ObjectiveTo investigate the procedural outcomes and the long-term survival of patients undergoing transcatheter aortic valve implantation （TAVI） and compare study results of patients≤ 80 years and patients ＆gt; 80 years *** total of 240 patients treated with TAVI were divided into two groups according to age≤ 80 years （n = 105; 43.8%） and ＆gt; 80 years （n= 135; 56.2%）. The baseline characteristics and the pro-cedural outcomes were compared between these two groups of *** the exception of peripheral artery disease and hypercho-lesterolemia, which were more frequently observed in the older age group, baseline characteristics were comparable between groups. Complica-tion rates did not differ significantly between patients≤ 80 years and patients ＆gt; 80 years. There were no differences in 30-day mortality rates be-tween patients aged≤ 80 years and patients ＆gt; 80 years old （9.5%vs. 7.4%, respectively;P = 0.557）. After a median follow-up of 28 months （in-terquartile range： 16-42 months）, 50 （47.6%） patients aged≤ 80 years died compared to 57 （42%） deaths in the group of patients ＆gt; 80 years old （P= 0.404）.ConclusionThe results of the present single center study showed that age did not significantly impact the outcomes of TAVI.

BACKGROUND Early diagnosis is key to prevent bowel damage in inflammatory bowel disease(IBD).Risk factor analyses linked with delayed diagnosis in European IBD patients are scarce and no data in German IBD patients *** To identify risk factors leading to prolonged diagnostic time in a German IBD *** Between 2012 and 2022,430 IBD patients from four Berlin hospitals were enrolled in a prospective study and asked to complete a 16-item questionnaire to determine features of the path leading to IBD *** diagnostic time was defined as the time from symptom onset to consulting a physician(patient waiting time)and from first consultation to IBD diagnosis(physician diagnostic time).Univariate and multivariate analyses were performed to identify risk factors for each time *** The total diagnostic time was significantly longer in Crohn’s disease(CD)compared to ulcerative colitis(UC)patients(12.0 vs 4.0 mo;P<0.001),mainly due to increased physician diagnostic time(5.5 vs 1.0 mo;P<0.001).In a multivariate analysis,the predominant symptoms diarrhea(P=0.012)and skin lesions(P=0.028)as well as performed gastroscopy(P=0.042)were associated with longer physician diagnostic time in CD *** UC,fever was correlated(P=0.020)with shorter physician diagnostic time,while fatigue(P=0.011)and positive family history(P=0.046)were correlated with longer physician diagnostic *** We demonstrated that CD patients compared to UC are at risk of long diagnostic *** efforts should focus on shortening the diagnostic delay for a better outcome in these patients.

AIM： To review of the efficacy and safety outcomes of different single incision slings （SIS） systems, also in comparison with traditional ***： A literature search was conducted in PubMed/MEDLINE database. The research was re-stricted to randomized and/or prospective trials and retrospective studies, published after 2006, with at least 20 patients with non-neurogenic stress urinary incontinence （SUI）. The studies had to assess effcacy and/or safety of the SIS with a minimum follow-up of 12 mo. All the paper assessing the performance of tension free vaginal tape secur were excluded from this review. The fnal selection included 19 papers fulflling the aforementioned criteria. Two authors independently reviewed the selected ***： Four different SIS systems were analysed： Ajust , Ophira , Altis and MiniArc . The average objective cure rate was 88%. Overall no statistically significant differences were found between SIS and traditional mid-urethral slings （MUS） in terms of objective cure （all P 〉 0.005）. Only one paper showed a statistically lower success rate in MiniArc？ vs Advant-age？ slings （40% vs 90%） and higher rates of failure in the SIS group. Since there was a great variability in terms of tests performed, it was not possible to com-pare subjective cure between studies. The vast part of the studies showed no major complications after SIS surgery. We also observed very low reported pain rates in SIS patients. The RCTs on Ajust？ and MiniArc？, showed better outcomes in terms of post-operative pain compared to MUS. None of the patients reported long- term pain complains. CONCLUSION： SIS showed similar effcacy to that of traditional slings but lower short-term pain, complication and failure rates.

