Targeted therapy in advanced metastatic colorectal cancer: Current concepts and perspectives

The introduction of new cytotoxic substances as well as agents that target vascular endothelial growth factor (VEgF) and epidermal growth factor receptor (EgFR) signaling has improved clinical outcome of patients with metastatic colorectal cancer (mCRC). In this review we summarize the most relevant clinical data on VEgF and EgFR targeting regimens in mCRC. The effects of available treatment strategies for mCRC are often temporary, with resistance and disease progression developing in most patients. Thus, new treatment strategies are urgently needed. Some gI peptides including gastrin and gastrin releasing peptide, certain growth factors such as insulin-like growth factor-I and II and neuropeptides such as growth hormone releasing hormone (gHRH) are implicated in the growth of CRC. Experimental investigations in CRC with antagonistic analogs of bombesin/gastrin-releasing peptide, gHRH, and with cytotoxic peptides that can be targeted to peptide receptors on tumors, are summarized in the second part of the review.

Physico-chemical properties of 3-methoxy-2-nitrobenzoates of some rare earth elements(Ⅲ)

The complexes of 3-methoxy-2-nitrobenzoates of Pr（Ⅲ）, Nd（Ⅲ）, Sm（Ⅲ）, Eu（Ⅲ）, gd（Ⅲ）, Tb（Ⅲ）, Er（Ⅲ） and Tm（Ⅲ） with the formula： Ln（C8H6NO5）3·2H2O, where Ln=lanthanides（Ⅲ）, were synthesized and characterized by elemental analysis, Forier transform irtrared （FTIR） spectroscopy, magnetic and thermogravimetric studies and also by X-ray diffraction （XRD） measurements. The complexes had col- ours typical for Ln（Ⅲ） ions. The carboxylate groups bound as bidentate chelating. On heating to 1173 K in air they decomposed in the same way, at first, dehydrated in one step to anhydrous salts, and then decomposed to the oxides of respective metals with intermediate formation of the oxycarbonates. The enthalpy values of the dehydration process changed from 133.72 to 44.50 kJ/mol. Their solubility in water at 293 K was of the order of 10-4 mol/dm3. The magnetic moments of analysed complexes were determined by gouy＇s method in the range of 76-303 K.

Therapeutic aspects of c-MYC signaling in inflammatory and cancerous colonic diseases

Colonic inflammation is required to heal infections, wounds, and maintain tissue homeostasis. As the seventh hallmark of cancer, however, it may affect all phases of tumor development, including tumor initiation, promotion, invasion and metastatic dissemination, and also evasion immune surveillance. Inflammation acts as a cellular stressor and may trigger DNA damage or genetic instability, and, further, chronic inflammation can provoke genetic mutations and epigenetic mechanisms that promote malignant cell transformation. Both sporadical and colitis-associated colorectal carcinogenesis are multi-step, complex processes arising from the uncontrolled proliferation and spreading of malignantly transformed cell clones with the obvious ability to evade the host's protective immunity. In cells upon DNA damage several protooncogenes, including c-MYC are activated in parelell with the inactivation of tumor suppressor genes. The target genes of the c-MYC protein participate in different cellular functions, including cell cycle, survival, protein synthesis, cell adhesion, and microRNA expression. The transcriptional program regulated by c-MYC is context dependent, therefore the final cellular response to elevated c-MYC levels may range from increased proliferation to augmented apoptosis. Considering physiological intestinal homeostasis, c-MYC displays a fundamental role in the regulation of cell proliferation and crypt cell number. However, c-MYC gene is frequently deregulated in inflammation, and overexpressed in both sporadic and colitis-associated colon adenocarcinomas. Recent results demonstrated that endogenous c-MYC is essential for efficient induction of p53-dependent apoptosis following DNA damage, but c-MYC function is also involved in and regulated by autophagy-related mechanisms, while its expression is affected by DNA-methylation, or histone acetylation. The paper comprises new analytical data on the nature and occurrence of gold in solid pyrobitumen,closely associated with the main gold-bearing sulfide arsenic ores of the Bakyrchik gold deposit(Kazakhstan),related to post-collisional magmatic-hydrothermal *** mineralization of the deposit occurs mainly in the form of an“invisible”type of gold in the structures of arsenian pyrite and arsenopyrite,and the form of gold-organic compounds of pyrobitumen in carbonaceous-terrigenous sequences of Carboniferous *** and electron microscopic analysis,Raman and FT-Infrared analysis,mineralogical and three-step sequential extraction analysis(NH2OHHCl,H2O2,HNO3?HCl)has been carried out using 9 ore samples(from 3 different types of ores)for a comprehensive study of pyrobitumen and sulfide arsenic ores focusing mainly on organic *** sequentially extracted precious metal content of pyrobitumen reaches up to 7 ppm gold and other metals like Ag 4 ppm,Pt 31 ppb,and Pd 26 ppb,forming metal–organic compounds,while arsenic sulfide minerals incorporate 11 ppm gold,39 ppm Ag,0.49 ppm *** enrichment of gold associating with organic matter and sulfide ore minerals was confirmed in this *** matter was active in the migration of gold and the capture of gold by ***,the reductive organic matter agent released gold,most likely for the sulfide arsenic ore *** was a decisive factor in the concentration,transportation,and preservation of gold in the deposit.

