-thawing ***,a general model is proposed,wherein the unfrozen water content at arbitrary temperature is determined as the lesser of the current total water content and the reference value derived from saturated *** dynamic performance of this model is verified through test ***,in accordance with electric double layer(EDL)theory,the theoretical residual and minimum temperatures in SFCC are calculated to be-14.5℃to-20℃for clay particles and-260℃,*** ensure that the SFCC curve ends at minimum temperature,a correction function is introduced into the general ***,a simplified dynamic model is proposed and investigated,necessitating only three parameters inherited from the general ***,both general and simplified models are evaluated based on a test database and proven to fit the test data exactly across the entire temperature *** recommended parameter values for various types of soils are ***,this study provides not only a theoretical basis for most empirical equations but also proposes a new and more general equation to describe the SFCC.

Comparative analysis of the microbiome of sympatric wintering Bean Geese,Domestic Ducks,humans,and soil at Shengjin Lake of China reveals potential public risk to human health

The gut microbiota of migratory waterbirds is affected by various complex factors,including cross-species transmission,which increases the risk of pathogen spreading among sympatric animals and poses a potential public health risk to *** this study,we investigated the microbial communities of wintering Bean Geese(Anser fabalis),Domestic Ducks(*** domesticus),humans,and soil using high-throughput sequencing of the 16S rRNA gene region in Shengjin Lake,*** total,6,046,677 clean reads were obtained,representing 41,119 operational taxonomic units(OTUs)across the four *** dominant microbial phyla were the Proteobacteria,Firmicutes,Bacteroidota,and *** Sorensen similarity index and alpha and beta diversity results showed that the gut microbial communities of Bean Geese and Domestic Ducks were more similar to those of the other *** analysis revealed that Faecalibacterium prausnitzii,Pseudomonas fragi,and Bradyrhizobium elkanii were hubs of the three major *** common microbiomes were iden-tified in Bean Geese,Domestic Ducks,humans,and soil in Shengjin Lake.A total of 96 potential pathogens were identified among the four groups,with 20 specific potentially pathogenic microbiomes found in the gut of Bean *** of these pathogens are responsible for significant financial losses in the poultry industry and pose risks to human *** pneumoniae,Morganella morganii,Escherichia coli,and Ralstonia insidiosa are potential core pathogens found in the four groups at Shengjin Lake that can cause diseases in humans and an-imals and facilitate cross-species transmission through various ***,humans are at risk of con-tracting these pathogens from migratory birds because of their frequent contact with domestic ***,further studies are required to explore the potential pathogenic species and transmission pathways among sympatric wintering Bean Geese,Domestic Ducks,humans,and soil.

Soil bacterial and fungal communities resilience to long-term nitrogen addition in subtropical forests in China

Atmospheric nitrogen(N)deposition is predicted to increase,especially in the ***,the responses of soil microorganisms to long-term N addition at the molecular level in N-rich subtropical forests have not been clarified.A long-term nutrient addition experiment was conducted in a subtropical evergreen old-growth forest in *** four treatments were:control,low N(50 kg N ha^(-1)a^(-1)),high N(100 kg N ha^(-1)a^(-1)),and combined N and phosphorus(P)(100 kg N ha^(-1)a^(-1)+50 kg P ha^(-1)a^(-1)).Metagenomic sequencing characterized diversity and composition of soil microbial communities and used to construct bacterial/fungal co-occurrence ***-treated soils were more acidic and had higher levels of dissolved organic carbon than *** were no significant differences in microbial diversity and community composition across *** addition of nutrients increased the abundance of copiotrophic bacteria and potentially beneficial microorganisms(e.g.,Gemmatimonadetes,Chaetomium,and Aureobasidium).Low N addition increased microbiome network *** rare fungi were identified as module hubs under nutrient addition,indicating that low abundance fungi were more sensitive to increased *** results indicate that the overall composition of microbial communities was stable but not static to long-term N *** findings provide new insights that can aid predictions of the response of soil microbial communities to long-term N addition.

Mannanase improves the growth performance of broilers by alleviating inflammation of the intestinal epithelium and improving intestinal microbiota

This experiment aimed to discuss and reveal the effect and mechanism of mannanase on intestinal inflammation in broilers triggered by a soybean meal diet. In this experiment, 384 Arbor Acres broilers at 1 d old were randomly divided into 3 treatment groups. The broilers were fed a corn-soybean meal basal diet, a low-energy diet (metabolizable energy reduced by 50 kcal/kg), and a low-energy diet supplemented with 100 mg/kg mannanase for 42 d. The low-energy diet increased feed conversion ratio from 0 to 42 d, reduced ileal villus height and villus height-to-crypt depth ratio and upregulated the expression of nuclear factor kappa B ( NF-κB ) in the ileum ( P P P CLDN1 ) and zonula occludens-1 ( ZO-1 ) in the jejunum at 21 d, downregulated the expression of ilealNF-κB , and increased the content of isobutyric acid in the cecum of broilers ( P Lactobacillus reuteri in the ileum of broilers. The addition of mannanase increased the relative abundance ofLactobacillus-KC45b andLactobacillus johnsonii in broilers. furthermore, a low-energy diet reduced the relative abundance ofButyricicoccus in the intestine of broilers and inhibited oxidative phosphorylation and phosphoinositol metabolism. Mannanase increased the relative abundance ofOdoribacter , promoted energy metabolism and N-glycan biosynthesis, and increased the activities of GH3 and GH18. It is concluded that mannanase could improve the growth performance of broilers by reducing the expression ofNF-κB in the ileum, increasing the production of SCFA in the cecum, suppressing intestinal inflammation, balancing the intestinal microbiota, reducing damage to the intestinal barrier, and improving the efficiency of nutrient utilization to alleviate the adverse effects caused by the decrease in dietary energy *** The PDF
