A two-stage process with temperature-shift for enhanced anthocyanin production in strawberry cell suspension cultures

A two-stage process with temperature-shift has been developed to enhance the anthocyanin yield in suspension cultures of strawbeny cells. The effect of the temperature-shift interval and the shift-time point was investigated for the optimization of this strategy. In this process, strawbeny cells were grown at 30°C (the optimum temperature for cell gowth) for a certain period as the first stage, with the temperature shifted to a lower temperature for the second stage. In response to the temperature shift-down, anthocyanin synthesis was stimulated and a higher content could be achieved than that at both boundary temperatures but cell growth was suppressed. When the lower boundary temperature. was decreased, cell growth was lowered and a delayed, sustained maximum anthocyanin content was achieved. Anthocyanin synthesis was strongly influenced by the shift-time point but cell growth was not. Consequently, the maximum anthocyanin content of 2.7 mg.g-fresh cell-1 was obtained on day 9 by a temperature-shift from 309°C, after 3-d culture, to 15°C. The highest anthocyanin yield of 318 mg. L-1 on day 12 was achieved when the temperature was shifted from 30°C, after 5-d culture, to 20°C. For a global optimization of both the yield and productivity, the optimum anthocyanin yield and productivity of 272 mg.L-1 1 and 30.2 mg.L-1d1 on day 9 were achieved by a two-stage culture with a temperature-shift from 30°C after 3 d to 20°C.

Radical gastrectomy with an adequate lymphadenectomy is the main procedure which makes it possible to cure patients with resectable gastric cancer (GC). A number of randomized controlled trials and meta-analysis provide phase III evidence that laparoscopic gastrectomy is technically safe and that it yields better short-term outcomes than conventional open gastrectomy for early-stage GC. While laparoscopic gastrectomy has become standard therapy for early-stage GC, especially in Asian countries such as Japan and South Korea, the use of minimally invasive techniques is still controversial for the treatment of more advanced tumours, principally due to existing concerns about its oncological adequacy and capacity to carry out an adequately extended lymphadenectomy. Some intrinsic drawbacks of the conventional laparoscopic technique have prevented the worldwide spread of laparoscopic gastrectomy for cancer and, despite technological advances in recent year, it remains a technically challenging procedure. The introduction of robotic surgery over the last ten years has implied a notable mutation of certain minimally invasive procedures, making it possible to overcome some limitations of the traditional laparoscopic technique. Robot-assisted gastric resection with D2 lymph node dissection has been shown to be safe and feasible in prospective and retrospective studies. However, to date there are no high quality comparative studies investigating the advantages of a robotic approach to GC over traditional laparoscopic and open gastrectomy. On the basis of the literature review here presented, robot-assisted surgery seems to fulfill oncologic criteria for D2 dissection and has a comparable oncologic outcome to traditional laparoscopic and open procedure. NPV risk simulation of an open pit gold mine project under the O'Hara cost model by using Gas

This paper analyzes an open pit gold mine project based on the O'Hara cost model. Hypothetical data is proposed based on different authors that have studied open pit gold projects, and variations are proposed according to the probability distributions associated to key variables affecting the NPV, like production level, ore grade, price of ore, and others, so as to see what if, in a gold open pit mine project of 3000 metric tons per day of ore. Two case scenarios were analyzed to simulate the NPV, one where there is low certainty data available, and the other where the information available is of high certainty. Results based on genetic algorithm metaheuristic simulations, which combine basically Montecarlo simulations provided by the Palisade Risk software, the O'Hara cost model, net smelter return and financial analysis tools offered by Excel are reported, in order to determine to which variables of the project is more sensitive the NPV.

Simulation of a Shock Tube with a Small Exit Nozzle

Shock tubes are frequently used to rapidly heat up reaction mixtures to study chemical reaction mechanisms and kinetics in the field of combustion chemistry [1]. In the present work, the flow field inside a shock tube with a small nozzle in the end plate has been investigated to support the analysis of reacting chemical mixtures with an attached mass spectrometer and to clarify whether the usual assumptions for the flow field and the related ther- modynamics are fulfilled. In the present work, the details of the flow physics inside the tube and the flow out of the nozzle in the end plate have been investigated. Due to the large differences in the typical length scales and the large pressure ratios of this special device, a very strong numerical stiffness prevails during the simulation process. Second-order ROE numerical schemes have been employed to simulate the flow field inside the shock tube. The simulations were performed with the commercial code ANSYS Fluent [2]. Axial-symmetric boundary conditions are employed to reduce the consumption of CPU time. A density-based transient scheme has been used and vali- dated in terms of accuracy and efficiency. The simulation results for pressure and density are compared with ana- lytical solutions. Numerical results show that a density-based numerical scheme performs better when dealing with shock-tube problems [5]. The flow field near the nozzle is studied in detail, and the effects of the nozzle to pressure and temperature variations inside the tube are invcstigatcd. The results show that this special shock-tube setup can be used to study high-temperature gas-phase chemical reactions with reasonable accuracy.

The evolving profile and role of computer science

Technological innovation ushered in the computer era, and, after a few years of tutelage by established disciplines, computer science emerged as an independent discipline. In the subsequent decades computer science developed its special identity, sharing the dual character of engineering and mathematics. This evolution is revisited here based on my personal experience. In my view, the notion of computational model has been the enabler of extraordinary creativity, and at the same time the source of critical reflection two decades ago. However, capitalizing on a vibrant technology, computer science is reinventing itself as the indispensable enabler of applications. This is a crucial profile that calls for a pedagogical adaptation, where the notion of model morphs from means to end.

Shock Wave-Boundary Layer Interaction in Forced Shock Oscillations

The flow in transonic diffusers as well as in supersonic air intakes becomes often unsteady due to shock wave boundary layer interaction. The oscillations may be induced by natural separation unsteadiness or may be forced by boundary conditions. Significant improvement of CFD tools, increase of computer resources as well as development of experimental methods have *** the attention of researchers to this topic. To investigate the problem forced oscillations of transonic turbulent flow in asymmetric two-dimensional Laval nozzle were considered. A viscous, perfect gas flow, was numerically simulated using the Reynolds-averaged compressible Navier-Stokes solver SPARC, employing a two-equation, eddy viscosity, turbulence closure in the URANS *** time-dependent and stationary flow simulations, Mach numbers upstream of the shock between 1.2 and 1.4 were considered. Comparison of computed and experimental data for steady states generally gave acceptable agreement. In the case of forced oscillations, a harmonic pressure variation was prescribed at the exit plane resulting in shock wave motion. Excitation frequencies between 0 Hz and 1024 Hz were investigated at the same pressure *** main result of the work carried out is the relation between the amplitude of the shock wave motion and the excitation frequency in the investigated range. Increasing excitation frequency resulted in decreasing amplitude of the shock movement. At high frequencies a natural mode of shock oscillation (of small amplitude) was observed which is not sensitive to forced excitement.

Distribution of action movements （DAM）： a descriptor for human action recognition

Human action recognition from skeletal data is an important and active area of research in which the state of the art has not yet achieved near-perfect accuracy on many well- known datasets. In this paper, we introduce the Distribution of Action Movements Descriptor, a novel action descriptor based on the distribution of the directions of the motions of the joints between frames, over the set of all possible mo- tions in the dataset. The descriptor is computed as a normal- ized histogram over a set of representative directions of the joints, which are in turn obtained via clustering. While the descriptor is global in the sense that it represents the overall distribution of movement directions of an action, it is able to partially retain its temporal structure by applying a window- ing scheme.