Nanocrystalline (Fe0.5 Co0.5 )86 Hf7 B6 Cu1 HITPERM alloy was investigated as the candidate of soft magnetic material for high temperature applications, compared with Fe86 Hf7B6 Cu1 NANOPERM alloy. Amorphous alloy ribbons were prepared by single-roller melt-spinning technology. Crystallization process of as-quenched ribbon was investigated using differential scanning calorimeter at different heating rates. The coercivity was determined from quasi-static hysteresis loop measured at room temperature using a computerized hysteresis loop tracer. X-ray diffraction with Cu Kα radiation was used to determine the structure. The vibrating sample magnetometer was usedto measure the magnetization as a function of temperature of the nanocrystllized alloys. That Co substitution for Fein alloy enhances the Curie temperature of amorphous alloy and the magnetization of nanocrystalline alloy at hightemperature. After annealing amorphous precursor, the optimum nanocrystalline alloy obtained shows the local minimum coercivity. The coercivity increases with the increasing annealing temperature corresponding to the formation of ferromagnetic phase in the secondary crystallization.

Physiological and pathological role of local and immigrating colonic stem cells

The latest avenue of research is revealing the existence of and role for the colonic stem cells in the physiological renewal of the mucosa and in pathological circumstanc- es where they have both positive and negative effects. In the case of human colon, different levels of stem cell compartments exist. First, the crypt epithelial stem cells, which have a role in the normal crypt epithelial cell dynamics and in colorectal carcinogenesis. Close to the crypts, the second layer of stern cells can be found; the local subepithelial stem cell niche, including the pericryptic subepithelial myofibroblasts that regulate the epithelial cell differentiation and have a crucial role in cancer progression and chronic inflammation-related fibrosis. The third level of stem cell compartment is the immigrating bone-marrow-derived stem cells, which have an important role in wound healing after severe mucosal inflammation, but are also involved in cancer invasion. This paper focuses on stem cell biology in the context of physiological and pathological processes in the human colon.

In their manuscript,“Whole-grain Intake and Pancreatic Cancer Risk-The Danish,Diet,Cancer and Health Cohort”,Schacht et al.(1)report that total whole-grain product(per 50 g/d serving)intake was significantly associated with lower incidence of pancreatic cancer(HR:0.93;95%CI:0.86,1.00).A sex-specific analysis revealed that this association only held among ***,neither total whole-grain(per 16-g serving)or specific individual whole-grain products and cereals were significantly associated with lower pancreatic cancer *** this commentary,I will describe some of the assumptions and biases associated with assessing dietary intake and attempting to find associations with disease.

Background:Proprioceptive accuracy refers to the individual’s ability to perceive proprioceptive information,that is,the information referring to the actual state of the locomotor system,which originates from mechanoreceptors located in various parts of the locomotor system and from tactile receptors located in the *** accuracy appears to be an important aspect in the evaluation of sensorimotor functioning;however,no widely accepted standard assessment *** this systematic review,our goal was to identify and categorize different methods that are used to assess different aspects of proprioceptive ***:A literature search was conducted in 5 different databases(PubMed,SPORTDiscus,PsycINFO,ScienceDirect,and SpringerLink).Results:Overall,1139 scientific papers reporting 1346 methods were included in this *** methods assess 8 different aspects of proprioception:(a)the perception of joint position,(b)movement and movement extent,(c)trajectory,(d)velocity,and the sense of(e)force,(f)muscle tension,(g)weight,and(h)*** apply various paradigms of psychophysics(i.e.,the method of adjustment,constant stimuli,and limits).Conclusion:As the outcomes of different tasks with respect to various body parts show no associations(i.e.,proprioceptive accuracy is characterized by site-specificity and method-specificity),the appropriate measurement method for the task needs to be chosen based on theoretical considerations and/or ecological validity.

Analysing the effect of stand density and site conditions on structure and growth of oak species using Nelder trials along an environmental gradient:experimental design,evaluation methods,and results

Background: Most current approaches in forest science and practice require information about structure and growth of individual trees rather than- or in addition to- sum and mean values of growth and yield at forest stand level as provided by classic experimental designs. By inventing the wheel design, Nelder provided the possibility to turn to the individual tree as basic information unit. Such trials provide valuable insights into the dependency of growth on stand density at particular ***: Here, we present an extension of the original design and evaluation by Nelder.(i) We established Nelder wheels along an environmental gradient through Europe in atlantic climate in Belgium and germany, Mediterranean climate in Italy, continental climate in Hungary as well as on high land climate in Mexico. Such disjunct Nelder wheels along an environmental gradient can be regarded and analysed as a two-factor design with the factors of site condition and stand density.(ii) We present an advanced statistical approach to evaluate density dependent growth dynamics of trees planted in form of the Nelder design, which considers spatio-temporal autocorrelation.(iii)We prove the usefulness of the methods in improving ecological theory concerning density related productivity,trade-offs between facilitation and competition, and allometric relations between size ***: First evaluations based on remeasured Nelder wheels in oak(Quercus robur L.) show a size growth differentiation during the first observation period. In particular, height growth is accelerated under higher competition indicating facilitation effects. We detect furthermore a high variability in allometric ***: The proposed design, methods, and results are discussed regarding their impact on forest practice,model building, and ecological theory. We conclude that the extended Nelder approach is highly efficient in providing currently lacking individual tree level information.
