Unveiling plant defense arsenal: metabolic strategies in Brassica oleracea during black rot disease

Alterations in plant metabolism play a key role in the complex plant-pathogen ***,there is still a lack of knowledge about the connection between changes in primary and specialized metabolism and the plant defense against diseases that impact ***,we aim to study the metabolic reprograming in Brassica oleracea plants upon infection by Xanthomonas campestris ***(Xcc).To accomplish this,we utilized a combination of untargeted and targeted metabolomics,through UPLC-Q-TOF-MS/MS and 1H-NMR,in two crop lines differing in resistance thatwere evaluated at two-and four-week intervals following inoculation(T1 and T2,respectively).Besides,to depict the physiological status of the plant during infection,enzymatic activities related to the carbohydrate pathway and oxidative stress were *** results revealed different temporal dynamics in the responses of the susceptible *** crops *** *** line suppresses carbohydrate metabolism contributing to limit nutrient supplies to the bacterium and prioritizes the induction of defensive compounds such as indolic glucosinolates,salicylic acid,phenylpropanoids and phytoalexins precursors at early infection *** contrast,the susceptible line invests in carbohydratemetabolism,including enzymatic activities related to the hexoses turnover,and activates defense signaling related to reactive oxygen ***,each line triggers a different metabolic strategy that will affect how the plant overcomes the disease in terms of resistance and *** work provides first insights of a fine-tuned metabolic regulation during Xcc infection in *** that will contribute to develop new strategies for plant disease management.

Prognostic implication of the coronary microvascular dysfunction in patients with isolated left bundle branch block

The present study aims to determine the influence of microvascular dysfunction (MVD) in the prognosis of patients presenting isolated left bundle branch block (LBBB). Methods: We studied 30 patients (pts), 22 males, 8 females, mean age 57 ± 4 years, with isolated LBBB, with a mean follow up of 48 ± 6 months. The control group consisted of 20 healthy individuals, 12 males, mean age 52 ± 10 years. Both groups were screened for cardiovascular risk factors (RF);they also had an echocardiogram and Coronary CT Scan, ruling out both structural heart disease and obstructive lesions of the epicardial coronary arteries. A myocardial perfusion study was then performed, with two groups emerging according to these results: Group A, 8 pts (26%), with reversible perfusion defects, in which the diagnosis of MVD was suspected, and Group B, 22 pts (74%), with either normal perfusion or minor septal/apical reversible defects (related to LBBB). All Group A pts, and 9 of the Group B pts, underwent coronary arteriography, with intracoronary acetylcholine and nitroglycerine infusion, thus evaluating vasomotor response as endothelium dependent (acetylcholine) or endothelium independent (nitroglycerine). During follow up, we reviewed functional class, 12 lead ECG and echocardiogram on a yearly basis. Results: All Group A patients had an abnormal acetylcholine response;only three of them had abnormal response to nitroglycerine infusion, suggesting endothelium dependent MVD. Of those in Group B, only one patient had abnormal acetylcholine response. At the end of the follow up period, 3 pts (37%) in Group A, showed functional class decrease vs 5 pts (22%) of those in Group B. Ulcerative colitis is a global health problem,affecting millions of individuals *** an inflammatory condition localised in the large intestine,rectal delivery of immunosuppressive therapies such as tacrolimus is a promising strategy to maximise drug concentration at the site of action whilst minimising systemic side ***,for the first time,self-supporting 3D-printed tacrolimus suppositories were prepared without the aid of moulds using a pharmaceutical semi-solid extrusion(SSE)3D *** suppositories were printed vertically in three different sizes using combinations of two lipid pharmaceutical excipients(Gelucire 44/14 or Gelucire 48/16)and coconut *** both suppository formulations had the appropriate viscosity characteristics for printing,the Gel 44 formulation required less energy and force for extrusion compared to the Gel 48 *** Gel 44 disintegrated more rapidly but released tacrolimus more slowly than the Gel 48 *** the tacrolimus release profiles were significantly different,both suppository systems released more than 80% drug within 120 *** and XRD analysis was inconclusive in determining the solid-state properties of the drug in the *** summary,this article reports on the fabrication of 3D printed selfsupporting suppositories to deliver personalised doses of a narrow therapeutic index drug,with potential benefits for patients with ulcerative colitis.

Innovative circular path harvester for mechanical harvesting of irregular and large-canopy olive trees

The harvesting process of the olive tree is mainly performed by manual means,because traditional olive orchards(the main planting typology)are formed of irregular,large-canopy trees that are very difficult to harvest *** that reason,the cost of harvesting is very high,and it threatens the future of these plantations whose conversion to other more modern layouts is not always possible due to several *** introduction of a harvester may represent the technological change that is the key factor for improved *** main purpose of this work was to develop a harvester based on canopy shaker technology for work on irregular,large trees in a circular *** design of the harvester was based on a determination of tree geometry,together with tree *** tests were used to determine machine-tree interaction,and to evaluate the removal,catch frame and driven *** proposed innovation allowed the fully mechanical harvest of previously planted trees with a removal efficiency of over 84%,achieving an effective field capacity of 0.21 hm^(2)/*** the results so far have been promising,further improvements are advisable in machine and tree adaptation.

