AIM to test whether Nox1 plays a role in typhlitis induced by Salmonella enterica serovar typhimurium(S. tm) in a mouse *** Eight-week-old male wild-type(wt) and Nox1 knockout(KO) c57BL6/J(B6) mice were administered metronidazole water for 4 d to make them susceptible to S. tm infection by the oral route. the mice were given plain water and administered with 4 different doses of S. tm by oral gavage. the mice were followed for another 4 d. From the time of the metronidazole application, the mice were observed twice daily and weighed daily. the ileum, cecum and colon were removed for sampling at the fourth day post-inoculation. Portions of all three tissues were fixed for histology and placed in RNAlater for m RNA/c DNA preparation and quantitative real-time PcR. the contents of the cecum were recovered for estimation of S. tm *** we found Nox1-knockout(Nox1-KO) mice were not more sensitive to S. tm colonization and infection than wt B6 mice. this conclusion is based on the following observations:(1) S. tm-infection induced similar weight loss in Nox1-KO mice compared to wt mice;(2) the same S. tm cFU was recovered from the cecal content of Nox1-KO and wt mice regardless of the inoculation dose, except the lowest inoculation dose(2 × 106 cFU) for which the Nox1-KO had one-log lower cFU than wt mice;(3) there is no difference in cecal pathology between wt and Nox1-KO groups; and(4) there are no S. tm infection-induced changes in gene expression levels(IL-1b, tNF-α, and Duox2) between wt and Nox1-KO groups. the Alpi gene expression was more suppressed by S. tm treatment in wt than the Nox1-KO cecum. cONcLUSION Nox1 does not protect mice from S. tm colonization. Nox1-KO provides a very minor protective effect against S. tm infection. Using NOX1-specific inhibitors for colitis therapy should not increase risks in bacterial infection.

Invariant natural killer t(iNKt)cells are part of the family of unconventional t cells,and they recognize glycolipid antigens presented on the MHc class I like molecule cD1^([1]).In humans there are five cD1 molecules named cD1a to cD1e,whereas mice use only cD1d for lipid ***,iNKt cells can elicit both cytotoxicity similarly to conventional cD8 t cells and secrete cytokines that influence the adaptive immune response including providing direct t cell help to B cells^([2]).In the context of tumors,they can use both release of granzyme B and use Fas-mediated mechanisms for killing of tumor *** iNKt cells use a restricted tcR repertoire and cD1 molecules are non-heterogenous the risk of graft vs host disease is limited and thus these t cells have been an attractive option for off the shelf immunotherapy to treat cancer^([3]).

In this study, interval-parameter programming, two-stage stochastic progranaming （tSP）, and conditional value-at-risk （cVaR） were incorporated into a general optimization framework, leading to an interval-parameter cVaR-based two-stage programming （IctP） method. the IctP method had several advantages：（i） its objective function simultaneously took expected cost and risk cost into consideration, and also used discrete random variables and discrete intervals to reflect uncertain properties; （ii） it quantitatively evaluated the right tail of distributions of random variables which could better calculate the risk of violated environmental standards; （iii） it was useful for helping decision makers to analyze the trade-offs between cost and risk; and （iv） it was effective to penalize the second-stage costs, as well as to capture the notion of risk in stochastic programming. the developed model was applied to sulfur dioxide abatement in an air quality management system. the results indicated that the IctP method could be used for generating a series of air quality management schemes under different risk-aversion levels, for identifying desired air quality management strategies for decision makers, and for considering a proper balance between system economy and environmental quality.

Background Hypertension(HtN)involves genetic variability in the renin-angiotensin system and influences antihypertensive *** previously reported that angiotensinogen(AGt)messenger RNA(mRNA)is endogenously bound by miR-122-5p and rs699 A>G decreases reporter mRNA in the microRNA functional-assay *** AGt promoter variant rs5051 c>t is in linkage disequilibrium(LD)with rs699 A>G and increases AGt *** independent effect of these variants is understudied due to their LD therefore we aimed to test the hypothesis that increased AGt by rs5051 c>t counterbalances AGt decreased by rs699 A>G,and when these variants occur independently,it translates to HtN-related *** we used in silico,in vitro,in vivo,and retrospective models to test this *** In silico,rs699 A>G is predicted to increase miR-122-5p binding affinity by 3%.Mir-ecLIP results show rs699 is 40–45 nucleotides from the strongest microRNA-binding site in the AGt ***,rs699 A>G increases AGt mRNA in an AGt-plasmid-cDNA HepG2 expression ***-tissue Expression(GtEx)and UK Biobank analyses demonstrate liver AGt expression and HtN phenotypes are not different when rs699 A>G occurs independently from rs5051 c>***,GtEx and the in vitro experiments suggest rs699 A>G confers cell-type-specific effects on AGt mRNA abundance,and suggest paracrine renal renin-angiotensin-system perturbations could mediate the rs699 A>G associations with *** we found that rs5051 c>t and rs699 A>G significantly associate with systolic blood pressure in Black participants in the UK Biobank,demonstrating a fourfold larger effect than in white *** studies are warranted to determine if altered antihypertensive response in Black individuals might be due to rs5051 c>t or rs699 A>*** like this will help clinicians move beyond the use of race as a surrogate for genotype.

t-cell-based immunotherapy is gaining momentum in cancer treatment;however,our comprehension of the transcriptional regulation governing t cell antitumor activity remains *** objective of this study was to explore the function of interferon regulatory factor 4(IRF4)in antitumor cD8^(+)t cells using the tRAMP-c1 prostate cancer and B16F10 melanoma *** achieve this,we generated an Irf4^(GFP-DtR) mouse strain and discovered that cD8^(+)tumor-infiltrating lymphocytes(tILs)expressing high levels of *** exhibited a more differentiated PD-1high cell *** administering diphtheria toxin to tumor-bearing Irf4^(GFP-DtR) mice,we partially depleted ***^(+)tILs and observed an accelerated tumor *** specifically explore the function of IRF4 in antitumor cD8^(+)t cells,we conducted 3 adoptive cell therapy(Act)***,depleting ***^(+)cD8^(+)tILs derived from Act significantly accelerated tumor growth,emphasizing their crucial role in controlling tumor ***,deleting the Irf4 gene in antitumor cD8^(+)t cells used for Act led to a reduction in the frequency and effector differentiation of cD8^(+)tILs,completely abolishing the antitumor effects of ***,we performed a temporal deletion of the Irf4 gene in antitumor cD8^(+)t cells during Act,starting from 20 days after tumor implantation,which significantly compromised tumor ***,sustained expression of IRF4 is essential for maintaining cD8^(+)t cell immunity in the melanoma model,and these findings carry noteworthy implications for the advancement of more potent immunotherapies for solid tumors.
