MET receptor tyrosine kinase promotes the generation of functional synapses in adult cortical circuits

Loss of synapse and functional connectivity in brain circuits is associated with aging and neurodegeneration,however,few molecular mechanisms are known to intrinsically promote synaptogenesis or enhance synapse *** have previously shown that MET receptor tyrosine kinase in the developing cortical circuits promotes dendritic growth and dendritic spine *** investigate whether enhancing MET in adult cortex has synapse regenerating potential,we created a knockin mouse line,in which the human MET gene expression and signaling can be turned on in adult(10–12 months)cortical neurons through doxycycline-containing *** found that similar to the developing brain,turning on MET signaling in the adult cortex activates small GTPases and increases spine density in prefrontal projection *** findings are further corroborated by increased synaptic activity and transient generation of immature silent *** MET signaling resulted in an increasedα-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid/N-methyl-Daspartate(AMPA/NMDA)receptor current ratio,indicative of enhanced synaptic function and *** data reveal that enhancing MET signaling could be an interventional approach to promote synaptogenesis and preserve functional connectivity in the adult *** findings may have implications for regenerative therapy in aging and neurodegeneration conditions.

Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tNGCs). However, the use of tNGCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). Given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tNGC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tNGCs and compared the various features of these improved silk^(+) tNGCs with the tubes without holes(silk^(–) tNGCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tNGCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tNGCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tNGCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tNGCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. The discovery of genes related to Parkinson's disease(PD)provides an imporant basis for the progress of research into PD,helps to clarify is mechanism,and provides many new opportunities to find therapeutic targelts[1.2].The article by Deng and colleagues in 2016 found a new autosomal dominant PD(ADPD)gene,transmembrane protein 230(TMEM230).Four PD-linked mutations were identifed in PD patients:Y92C,RI4IL.184Wexr*5(replacing the stop codon with six ***,in the C-terminus in North American patients),and 184PGext*5(replacing the stop codon with proline and glycine,thus adding seven amino acids,PGHPPHS,to the C-lerminus in Chinese families with PD)[3].Subsequenly,studies have suggested thal TMEM230 variants are related to PD pathogenesis in viro[4-6].

The current study analyzes the contribution of 10 water quality parameters(including pH,turbidity,conductivity,total dissolved solids(TDS),hardness,total organic carbon(TOC),alkalinity,calcium ions,chlorides and sulfates) to corrosion extent of stainless steel valves taken from different locations in a reverse osmosis system of a reclaimed water *** valves were in service for 5 *** spectroscopy and X-ray photoelectron spectroscopy analyses are conducted to quantify corrosion products on different valves under various water quality *** that basis,bivariate and multivariate regression analyses between the 10 water quality parameters and the corrosion extent of valve specimens(represented by metal loss percentage(MLP) values) are carried out to check the contribution of those water quality parameters to *** results indicate that the proportions of metal oxides as corrosion products vary according to the corrosion extent of the *** no linear correlation is founds all 10 water quality parameters except for pH show a significant positive correlation with the MLP values of the valve ***,results of multivariate regression suggest that the variation of MLP can be explained by turbidity,TDS,TOC and sulfates.A positive contribution of turbidity,TDS and TOC to MLP is observed,whereas the contribution of sulfates is *** results from the current work help to identify the reasons for water quality-induced failure of stainless steel equipment in RO systems.

Biochar pores in the micrometer range(1-100μm)derive from cellular structures of the plant biomass subjected to pyrolysis or can be the result of mechanical processing,such as *** this study,synchrotron X-ray microtomography was used to investigate the internal pore structure of softwood pellet biochar produced by slow pyrolysis at 550 and 700°*** microtomographic data sets consisted of 2025 images of 2560×2560 voxels with a voxel side length of 0.87μ*** three-dimensional reconstructions revealed that pelleting and pyrolysis significantly altered the pore structures of the wood feedstock,creating a network of connected pores between fragments that resembled the wood *** higher pyrolysis temperature increased the specific surface area(as determined by BET nitrogen adsorption),it did not affect the total observed *** analysis was applied to assess the characteristics of the frequency distribution of pores along each of the three dimensions of reconstructed images of five softwood pellet biochar *** resulting singular-ity and Rényi spectra(generalized dimensions)indicated that the distribution of porosity had monofractal scaling behavior,was homogeneous within the analyzed volumes and consistent between replicate ***,the pore distributions were isotropic(direction-independent),which is in strong contrast with the anisotropic pore structure of *** pores at the scale analyzed in this study are relevant,for example,for the supply of plant accessible water and habitable space for microorganisms,our findings combined with the ability to reproduce biochar with such pore distribution offer substantial advantages in various biochar applications.
