Evaluation of the effects of acyclovir and/or human amniotic membrane on herpes virus culture and quantitative virus inactivity by real-time polymerase chain reaction

·AIM: To investigate the permeability of amniotic membrane in herpes virus cell culture to acyclovir with real time polymerase chain reaction(RT-PCR).·METHODS: Madin-Darby Bovine Kidney(MDBK) cell culture and Bovine Herpes Virus(BHV1) type 1 were used in the study. Cell cultures were grouped into two on the basis of herpes virus inoculation. Each group was sub-grouped into three. Amniotic membrane(V-HAM),acyclovir(V-A), and amniotic membrane and acyclovir(V-HAM-A) were applied to these subgroup cultures,respectively. After the application of the membrane and the drug, the cultures were evaluated at 24 and 48 h for cytopathic effect positive(CPE +) with a tissue culture microscope. In the CPE(+) samples, the DNA was extracted for viral DNA analysis by RT-PCR.·RESULTS: In control cultures without herpes virus CPE was not detected. Besides, amniotic membrane and acyclovir did not have cytotoxic effect on cell *** were detected in Bovine Herpesvirus type-1inoculated cell cultures after amniotic membrane and/or acyclovir application. DNA analysis with RT-PCR indicated that Cycle threshold(Ct) values were lower in the BHV1 and membrane applied group(amniotic membrane group < acyclovir group < membrane and acyclovir group). This showed that membrane did not have antiviral effect. The membrane and acyclovir cell culture groups with high Ct values indicated thatmembrane was permeable and had a low barrier effect to drug.·CONCLUSION: In our in-vitro study, we found that amniotic membrane, which can be used in the treatment of corneal diseases, did not have antiviral effect. Besides,we detected that amniotic membrane was permeable to acyclovir in BHV-1 inoculated MDBK cell ***, more studies are necessary to investigate the quantitative effects of amniotic membrane and acyclovir.

Multivariate statistical methods and GIS based evaluation of the health risk potential and water quality due to arsenic pollution in the K?z?l?rmak River

The K?z?l?rmak River is Turkey?s longest(1,355 km) river and it is important since it constitutes the drinking and irrigation water source of the Central Anatolia Region. In the current study, the spatial distribution, effect on irrigation water quality, environmental pollution status, ecotoxicological and health risks as well as possible sources of 12 potential toxic elements?(PTEs)(manganese(Mn),aluminum(Al), arsenic(As), chromium(Cr), zinc(Zn), iron(Fe), nickel(Ni), cadmium(Cd), cobalt(Co),copper(Cu), mercury(Hg), and lead(Pb)) concentrations from surface water samples in the K?z?l?rmak River were analyzed by using geographical information systems(GIS) software, ecotoxicological indices,and multivariate statistical analysis. As a result of the current study, it was determined that the As concentration is above the World Health Organization(WHO) limit values, and, in terms of the 12 PTEs,there is slight heavy metal contamination in the river according to the Heavy Metal Pollution Index(HPI)and Heavy Metal Evaluation Index(HEI) values and there is a low level of pollution. The Hazard Quotient for ingestion(HQingestion) and Hazard Index(HI) values indicate that non-carcinogenic effects may *** to river water is likely to result in a low level of carcinogenic risk(CR) in adults, mainly due to the As concentration. The water at Sampling Station S3(131.79) cannot be used for drinking and is not suitable for consumption. According to the sodium percentage(Na%), Sodium Absorption Ratio(SAR),and Magnesium Hazard(MH) index results, river water is suitable for agricultural use.

Naringin attenuates oxidative stress,inflammation,apoptosis,and oxidative DNA damage in acrylamide-induced nephrotoxicity in rats

Water quality and sediment contamination assessment of Pazarsuyu Stream, Turkey using multivariate statistical methods and pollution indicators

The spatio-temporal variations of surface water quality and the level of metal contamination in the sediment of Pazarsuyu Stream were determined by using multivariate statistical techniques and pollution indicators,*** water and sediment samples were collected monthly from four different stations along the Pazarsuyu Stream between June 2014 and May 2015 and analyzed using standard *** were assessed according to national and international water quality criteria and the water quality parameters in some stations were found to be over these criteria *** present study showed that statistical techniques such as Pearson Correlation Index (PCI),Principal Component Analysis (PCA),Cluster Analysis (CA),One Way ANOVA need to be used to get better information for the studies on the water quality of surface *** identified six factors accounting for 82.88% of the total variation in the *** abundance of sediment heavy metals was in the order of Fe > Mn > Zn > Pb > Cu> Cr > Co > *** heavy metal contamination in the sediments was evaluated by applying the enrichment factor (EF),contamination factor (CF),geoaccumulation index (lgeo) and potential ecological risk index (PERI).Pb was heavily contaminated and has a significant enrichment of contamination according to calculated EF ***,the stream water can be used for irrigation with precaution but extensive treatment required before using for domestic purposes to prevent adverse public health effects.