In allogeneic stem cell transplantation （SCT）, natural killer （NK） cells lacking their cognate inhibitory ligand can induce graft-versus-leukemia responses, without the induction of severe graft-versus-host disease （GVHD）. This feature can be exploited for cellular immunotherapy. In this study, we examined selective expansion of NK cell subsets expressing distinct killer immunoglobulin-like receptors （KIRs） within the whole human peripheral blood NK cell population, in the presence of HLA-Cw3 （C1） or Cw4 （C2） transfected K562 stimulator cells. Coculture of KIR＋ NK cells with C1 or C2 positive K562 cells, in the presence of IL-2＋ IL-15, triggered the outgrowth of NK cells that missed their cognate ligand. This resulted in an increased frequency of alloreactive KIR＋ NK cells within the whole NK cell population. Also, after preculture with K562 cells lacking their cognate ligand, we observed that this alloreactive NK population revealed higher numbers of CD107~ cells when cocultured with the relevant K562 HLA-C transfected target cells, as compared to coculture with untransfected K562 cells. This enhanced reactivity was confirmed using primary leukemic cells as target. This study demonstrates that HLA class I expression can mediate the skewing of the NK cell repertoire and enrich the population for cells with enhanced alloreactivity towards leukemic target cells. This feature may support future clinical applications of NK cell-based immunotherapy.

A study was conducted to evaluate the current production capabilities of the Mark One? 3D printer in printing carbon fibre reinforced thermoplastic (CFRTP) tensile test specimens according to the JIS K 7073 by making use of fused deposition modelling. Several different types of CFRTP tensile test specimens are printed and are tensile tested in the longitudinal direction to obtain an overview of the mechanical properties of 3D printed CFRTP material. These properties are compared with the literature values known for composite materials to see if these agree. The main goal of this research is to increase the knowledge of the 3D printing process of CFRTP and to later use this knowledge to further improve the 3D printing process to obtain stronger 3D printed CFRTP materials.

Hydrogen peroxide （H2O2） operates as a signaling molecule in eukaryotes, but the specificity of its signal- ing capacities remains largely unrevealed. Here, we analyzed whether a moderate production of H2O2 from two different plant cellular compartments has divergent effects on the plant transcriptome. Arabidopsis thaliana overexpressing glycolate oxidase in the chloroplast （Fahnenstich et al., 2008; Balazadeh et al., 2012） and plants deficient in peroxisomal catalase （Queval et al., 2007; Inze et al., 2012） were grown under non-photorespiratory conditions and then transferred to photorespiratory conditions to foster the production of H202 in both organelles. We show that H202 originating in a specific organelle induces two types of responses： one that integrates signals independently from the subcellular site of H202 production and another that is dependent on the H2O2 production site. H2O2 produced in peroxisomes induces transcripts involved in protein repair responses, while H2O2 produced in chloroplasts induces early signaling responses, including transcription factors and biosynthetic genes involved in production of secondary signaling messengers. There is a significant bias towards the induction of genes involved in responses to wounding and pathogen attack by chloroplas- tic-produced H202, including indolic glucosinolates-, camalexin-, and stigmasterol-biosynthetic genes. These transcriptional responses were accompanied by the accumulation of 4-methoxy-indol-3-ylmethyl glucosinolate and stigmasterol.

Iatrogenic preterm premature rupture of fetal membranes(iPPROM)after fetal surgery remains a strong trigger for premature *** fetal membrane defects do not heal spontaneously and amniotic fluid leakage and chorioamniotic membrane separation may occur,we developed a biocompatible,fetoscopically-applicable collagen plug with shape memory to prevent *** plug expands directly upon employment and seals fetal membranes,hence preventing amniotic fluid leakage and potentially *** typeⅠcollagen plugs were given shape memory and crimped to fit through a fetoscopic cannula(Ø3 mm).Expansion of the plug was examined in phosphate buffered saline(PBS).Its sealing capacity was studied ex vivo using human fetal membranes,and in situ in a porcine bladder *** crimped plug with shape memory expanded and tripled in diameter within 1 min when placed into PBS,whereas a crimped plug without shape memory did *** both human fetal membranes and porcine bladder,the plug expanded in the defect,secured itself and sealed the defect without membrane *** conclusion,collagen plugs with shape memory are promising as medical device for rapid sealing of fetoscopic defects in fetal membranes at the endoscopic entry point.
