Inhibition of rho kinase attenuates high flow induced pulmonary hypertension in rats

Background The RhoA/Rho kinase pathway may participate in the pathogenesis of hypoxia and monocrotaline induced pulmonary hypertension. This study tested whether RhoA/Rho kinase pathway is involved in the pathogenesis of high flow induced pulmonary hypertension in rats. Methods Male Wistar rats （4 weeks） were randomly divided into 4 shunt groups, 4 treated groups and 4 control groups. Shunt and treated groups underwent left common carotid artery/external jugular vein shunt operation. Control groups underwent sham operation. Treated groups received fasudil treatment and the others received same dose of saline. At weeks 1, 2, 4 and 8 of the study, nght ventricular systolic pressure was measured and blood gases were analysed to calculate Qp/Qs. The weight ratio of right ventricle to left ventricle plus septum and the mean percentage of medial wall thickness in moderate sized pulmonary arteries were obtained. RhoA activity in pulmonary arteries was detected using Rho activity assay reagent. Rho kinase activity was quantified by the extent of MYPT1 phosphorylation with Western blot. Proliferating cells were evaluated using proliferating cell nuclear antigen immunohistological staining, Results Carotid artery/jugular vein shunt resulted in high pulmonary blood flow, both an acute and a chronic elevation of right ventricular systolic pressure, significant medial wall thickening characterized by smooth muscle cells proliferation, nght ventricular hypertrophy and increased activation of RhoA and Rho kinase. Fasudil treatment lowered pulmonary artery systolic pressure, suppressed pulmonary artery smooth muscle cells proliferation, attenuated pulmonary artery medial wall thickening and inhibited right ventricular hypertrophy together with significant suppression of Rho kinase activity but not Rho activity. Analog simulation of urban construction land supply and demand in Chang-Zhu-Tan Urban Agglomeration based on land intensive use

Urban land intensive use is an important indicator in harmonizing the relationship between land supply and demand. The system dynamics(SD) can be used to construct the feedback loop between urban construction land supply and demand and index variable function. Based on this, this study built a supply and demand system dynamic model of urban construction land for Chang-Zhu-Tan urban agglomeration. This model can simulate the change trends of supply and demand of construction land, industrial land, and residential land in 2016–2030 by three scenarios of low, medium, and high intensity modes. The results showed that the scale of construction land of urban agglomeration is expanding, with a rapid increase rate for the urban construction land. The scale and speed of land use based on the three intensity modes existed differences. The large scale and supply of construction land in the low intensity mode caused easily the waste of land resources. In high intensity mode, the scale and supply of construction land were reduced against the healthy development of new-type urbanization. In the medium intensity mode, the scale and supply of land use adapted to the socio-economic development and at the same time reflected the concept of modern urban development. In addition, the results of this study found that the proportion of industrial land in construction land ranged from 15% to 21%, which increased year by year in the low intensity mode, and decreased slowly and stabilized in medium and high intensity modes. The proportion of residential land in construction land ranged from 27% to 35%, which decreased in the low and the medium intensity modes, and maintained a high level in the higher intensity mode. This study contributes to provide scientific reference for decision-making optimization of land supply and demand, urban planning, and land supply-side reform.

Study on biological characters of SGC7901 gastric cancer cell-dendritic cell fusion vaccines

AIM： To detect the biological characters of the SGC7901 gastric cancer cell-dendritic cell fusion ***： The suspending living SGC7901 gastric cancer cells and dendritic cells were induced to be fusioned by polyethylene glycol. Pure fusion cells were obtained by selective culture with the HAT/HT culture systems. The fusion cells were counted at different time points of culture and their growth curves were drawn to reflect their proliferative activities. The fusion cells were also cultured in culture medium to investigate whether they could grow into cell clones. MTT method was used to test the stimulating abilities of the fusion cells on T lymphocytes＇ proliferations. Moreover, the fusion cells were planted into nude mice to observe whether they could grow into new planted tumors in this kind of immunodeficiency ***： The fusion cells had weaker proliferative activity and clone abilities than their parental cells. When they were cultured, the counts of cells did not increase remarkably, nor could they grow into cell clones in culture medium. The fusion cells could not grow into new planted tumors after planted into nude mice. The stimulating abilities of the fusion cells on T lymphocytes＇ proliferations were remarkably increased than their parental dendritic cells. CONCLUSION： The SGC7901 gastric cancer cell-dendritic cell fusion vaccines have much weaker proliferative abilities than their parental cells, but they keep strong abilities to irritate the T lymphocytes and have no abilities to grow into new planted tumors in immunodeficiency animals. These are the biological basis for their antitumor biotherapies.

