The vaginal microbiome likely influences host signaling compounds within the reproductive tract,including pro-inflammatory signals,which may play an important role during *** lactobacilli are associated with positive pregnancy outcome,whereas bacterial vaginosis,a dysbiosis of the vaginal microbiome,is associated with an increased risk of adverse pregnancy outcomes including preterm *** the host response could be predicted based on the taxonomic composition of the vaginal microbiome,particularly early in pregnancy,then those predictions could potentially be used to personalize intervention methods to reduce preterm birth and other adverse *** this proof of principle study,we apply multivariate strategies to analyze 16s rRNA-based taxonomic surveys in conjunction with targeted immuno-proteomic and lipidomic data from vaginal samples from 58 women enrolled in the Multi-Omic Microbiome study-Pregnancy Initiative during early *** between the vaginal microbiome and the vaginal lipidome have not been previously *** from this study reveal significant multiple pairwise associations between microbial taxa,specific eicosanoids and sphingomyelins,and *** the biologic significance of these associations is not yet known,these results support the utility of such multi-omic approaches as a means to predict the impact of the microbiome on the host.

The gut hormone apelin is a major therapeutic focus for several diseases involving inflammation and aberrant cell growth. We investigated whether apelin-36 contained alternative bioactive peptides associated with normal physiology or disease. Amino acid sequence analysis of apelin-36 identified an amidation motif consistent with the formation of a secondary bioactive peptide (sCNH2). sCNH2 is proven to be mitogenic and chemotactic in normal/malignant cells and augments angiogenesis via a PTX-resistant/CT-X-sensitive G protein-coupled receptor (GPCR). Notably, sCNH2 is substantially more potent and sensitive than apelin-13 and vascular endothelial growth factor-A. Endogenous sCNH2 is highly expressed in human tumors and placenta and in mouse embryonic tissues. Our findings demonstrate that sCNH2 is a new apelinergic member with critical pluripotent roles in angiogenesis related diseases and embryogenesis via a non-APJ GPCR.

Cancer of unknown primary(CUP)is a recalcitrant disease with poor prognosis because it lacks standard first-line *** consists of diverse malignancy groups,making personalized precision therapy *** present study aimed to identify an effective therapy for a CUP patient using a patient-derived orthotopic xenograft(PDOX)*** paper reports the usefulness of the PDOX model to precisely identify effective and ineffective chemotherapy and to compare the efficacy of *** A1-R with first-line chemotherapy using the CUP PDOX *** present study is the first to use a CUP PDOX model,which was able to precisely distinguish the chemotherapeutic *** found that a carboplatinum(CAR)-based regimen was effective for this CUP *** also demonstrated that *** A1-R was more effective against the CUP tumor than first-line *** results indicate that *** A1-R has clinical potential for CUP,a resistant disease that requires effective therapy.
