Effects of weaning on intestinal longitudinal muscle-myenteric plexus function in piglets

Weaning piglets usually suffer from severe diarrhea(commonly known as postweaning diarrhea,PWD)along with intestinal motility *** peristalsis is mainly regulated by the longitudinal muscle-myenteric plexus(LM-MP).To understand the relationship between intestinal LM-MP function and the development of PWD,we compared the intestinal electrical activity,and the transcriptional profile of the LM-MP between 21-day-old piglets(just weaned,n=7)and 24-day-old piglets(suffered the most severe weaning stress,n=7).The results showed that 24-day-old piglets exhibited different degrees of diarrhea.A significant increase in the slow-wave frequency in the ileum and colon was observed in 24-day-old piglets,while c-kit expression in the intestinal LM-MPs was significantly decreased,indicating that PWD caused by elevated slow-wave frequency may be associated with loss of *** real-time quantitative PCR(RT-q PCR)and enzyme-linked immunosorbent assay(ELISA)showed that intestinal LM-MPs in 24-day-old piglets may undergo inflammation and oxidative *** increases in 8-hydroxy-2′-deoxyguanosine and decreases in thioredoxin suggest that weaning may lead to DNA damage in the LM-MP of 24-day-old *** addition,activating transcription factor 3 was significantly upregulated,indicating nerve damage in the LM-MP of 24-day-old *** transcriptomic results showed that most of the differentially expressed genes in the ileal LM-MP after weaning were downregulated and closely related to the cell cycle *** RT-q PCR analysis showed that the relative expression of p21 was upregulated,while the expression of cyclin A2,cyclin B1,and proliferating cell nuclear antigen was downregulated in the ileal and colonic LM-MP of 24-day-old piglets,suggesting that weaning may inhibit cell proliferation and cause G1/S cell cycle arrest in ileal and colonic *** conclusion,weaning may lead to cell cycle arrest by causing DNA damage in the LM-MP,impairing i

金花茶花对2型糖尿病小鼠的降糖及抗氧化作用

目的:综合评价金花茶花的降血糖和抗氧化作用。方法:制备金花茶花水提物(aqueous extract of Camellia nitidissima flower,CFA),利用高效液相色谱-质谱联用法进行组分鉴定;同时,建立2型糖尿病小鼠模型并随机分为模型组,阿卡波糖阳性组(20 mg/kg mb),CFA低、中、高剂量组(200、400、800 mg/kg mb)。灌胃干预5周后,分析各组小鼠一般生长特征,血清中血糖、空腹胰岛素及血脂水平,胰、肝脏氧化应激水平、组织形态变化及细胞凋亡情况。结果:金花茶花具有较高的多酚、多糖含量,清除1,1-二苯基-2-三硝基苯肼自由基和抑制α-葡萄糖苷酶的半抑制浓度分别为(24.14±0.64)、(69.99±1.97)μg/m L,从金花茶花中鉴定出7种主要化合物。此外,金花茶花可有效改善糖尿病小鼠“三多一少”症状,显著降低小鼠空腹血糖、餐后血糖、总胆固醇、甘油三酯、低密度脂蛋白胆固醇、丙二醛水平,升高胰岛素、高密度脂蛋白胆固醇的含量,提升总超氧化物歧化酶、谷胱甘肽过氧化物酶的活性,减轻胰脏、肝脏组织的病理损伤。结论:金花茶花对2型糖尿病小鼠具有显著的降血糖及抗氧化作用。

中国野生刺梨的遗传多样性及遗传结构分析

基于10对EST-SSR引物,对中国8个省29个居群的261份野生刺梨(Rosa roxburghii Tratt.)进行毛细管电泳检测,探究其遗传多样性,并进行聚类分析。结果显示,共扩增出了95个等位基因,位点多态性信息含量(PIC)平均值为0.568;群体水平上,平均等位基因数(NA)和有效等位基因数(NE)分别为3.131和2.331,平均观测杂合度(HO)和预期杂合度(HE)分别为0.508和0.488,平均Shannon’s信息指数(I)为0.858,表明野生刺梨种质具有较高的遗传多样性水平。群体遗传分化分析结果显示,平均遗传分化系数(FST)为0.067,基因流(Nm)为4.511,说明刺梨群体间基因流动较频繁。分子方差分析(AMOVA)结果表明刺梨的遗传变异主要来源于居群内(93.64%)。各居群间的Nei氏标准遗传距离范围为0.054~1.269,平均值为0.657,与地理距离表现为显著相关(r=0.467,P<0.0001)。聚类分析和PCA结果均将29个居群分为3个分支。研究结果说明中国野生刺梨资源核心分布区在我国西南地区。

