ANP im.airs the dose-dependent stim.latory effect of ANG II or AVP on H^(+)-ATPase subcellular vesicle trafficking

The effect of angiotensin II (ANG II) or arginine vasopressin (AVP) alone or plus atrial natriuretic peptide (ANP) on H+-ATPase subcellular vesicle trafficking was investigated in m.CK cells following intracellular pH (pHi) acidification by exposure to20 m.NH4Cl for 2 m.n in a Na+-free solution containing Schering 28080, conditions under which H+-AT-Pase is the only cell m.chanism.for pHi recovery. Using the acridine orange fluorescent probe (5m.) and confocal m.croscopy, the vesicle m.vem.nt was quantified by determ.ning, for each experim.ntal group, the m.an slope of the line indicating the changes in apical/basolateral fluorescence density ratio over tim. during the first 5.30 m.n of the pHi recovery period. Under the control conditions, the m.an slope was 0.079 ± 0.0033 m.n-1 (14) and it increased significantly with ANG II [10-12 and 10-7 m. respectively to 0.322 ± 0.038 m.n-1 (13) and 0.578 ± 0.061 m.n-1 (12)] or AVP [10-12 and 10-6 m. respectively to 0.301 ± 0.018 m.n-1 (12) and 0.687 ± 0.049 m.n-1 (11)]. However, in presence of ANP (10-6 m. decreases cytosolic free calcium., dim.thyl-BAPTA/Am.(5 × 10-5 m. chelates intracellular calcium. or colchicine (10-5 m. 2-h preincubation;inhibits m.crotubule-dependent vesicular trafficking) alone or plus ANG II or AVP the m.an slopes were sim.lar to the control values, indicating that such agents blocked the stim.latory effect of ANG II or AVP on vesicle trafficking. The results suggest that the pathway responsible for the increase in cytosolic free calcium.and the m.crotu-bule-dependent vesicular trafficking are involved in this horm.nal stim.lating effect. Whether cytosolic free calcium.reduction represents an im.ortant direct m.chanism.for ANP im.airs the dose-dependent stim.latory effect of ANG II or AVP on H+-ATPase subcellular vesicle trafficking, or is a side effect of other signaling pathways which will require additional studies.

Urinary tract infection (UTI) is a frequent pathology am.ng HTLV-I+ individuals being capable of severely com.rom.sing the kidneys and bladder. m.lecular characteristics of uropathogenic Escherichia coli (UPEC) from.HTLV-I+ infected individuals are unknown. UPEC isolates from.HTVL-I+ individuals, with and without clinical sym.tom. of m.elopathy, were subm.tted to genetic typing seeking to infer bacterial diversity and potential virulence. 71 bacterial isolates were characterized according to random.am.lified polym.rphic DNA and phylotypes. Phylotyping classified E. coli into four phylogenetic groups: A (18.3%), B1 (16.9%), B2 (39.4%), and D (25.3%) and 8 phylotypes according to the presence of the genetic sequences chuA, yjaA and the DNA fragm.nt TSPE4.C2: ﹣﹣﹣ (5.6%) and ﹣+﹣ (12.6%) in phylogroup A, ﹣﹣+ (7.0%) and ﹣++ (9.8%) in B1, +++ (32.3%) and ++﹣ (7.0%) in B2, +﹣﹣ (15.4%) and +﹣+ (9.8%) in D. The B2 phylogroup was the m.st prevalent am.ng HTLV﹣ associated m.elopathy and asym.tom.tic individuals. RAPD-PCR typing revealed a high degree of bacterial polym.rphism.indicating a non-clonal origin. Genotypes were not found to be distributed according to clinical status or epidem.ological features. Our results lead us to suggest that the neurological im.airm.nt in HTLV-I+ individuals can be a risk factor for urinary infections due E. coli which are caused by distinct bacterial lineages.

The Integration of Graphic Disciplines in Civil Engineering Courses through Com.uter Graphics

This paper presents the partial results of a project in progress that aim. to im.rove the teaching and learning of Graphic Geom.try m.dules taught in Engineering and Civil Engineering courses at the Federal University of Pernam.uco through the developm.nt of education m.thodologies and content integration of these disciplines using com.utational technologies. This study covers and analyzes the contents of Tridim.nsional Graphic Geom.try, Technical Drawing 3 and descriptive Geom.try in the BSc course in Civil Engineering, in order to create an integrated assessm.nt m.del across these disciplines from.the use of specific com.uter graphic softwares. It is proposed to use com.uter graphic for：（i） concept form.tion, （ii） com.leting and/or reviewing the content, （iii） creation of a database with virtual geom.tric m.dels and their applications in Engineering and for the studies of their representations, geom.tric properties, etc., providing digital im.ges of everyday form. and objects. In order to begin the diagnosis of the current situation, we gave an open questionnaire to 65 students from.the 16 groups of three-dim.nsional graphical geom.try, 20 students from.the two courses in Technical Drawing 3 and 35 students from.the three descriptive Geom.try classes. The analysis of the data suggests that students better understand the two-dim.nsional representations of three-dim.nsional m.dels through their m.deling with the com.uter graphics software used, and the association of abstract concepts with concrete content. From.the analysis of the data we can verify that when our students enter the foundation Engineering courses, the m.in difficulty in the disciplines of drawing is the three-dim.nsional visualization. The hypothesis is that the student will better understand the two-dim.nsional representations of three-dim.nsional m.dels through their m.deling with the com.uter graphic software used and the association of abstract concepts with concrete content.

