Inhibitory effect of three biopolymer materials on scar formation following trabeculectomy

AIM: To investigate the inhibitory effects of amniotic membrane, polylactic acid membrane and chitosan membrane on scar formation following trabeculectomy. METHODS: A total of 24 New Zealand white rabbits (48 eyes) were randomly divided into 4 groups: amniotic membrane group, polylactic acid membrane group, chitosan membrane group, and control group, with 6 rabbits (12 eyes) in each group. The left eyes underwent routine trabeculectomy, and the right eyes were considered as controls. Amniotic membrane, polylactic acid membrane and chitosan membrane were respectively installed under sclera flap in three groups, but any treatment was not applied in control group. Intraocular pressure, conjunctival filtering bleb, and anterior chamber inflammation responses were monitored at day 1, 3, 7, 14, 28 and 56 post-operatively. Eyeball tissue underwent histopathological examination at day 56 post-operatively. RESULTS: Fibrocytes and inflammatory cells were reduced in amniotic membrane, polylactic acid membrane and chitosan membrane groups compared to that in control group. At day 1 post-operatively, intraocular pressure was decreased in three membrane groups compared to that in control group. At day 14 post-operatively, the intraocular pressure was decreased significantly, while it of three membrane groups was significantly lower than that of preoperative (P0.05). Filtering bleb of four groups was clearly observed at day 7 post-operatively, but there was no significant difference in pair-wise comparison. At day 28 and 56 post-operatively, filtering bleb in control group was significantly narrowed compared to that in three membrane groups (P <0.05), but there was no significant difference in pair-wise comparison of three membrane groups. CONCLUSION: All amniotic membrane, polylactic acid membrane and chitosan membrane can effectively inhibit scar formation following trabeculectomy, the effect of amnioti

Impact of Arsenic Trioxide on LS-174T Cell Growth in vitro and the Activity of Telomerase

The therapeutic action of arsenic trioxide（ As203 ） on solid tumors has aroused widespread interest among scholars. To study the impact of As2O3 on human colorectal carcinoma cells（ LS-174T cell） and the activity of telomerase, the methods of PCR-ELISA, flow cytometry （FCM） and MTT assay in vitro were utilized. The results show that （1） with an increase in the concentration of As2O3, the ratio of living the cells to dead cells decreases significantly, and the IC50 value is 5.23 μg/mL; （2） the cells of the experimental groups can endure a series of morphological changes similar to the features of apoptosis ; （3） the apoptotic curves of FCM pictures appear after 24 h, and the cells show the apoptosis in a time-dependent manner; （4） As2O3 can inhibit the activity of telomerase of the cell extraction obviously in a concentration-dependent and time-dependent manner after 24 h. It can be concluded from the experiment results in vitro that As2O3 can induce the apoptosis of LS-174T cells and inhibit the telomerase activity. Therefore, it has been proposed, for the first time, that these two factors （the apoptosis of LS-174T cells and the inhibition to the telomerase activity） are important causes of the LS-174T cell death caused by As2O3.

o understand the driving forces leading to mixed-ligand complexes in biologi-cal *** complexes of M (ATP(L￣(2-) type, where M = Cu￣(2+), Ni￣(2+) orCo￣(2+) , and L= pyridine(Py) , 4-picoline, 3, 5-lutidine, isoquinoline (lq) or benzimi-dazole(Bi) in aqueous solution were studied by spectropliotometry, some of them(M=Cu￣(2+), L = isoquinoline or benzimidazole) were also separately studied hy po-tentiometric pH titration. The results show that an intramolecular stacking interac-tion exists between the heteroaromatic ring of the ligands and the purine moiety ofATP.

Effects of cloned tumstatin-related and angiogenesis-inhibitory peptides on proliferation and apoptosis of endothelial cells

Background Tumstatin is a recently developed endogenous vascular endothelial growth inhibitor that can be applied as an anti-angiogenesis and antineoplastic agent. The study aimed to design and synthesize the small molecular angiogenesis inhibition-related peptide （peptide 21）, to replicate the structural and functional features of the active zone of angiogenesis inhibition using tumstatin and to prove that synthesized peptide 21 has a similar activity： specifically inhibiting tumor angiogenesis like tumstatin. Methods Peptide 21 was designed and synthesized using biological engineering technology. To determine its biological action, the human umbilical vein endothelial cell line ECV304, the human ovarian cancer cell line SKOV-3 and the mouse embryo-derived NIH3T3 fibroblasts were used in in vitro experiments to determine the effect of peptide 21 on proliferation of the three cell lines using the MTT test and growth curves. Transmission electron microscopy （TEM） and flow cytometry （FCM） were applied to analyze the peptide 21-induced apoptosis of the three cell lines qualitatively and quantitatively. In animal experiments, tumor models in nude mice subcutaneously grafted with SKOV-3 were used to observe the effects of peptide 21 on tumor weight, size and microvessel density （MVD）. To initially investigate the role of peptide 21, the effect of peptide 21 on the expression of vascular endothelial growth factors （VEGFs） by tumor tissue was semi-quantitatively analyzed. Results The in vitro Ml-r test and growth curves all indicated that cloned peptide 21 could specifically inhibit ECV304 proliferation in a dose-dependent manner （P 〈0.01）; TEM and FCM showed that peptide 21 could specifically induce ECV304 apoptosis （P 〈0.01）. Results of in vivo experiments showed that tumors in the peptide 21 group grew more slowly. The weight and size of the tumors after 21 days of treatment were smaller than those in the control group （P 〈0.05）, with a mean tumor inhibition rate of 67.