Tropical alpine ecosystems exhibit outstanding plant diversity and endemism while being particularly sensitive to the impacts of climate *** understanding spatiotemporal changes in plant species composition,richness and community structure along tropical alpine altitudinal gradients is of primary importance,both the functional and historical/biogeographic dimensions of vegetation diversity remain largely *** used Generalized Linear Models and multivariate analyses to assess changes in species,growth forms,and biogeographic groups richness and abundance,in response to habitat variables along an elevation gradient in seven summits(3800 to 4600 m asl)in the Venezuelan Andes,studied using the standardized approach of the GLORIA-Andes monitoring *** habitat variables assessed were soil temperature(-10 cm),soil organic matter,slope inclination,and substrate *** found 113 species,representing72 genera,32 families,13 growth forms,and seven biogeographic origins,that included 25%of endemic *** observed richer vegetation,both in terms of species and growth forms,in summits with higher soil temperatures and higher SOM content,as well as higher biogeographic origin richness with increasing soil *** presence of holarctic elements increased toward higher elevations,while the occurrence of austral antarctic elements increased toward lower *** results indicate that biogeographic and functional approaches to vegetation diversity capture well the effect of abiotic filtering on community structuring in these tropical alpine *** findings constitute an important baseline for monitoring vegetation dynamics linked to climate change in the Venezuelan Andes by highlighting the functional and historical perspective on vegetation analyses,in contrast with more traditional approaches,based only on taxonomic species diversity.

Xeroderma pigmentosum group D 751 polymorphism as a predictive factor in resected gastric cancer treated with chemo-radiotherapy

AIM: To evaluate the potential association of xeroderma pigmentosum group D (XPD) codon 751 variant with outcome after chemo-radiotherapy in patients with resected gastric cancer. METHODS: We used PCR-RFLP to evaluate the genetic XPD Lys751Gln polymorphisms in 44 patients with stage Ⅲ (48%) and Ⅳ (20%) gastric cancer treated with surgery following radiation therapy plus 5-fluorouracil/ leucovorin based chemotherapy. RESULTS: Statistical analysis showed that 75% (12 of 16) of relapse patients showed Lys/Lys genotype more frequently (P = 0.042). The Lys polymorphism was an independent predictor of high-risk relapse-free survival from Cox analysis (HR: 3.07, 95% CI: 1.07-8.78, P = 0.036) and Kaplan-Meir test (P = 0.027, log-rank test). CONCLUSION: XPD Lys751Gln polymorphism may be an important marker in the prediction of clinical outcome to chemo-radiotherapy in resected gastric cancer patients.

Evaluation of the Ex-PRESS~ P-50 implant under scleral flap in combined cataract and glaucoma surgery

AIM： To evaluate the efficacy and safety of glaucoma drainage device Ex-PRESS~ P-50 for combined cataract surgery and glaucoma.·METHODS： Patients having cataract and open angle glaucoma or patients with open advanced glaucoma which needed two or more antiglaucoma medications were included. Combined cataract surgery and glaucoma with Ex-PRESS~ P-50 model placed under scleral flap was performed.·RESULTS： Out of 40 eyes of 40 patients（55% male and45% female） completed the study during one-year follow-up. The mean of age was 76.6 ±11.02 y. The intraocular pressure（IOP） decreased significantly during the 12-month follow-up from 23.5 mm Hg to 16.8 mm Hg（Wilcoxon signed ranks test, P 〈0.001）. A 59.5% of patients did not need any topical treatment, 10.8% of them needed one active principle, 27% needed two active principles, and 2.7% of them needed three active principles for successful IOP control（〈21 mm Hg）.·CONCLUSION：Combinedsurgeryofphacoemulsification with Ex PRESS~ P-50 lowers IOP from the preoperative baseline and reduces significantly the number of antiglaucoma active principles for IOP control after the operation.

Nitric oxide and hydrogen sulfide share regulatory functions in higher plant events

Nitric oxide(NO)and hydrogen sulfide(HS)are two molecules that share signaling properties in plant and animal cells NO and H2S originate two farmilies of de rived mol ecules designated reactive nitrogen and sulfur species(RNS and RSS,respectively).These molecules are responsible for certain protein regulatory processes through posttranslational modifications(PTMs),being the most remarkable S nitrosation and persufidation,which afect the thiol group of cysteine *** and H2S can also exert regulatory functions due to their interaction through the iron present in proteins that contain heme groups or iron-sulfur dlusters,as reported mainly in animal ***,the available information in plant cells is still very limited thus *** higher plants,NO and H2S are involved in a myriad of physiological events from seed germination to fruit ripening,but also the mec hanism of response to biotic and abiotic stress *** vie wpoint manuscript highlights the functional regulatory parllelism of these two molecules which also interact with the metabolism of reactive oxygen species(ROS)in plant cells.
