Mammalian herbivory in post-fire chaparral impacts herbaceous composition but not N and C cycling

Aims Classical theory predicts that herbivores impact herb assemblages and soil nitrogen(N)cycling through selective plant consumption and the deposition of N-rich waste,with effects dependent upon ecosystem N *** are predicted to accelerate N cycling when N availability is high and decelerate cycling when availability is ***,experimental tests of these theories in natural systems are limited and have yielded contradictory ***’s widespread chaparral shrublands provide a tractable system in which to test these *** are prone to periodic crown fire,which temporarily removes living shrub cover,deposits mineral N on soils and allows diverse herbaceous assemblages to dominate the landscape for 3–5 *** is also increasingly vulnerable to herbaceous invasion;mammalian herbivory may limit the establishment of non-native herbs in the shrub *** We implemented a 2-year herbivore-exclosure experiment(Hopland,CA)to assess the impact of mammalian herbivory during early post-fire chaparral succession,both on herbaceous plant assemblages and soil N and C *** predicted that,in high-N post-fire conditions,mammalian herbivory would not demonstrate a strong preference for N-fixing herbs,would accelerate N cycling and would reduce the abundance of non-native *** Findings Excluding mammalian herbivores increased herb standing biomass by 54%,but changed neither the relative abundance of N-fixing ***-N-fixing herbs nor.any measure of N or C *** impacts on nutrient cycling may not be significant over the 2-year time scale of the experiment and physical effects of herbivore activity could have counteracted the influence of plant litter and animal dung/urine *** herbivores concentrated their feeding on typical non-native herbs,slightly decreasing their relative abundance;however,mammalian herbivory was not sufficient to stem the invasion of chaparral by invasive herbs

Infliximab does not increase colonic cancer risk associated to murine chronic colitis

AIM To explore the influence of Infliximab(IFX) on cancer progression in a murine model of colonic cancer associated to chronic *** AOM/DSS model was induced in C57BL/6 mice. Mice were injected with IFX(5 mg/kg) during each DSS cycle while control mice received saline. Body weight, occult blood test and stool consistency were measured to calculate the disease activity index(DAI). Mice were sacrificed at week 10 and colons were analyzed macroscopically and microscopically for number of cancers and degree of inflammation. MTT assay was performed on CT26 to evaluate the potential IFX role on metabolic activity and proliferation. Cells were incubated with TNF-α or IFX or TNF-α plus IFX, and cell vitality was evaluated after 6, 24 and 48 h. The same setting was used after pre-incubation with TNF-α for 24 *** IFX significantly reduced DAI and body weight loss in mice compared with controls, preserving also colon length at sacrifice. Histological score was also reduced in treated mice. At macroscopic analysis, IFX treated mice showed a lower number of tumor lesions compared to controls. This was confirmed at microscopic analysis, although differences were not statistically significant. In vitro, IFX treated CT26 maintained similar proliferation ability at MTT test, both when exposed to IFX alone and when associated to TNF-α.CONCLUSION IFX did not increase colonic cancer risk in AOM-DSS model of cancer on chronic colitis nor.influence directly the proliferation of murine colon cancer epithelial cells.

Deposition of the H2A.Z histone variant by the SWR1 complex （SWRI-C） in regulatory regions of specific loci modulates transcription. Characterization of mutations in Arabidopsis thaliana homologs of yeast SWRI-C has revealed a role for H2A.Z exchange in a variety of developmental processes. Nevertheless, the exact composition of plant SWRI-C and how it is recruited to target genes remains to be established. Here we show that SWC4, the Arabidopsis homolog of yeast SANT domain protein Swc4/Eaf2, is a DNA-binding protein that interacts with SWR1-C subunits. We demonstrate that the swc4-1 knockout mutant is embryo- lethal, while SWC4 RNAi knockdown lines display pleiotropic phenotypic alterations in vegetative and repro- ductive traits, including acceleration of flowering time, indicating that SWC4 controls post-embryonic processes. Transcriptomic analyses and genome-wide profiling of H2A.Z indicate that SWC4 represses tran- scription of a number of genes, including the floral integrator FT and key transcription factors, mainly by modulating H2A.Z deposition. Interestingly, SWC4 silencing does not affect H2A.Z deposition at the FLC locus nor.expression of this gene, a master regulator of flowering previously shown to be controlled by SWR1-C. Importantly, we find that SWC4 recognizes specific AT-rich DNA elements in the chromatin regions of target genes and that SWC4 silencing impairs SWRI-C binding at FT. Collectively, our data suggest that SWC4 regulates plant growth and development by aiding SWR1-C recruitment and modulating H2A.Z deposition.