Intranodal palisaded myofibroblastoma （IPM）, a benign, rare primary mesenchymal neoplasm, located in the inguinal lymph nodes, was described as “ palisaded myofibroblastoma” by Weiss et al, and “intranodal hemorrhagic spindlecell tumor with amianthoid fibers” by Suster and Rosai. Similar neoplasms have been previously reported as intranodal schwannoma. The age of the patients ranged between 19-70 years, and more men are affected than women.

A comprehensive study of hydrogen production from ammonia borane via PdCoAg/AC nanoparticles and anodic current in alkaline medium:experimental design with response surface methodology

In this paper,the optimization of hydrogen(H2)production by ammonia borane(NH3BH3)over PdCoAg/AC was investigated using the response surface ***,the electro-oxidation of NH3BH3 was determined and optimized using the same method to measure its potential use in the direct ammonium boran fuel ***,the ternary alloyed catalyst was synthesized using the chemical reduction *** synergistic effect between Pd,Co and Ag plays an important role in enhancement of NH3BH3 *** addition,the support effect could also efficiently improve the catalytic ***,the effects of NH3BH3 concentration(0.1-50 mmol/5 mL),catalyst amount(1-30 mg)and temperature(20℃-50℃)on the rate of H2 production and the effects of temperature(20℃-50℃),NH3BH3 concentration(0.05-1 mol/L)and catalyst amount(0.5-5 jiL)on the electro-oxidation reaction of NH3BH3 were investigated using the central composite design experimental *** implementation of the response surface methodology resulted in the formulation of four models out of which the quadratic model was adjudged to efficiently appropriate the experimental data.A further statistical analysis of the quadratic model demonstrated the significance of the model with a pvalue far less than 0.05 for each model and coefficient of determination(R2)of 0.85 and 0.95 for H2 production rate and NH3BH3 electrroxidation peak current,respectively.

Probiotic bacteria attenuates cisplatin-induced nephrotoxicity through modulation of oxidative stress, inflammation and apoptosis in rats

Objective: To investigate the effects of probiotic bacteria on cisplatin(CP)-induced nephrotoxicity. Methods: In the present study, 50 Sprague-Dawley rats were used and randomly divided into five groups including control, CP, probiotic bacteria treatment groups with different doses(0.5 and 1 mL) and only probiotic bacteria group. After CP and probiotic administration on seven days, rats sacrificed under anesthesia on the eighth day. The serum urea, creatinine, and blood urea nitrogen levels were analyzed. In renal tissue, malondialdehyde levels, superoxide dismutase and glutathione activity, interleukin-8, interleukin-1β and tumor necrosis factor-alpha levels were determined and histopathological and immunohistochemical changes were also examined. Results: According to results, urea, creatinine and blood urea nitrogen levels as well as kidney weights increased in CP group. Also, CP induced inflammation, oxidative stress, DNA damage and apoptosis in kidney tissue and caused histopathological changes. Administration of the high dose of probiotic bacteria could prevent these changes and damages. Conclusions: This study reveals that probiotic bacteria has protective effects on CP-induced renal damage in rats.

Free and liposome form of gallic acid improves calvarial bone wound healing in Wistar rats

Comparative Analysis of Bridges with AASHTO and Florida I-Beam Girders

A comparative study of two pre-stressed girder bridges, one with AASHTO (American Association of State Highway and Transportation Officials) Type III girders and the other with new FIB (Florida I-beam) girders, is presented. FIB girders are expected to provide increased lateral stiffness, higher load carrying capacity, cost-efficiency and better reliability. In this paper, the first bridge that is analyzed is a 3-span bridge designed with six AASHTO Type III girders, and the second bridge has four FIB girders with the same span length, width and girder depth. The bridges are analyzed for Florida state legal loads SU4 and C5. Both bridges are analyzed using a sophisticated finite element method. The deflections, moment envelopes, section capacity and live load rating of the two bridges are obtained and compared. FIB girders have higher vertical stiffness, higher section capacity providing higher load rating than the AASHTO girders.