目的探究血浆circRNA_0001445在老年性骨质疏松症(senile osteoporosis,SOP)中的表达情况及其临床意义。方法选取2020年11月至2022年5月于常州市第一人民医院老年科住院的患者为研究对象,应用双能X射线吸收检测法(DXA)测量骨密度(bone mineral density,BMD)根据BMD结果分为SOP组、骨量减少组及骨量正常组,比较各组间临床资料及circRNA_0001445表达量,采用Pearson相关分析circRNA_0001445与骨密度的的相关性;采用二元Logistic回归分析SOP的危险因素;采用受试者工作特征(ROC)曲线分析circRNA_0001445对SOP的诊断价值;采用配对t检验或配对秩和检验比较SOP治疗前后circRNA_0001445表达量变化。结果共纳入150例患者,每组各50例。SOP组circRNA_0001445表达量(0.58±0.30)明显低于骨量减少组(0.88±0.35)及骨量正常组(1.15±0.62),P<0.001;Pearson相关分析示circRNA_0001445表达量与腰椎BMD(r=0.427,P<0.001)、股骨颈BMD(r=0.363,P<0.001)及髋部BMD(r=0.435,P<0.001)存在正相关;二元Logistic回归示高TC(OR=2.740,95%CI:1.473-5.099)、低BMI(OR=0.674,95%CI:0.543-0.837)及circRNA_0001445(OR=0.019,95%CI:0.002~0.144)是SOP发生的危险因素,P<0.05。ROC曲线分析显示,circRNA_0001445可将SOP组与骨量正常组(AUC=0.810,95%CI:0.728~0.892)、骨量减少组(AUC=0.756,95%CI:0.662~0.849)及非SOP组(AUC=0.783,95%CI:0.706~0.860)区分开来。SOP治疗1年后circRNA_0001445表达量较前升高(1.13±0.61 vs 2.14±0.61),P<0.001。结论circRNA_0001445在SOP中低表达,对SOP的诊断及治疗监测具有一定临床价值。

为探讨蓝莓叶提取物(bluberry leaf extract, BLE)对2型糖尿病(type 2 diabetic, T2D)小鼠糖脂代谢的调节作用,采用高糖高脂饮食诱导辅以链脲佐菌素腹腔注射建立T2D小鼠模型,将其分为模型组、阳性组(20 mg·kg^(-1)·d^(-1)阿卡波糖)、BLE低中高剂量组(200,400,800 mg·kg^(-1)·d^(-1)BLE),另设正常组.灌胃干预35 d,分析小鼠血清中血糖、胰岛素和血脂水平,肝脏中糖代谢水平,胰脏和肝脏中的抗氧化水平变化,结合苏木精-伊红染色切片和原位末端标记染色切片,评估BLE对T2D小鼠胰脏和肝脏组织病理损伤的缓解作用.结果表明,BLE能显著降低T2D小鼠空腹血糖、餐后2 h血糖、总胆固醇、甘油三酯、低密度脂蛋白胆固醇和丙二醛的水平,提升胰岛素、高密度脂蛋白胆固醇、肝糖原和谷胱甘肽过氧化物酶的水平,增强葡萄糖激酶和总超氧化物歧化酶的活性,抑制葡萄糖-6-磷酸酶的活性,减轻胰脏和肝脏组织的病理损伤.综上所述,BLE具有显著的降血糖和降血脂作用,推测其可通过减少T2D小鼠胰脏和肝脏的氧化应激损伤,保护脏器组织细胞的生理形态结构,从而在提升胰腺胰岛素分泌功能的同时改善肝脏糖代谢功能,完成机体血糖和血脂水平的整体性调节.

采集昌平南口镇473件表层土壤样品,收集65件土壤样品(1970年采集的),测试土壤中砷(As)、镉(Cd)、铜(Cu)、铬(Cr)、汞(Hg)、铅(Pb)、镍(Ni)、锌(Zn)等8种重金属元素及三氧化二铝(Al_(2)O_(3))、二氧化硅(SiO_(2))和三氧化二铁(Fe_(2)O_(3))等地球化学指标。采用多种污染评价方法和统计学方法,运用Pearson相关性分析和主成分分析,评估土壤重金属污染风险,分析污染来源。结果表明:重金属Cd含量普遍较高,超筛选值点位较广,Pb、Hg、Cr、Ni、Cu、Zn存在个别点位超筛选值的情况;Cd、Hg、Pb、Zn存在轻微及以上污染点位,Pearson相关性分析和主成分分析显示Cd、Hg、Zn来源相近;Cd和Zn的人为贡献率在1970—2021年间增长明显,且平均值明显高于北京地区背景值,表明研究区可能存在一定的人为污染。