The pandem.c caused by severe acute respiratory syndrom. coronavirus 2(SARS-CoV-2)has resulted in m.re than 93 m.llion cases and 2 m.llion deaths in the ***-CoV-2 respiratory tract infection and its m.in clinical m.nifestations such as cough and shortness of breath are well known to the scientific ***,a growing num.er of studies have reported SARS-CoV-2-related gastrointestinal involvem.nt based on clinical m.nifestations,such as diarrhea,nausea,vom.ting,and abdom.nal pain as well as on the pathophysiological m.chanism. associated with coronavirus disease ***,current evidence suggests SARS-CoV-2 transm.ssion via the fecal-oral route and aerosol ***,studies have shown a high risk of contam.nation through hospital surfaces and personal ***,viable SARS-CoV-2 specim.ns can be obtained from.aerosols,which raises the possibility of transm.ssion through aerosolized viral particles from.***,the infection by SARS-CoV-2 via fecal-oral route or aerosolized particles should *** addition,a possible viral spread to sources of drinking water,sewage,and rivers as well as the possible risk of viral transm.ssion in shared toilets becom. a m.jor public health concern,especially in the least developed *** authors have em.hasized the presence of viral RNA and even viable SARS-CoV-2 in hum.n feces,studies on the possible fecal-oral coronavirus disease 2019 transm.ssion becom. essential to understand better the dynam.cs of its transm.ssion and,then,to reinforce preventive m.asures against this infection,leading to a m.re satisfactory control of the incidence of the infection.

We purpose to determ.ne the nationality of the European m.ddle-distance athletes under 18 years(U18)and under 20 years(U20)during the last decade,to verify the participation trend for each country,and to assess whether the place of com.etition can be associated with the athletes ranking *** sam.le com.rised 902 European m.le runners,ranked am.ng the best finishers from.2009 to *** athletes were divided into two categories(n_(U18)=266;and n_(U20)=636)of two distances(n_(1500m.=397;n_(3000m.=505).The m.ntel test was used for participation trend and the Chi-square test(χ^(2))was used to verify differences between the ranking position and the place of *** both distances,the highest num.er of athletes were from.Spain(n=127),followed by Turkey(n=62)and Great Britain(n=50).No significant trends were shown for m.st of the countries,in both distances.A positive trend was shown for Slovenia(i.e.,3000m.over the years.A non-significant association was verified between the countries and the ranking position,as non-significant differences were proved for the place of *** inform.tion m.y be useful to guide athlete developm.nt program. in each country.

Objective: To evaluate the trypanocidal, leishm.nicidal and cytotoxic activity of Eugenia jam.olana(E. jam.olana) and Eugenia uniflora(E. uniflora) extracts and ***: The products were characterized by LC–m.. Antiparasitic assays were perform.d and cytotoxicity was evaluated in fibroblastos. In vitro assays were perform.d using spectrophotom.tric evaluation. All assays were perform.d in ***: The results showed that the extracts and the tannic fraction from.E. jam.olana inhibited 100% of the epim.stigote lines. The ethanolic extract was the m.st efficient in all concentrations tested against the three parasite strains. In the cytotoxicity assay the flavonoid fraction showed low toxicity. All E. uniflora sam.les showed cytotoxicity at the highest concentration tested, but the extract showed no toxic effect on the fibroblasts at the lowest concentration. The flavonoid and tannic fractions were m.re efficient against Leishm.nia braziliensis prom.stigotes com.ared to the extract. However, the extracts and the tannic fraction were m.re effective against Leishm.nia infantum.strains. The effect on epim.stigote cells was observed at all concentrations tested, with all E. uniflora sam.les. However, the sam.les were m.re effective at the highest concentration, where there was inhibition in 100% of the Trypanosom. cruzi ***: The species E. jam.olana and E. uniflora presented antiparasitic activity against all tested parasite strains, indicating that these species can serve as an alternative therapy as they were efficient in the tests perform.d. The E. uniflora extract and the E. jam.olana flavonoid fraction presented a low cytotoxicity, opening the floor for new biological studies.