Response relationship between tourism disturbance and community resilience in agricultural heritage site:a case study of Honghe Hani Rice Terraced System

Agricultural heritage systems have the characteristics of ecological fragility and cultural *** a long time,the conservation and sustainable development of agricultural heritage systems have received attention from all ***,there are few measurements of tourism disturbance(TD)and community resilience(CR)in agricultural heritage sites,and research on the relationship between the two is even more *** study selects six villages in the Honghe Hani Rice Terraced System(HHRTS)as examples and constructs an evaluation system for TD and *** questionnaires and in-depth interviews,it explores the response relationship between TD and CR of heritage site,and compares the response relationship of villages in different stages of tourism *** results indicate that the heritage site's current response relationship between TD and CR shows a positive response type,and the development of tourism has brought positive responses and *** are differences in the interactive relationship between TD and CR among communities with different levels of tourism *** communities with poor tourism development,CR is more susceptible to the impact of TD than other *** stability of the relationship between TD and CR response gradually tends to stabilize from poor,good,and moderate tourism development.

Unsupervised multiplex graph diffusion networks with multi-level canonical correlation analysis for multiplex graph representation learning

In recent years, unsupervised multiplex graph representation learning(UMGRL) has received increasing research interest, which aims to learn discriminative node features from the multiplex graphs supervised by data without the guidance of labels. Although these designed UMGRL methods have obtained great success in various graph-related tasks, most existing UMGRL models still have the following issues: highly depending on complex self-supervised strategies(i.e., data augmentation,pretext tasks, and negative pairs sampling), restricted receptive fields, and only aggregating low-frequency information between nodes. In this paper, we propose a simple unsupervised multiplex graph diffusion network(UMGDN) with the aid of multi-level canonical correlation analysis to solve the above issues. Specifically, we first decouple the feature transform and propagation processes of the graph convolution layer to further improve the generalization of the learnable parameters. And then, we propose adaptive diffusion propagation to capture long-range dependency relationships between nodes, not the local neighborhood interactions. Finally, a multi-level canonical correlation analysis loss on both the feature transform and propagation processes is proposed to maximize the correlation of the same node features from multiple graphs for guiding model optimization. Compared to the existing UMGRL models, our proposed UMGDN does not need to introduce any data augmentation, negative pairs sampling techniques, complex pretext tasks, and also adaptively aggregates the optimal frequency information between nodes to generate more robust node embeddings. Extensive experiments on four popular datasets and two graph-related tasks demonstrate the effectiveness of the proposed method.

Bioenergy recovery and carbon emissions benefits of short-term bio-thermophilic pretreatment on low organic sewage sludge anaerobic digestion:A pilot-scale study

Sewage sludge in cities of Yangzi River Belt,China,generally exhibits a lower organic content and higher silt contentdue to leakage of drainage system,which caused low bioenergy recovery and carbon emission benefits in conventional anaerobic digestion(CAD).Therefore,this paper is on a pilot scale,a bio-thermophilic pretreatment anaerobic digestion(BTPAD)for low organic sludge(volatile solids(VS)of 4%)was operated with a long-term continuous flow of 200 *** VS degradation rate and CH_(4) yield of BTPAD increased by 19.93%and 53.33%,respectively,compared to those of *** analysis of organic compositions in sludge revealed that BTPAD mainly improved the hydrolysis of proteins in *** analysis of microbial community proportions by high-throughput sequencing revealed that the short-term bio-thermophilic pretreatment was enriched in Clostridiales,Coprothermobacter and Gelria,was capable of hydrolyzing acidified proteins,and provided more volatile fatty acid(VFA)for the subsequent *** combined with fluorescence quantitative polymerase chain reaction(PCR)analysis showed that the number of bacteria with high methanogenic capacity in BTPAD was much higher than that in CAD during the medium temperature digestion stage,indicating that short-term bio-thermophilic pretreatment could provide better methanogenic conditions for ***,the greenhouse gas emission footprint analysis showed that short-term bio-thermophilic pretreatment could reduce the carbon emission of sludge anaerobic digestion system by 19.18%.

Background The autosomal dominant form of retinitis pigmentosa （ADRP） can be caused by mutations in 14 genes and further loci remains to be identified. This study was intended to identify mutations in a Chinese pedigree with ADRP. Methods A large Chinese family with retinitis pigmentosa was collected. The genetic analysis of the family suggested an autosomal dominant pattern. Microsatellite （STR） markers tightly linked to genes known to be responsible for ADRP were selected for linkage analysis. Exons along with adjacent splice junctions of PRPF31 were amplified by polymerase chain reaction （PCR） and screened by direct sequencing. Results The caused gene of ADRP was mapped to 19q13.4 between markers D19S572 and D19S877, with a maximum LOD score of 3.01 at marker D19S418 （recombination fraction=0）. Conclusion The affected gene linked to the 19q13.4 in a Chinese family with ADRP, which is different from other mutations at the same loci in other Chinese families.

目的探讨p53基因过表达介导不同转移潜能肿瘤细胞的凋亡分子机理,寻找与细胞凋亡和肿瘤转移的相关基因。方法应用mRNA差异显示方法分析腺病毒介导的野生型p53基因感染不同转移潜能的肺癌细胞系前后存在的表达差异基因,并用逆转录-聚合酶链反应、Northern印迹和Western印迹方法加以验证。结果分析发现,在p53基因诱导后ANNEXIN A2基因的表达显著差异有统计学意义,经p53基因诱导后ANNEXINA2基因的表达有明显下调。并且以Anip973细胞的表达缺失最为显著。经逆转录-聚合酶链反应、Northern印迹和Western印迹方法也验证了这种差异的存在。结论表明ANNEXINA2基因的表达与p53基因诱导的肿瘤细胞凋亡存在密切的相关性。