Safety issues related to flammable electrolytes in lithium-ion batteries(LIBs) remain a major challenge for their extended *** use of non-flammable phosphate-based electrolytes has proved the validity in inhibiting the combustion of ***,the strong interaction between Li^(+) and phosphate leads to a dominant solid electrolyte interphase(SEI) with limited electronic shielding,resulting in the poor Li^(+) intercalation at the graphite(Gr) anode when using high-phosphate-content *** mitigate this issue and improve Li^(+) insertion,we propose an “In-N-Out” strategy to render phosphates “noncoordinative”.By employing a combination of strongly polar solvents for a “block effect” and weakly polar solvents for a “drag effect”,we reduce the Li^(+)–phosphate *** a result,phosphates remain in the electrolyte phase(“In”),minimizing their impact on the incompatibility with the Gr electrode(“Out”).We have developed a non-flammable electrolyte with high triethyl phosphate(TEP) content(>60 wt.%),demonstrating the excellent ion conductivity(5.94 mS cm^(-1) at 30 ℃) and reversible Li^(+) intercalation at a standard concentration(~1 mol L^(-1)).This approach enables the manipulation of multiple electrolyte functions and holds the promise for the development of safe electrochemical energy storage systems using non-flammable electrolytes.

Analysis and application of carbon-bearing rate of reservoirs in the Puguang gas field,northeastern Sichuan Basin

With wells Puguang 2 and 6 in the Puguang gas field as the research objects by choosing organic carbon as characterization reservoir carbon(indirectly reflecting the bitumen contents),this paper discusses the relationship between bitumen contents and lithologic characteristics,reservoir characteristics,and calculates the volume of paleo-oil and oil cracked *** study shows that the organic carbon contents of the reservoirs are related to lithologic *** to variations in the particle size or structure of rocks,the porosity of the reservoirs varies,furthermore,it will influence the contents of organic *** the relationship between these two parameters is *** on the analysis,the relationship between total organic carbon(TOC)and the contents of reservoir bitumen also appears to be *** volume of paleo-oil and cracked gas of the Changxing-Feixian’guan Formation are respectively 495 million tons and 360.4 billion m3 through the calculation of reservoir’s organic ***,the relationship between carbon-bearing rate and characteristics of the reservoirs can be used to indirectly evaluate reservoir ***,the carbon-bearing rate of the reservoirs also reflects the contents of reservoir bitumen,paleo-oil as well as natural *** makes a new approach for the study of the types of reservoirs and the amounts of paleo-oil and oil cracked gas in northeastern Sichuan Basin.

Substantial NO_(x) emission mitigation is crucial for the synergistic reduction of particulate matter and ozone(O_(3))pollution in *** traditional air quality model does not consider heterogeneous HONO chemistry,leading to uncertainties in estimating the benefits of NO_(x) *** studies have shown that the parameterization of heterogeneous HONO formation increases both the simulated value of sulfate–nitrate–ammonium(SNA)and that of O_(3),thus adding the heterogeneous reactions of HONO into air quality models inevitably leads to changes in the estimated benefits of NO_(x) *** we investigated the changes in SNA and O_(3)concentrations from NO_(x) emission reduction before and after adding heterogeneous HONO reactions in the Community Multi-Scale Air Quality(CMAQ)*** heterogeneous HONO reactions in the simulation improved the benefits of NO_(x) reduction in terms of SNA control in *** 80%NO_(x) reduction,the reduction in SNA increased from 36.9%without considering heterogeneous HONO reactions to 42.8%with heterogeneous HONO *** reduction in the maximum daily 8h average(MDA8)O_(3)in summer caused by NO_(x) reduction increased slightly from 4.7%to 5.2%after adding heterogeneous HONO *** results in this study highlight the enhanced effectiveness of NO_(x) controls for the reduction of SNA and O_(3)after considering heterogeneous HONO formation in a complex chemical ambient,demonstrating the importance of NO_(x) controls in reducing PM2.5 and O_(3)pollution in China.

Mesenchymal stem cells(MSCs)experience substantial viability issues in the stroke infarct region,limiting their therapeutic efficacy and clinical *** levels of deadly reactive oxygen radicals(ROS)and proinflammatory cytokines(PC)in the infarct milieu kill transplanted MSCs,whereas low levels of beneficial ROS and PC stimulate and improve engrafted MSCs’*** on the intrinsic hormesis effects in cellular biology,we built a microglia-inspired MSC bioengineering system to transform detrimental high-level ROS and PC into vitality enhancers for strengthening MSC *** system is achieved by bioorthogonally arming metabolic glycoengineered MSCs with microglial membrane-coated nanoparticles and an antioxidative extracellular protective *** this system,extracellular ROSscavenging and PC-absorbing layers effectively buffer the deleterious effects and establish a microlivable niche at the level of a single MSC for ***,the infarct’s inanimate milieu is transformed at the tissue level into a new living niche to facilitate *** engineered MSCs achieved viability five times higher than natural MSCs at seven days after transplantation and exhibited a superior therapeutic effect for stroke recovery up to 28 *** vitality-augmented system demonstrates the potential to accelerate the clinical translation of MSC treatment and boost stroke recovery.