Postnatal weight gain in very low birth weight infants in Beijing and the risk of retinopathy of prematurity

AIMTo analyze the low weight gain (WG) from birth to 4 and 6wk of life to predict the development of retinopathy of prematurity (ROP) among very low birth weight (VLBW) preterm *** hundred and three newborns with VLBW were analyzed. Body weight measurements were recorded weekly. In all patients, the proportion of the WG was defined as the preterm weight measured at the 4th and 6th weeks of life minus the birth weight (BW) divided by the BW. Other risk factors for ROP were also *** gestational age and mean BW of the whole cohort were 29.56±1.44wk and 1270.58±176.18g respectively. WG proportion at 4wk postnatal age (18.89%±13.58%) were significantly lower in infants with ROP (P=0.003). WG proportion at 6wk was not different between ROP and no ROP group (42.48%±20.36% vs 46.43%±15.65% P=0.118). When all the other risk factors signiﬁcant for ROP were included in the logistic regression poor WG did not arise as an independent risk factor. Area under the ROC curve was 0.591 (95%CI: 0.515-0.666; P=0.016). For ROP, the best discriminative cutoff of 18.06% of the proportional WG at the 4th week over the BW, sensitivity and specificity values were 67.3% and 50.0% *** WG proportion in the first 4wk of life is maybe an additional predictor of ROP in very low BW infants. Preterm babies with low BW and low WG proportion should be followed closely for ROP.

To the Editor： A 20-year-old woman complained of two painful pruritic lesions on her left index finger. She had been bitten by a lamb on her index finger a month earlier; the lamb died 2 days later. Five days after having been bitten, she noticed two papules on her left index finger that enlarged gradually. Physical examination revealed two indurated nodules 8-mm and 10-mm in diameter, each with central umbilication and surrounded by a reddish halo [Figure i a]. The patient had no fever, respiratory symptoms, or lymphadenopathy. The informed consent was written.

The hospitalization time, hospitalization costs, operation time, operative complications and post-operative liver functions of the two groups of patients were ***: The operation time and post-operative liver functions of the two groups of patients had no significant differences, while the hospitalization time, hospitalization costs and operative complications of the laparoscopic hepatic left lateral lobectomy combined with fiber choledochoscopic exploration in the common bile duct group were significantly lower than those in the traditional open operation ***: For patients with gallstones in the left lobe of liver and in the common bile duct, laparoscopic hepatic left lateral lobectomy combined with fiber choledochoscopic exploration of the common bile duct can significantly shorten the hospitalization time, reduce the hospitalization costs and the post-operative complications,without prolonging the operation time and bringing about more liver function damages compared with traditional open operation. This kind of operation has more advantages than traditional open operation.

Effect of initial ammonium concentration on a one-stage partial nitrification/anammox biofilm system:Nitrogen removal performance and the microbial community

One-stage partial nitrification coupled with anammox(PN/A)technology effectively reduces the energy consumption of a biological nitrogen removal *** nitrite-oxidizing bacteria(NOB)is essential for this technology to maintain efficient nitrogen removal *** ammonium concentration(IAC)affects the degree of inhibited *** this study,the effect of the IAC on a PN/A biofilm was investigated in a moving bed biofilm *** results showed that nitrogen removal efficiency decreased from 82.49%±1.90%to 64.57%±3.96%after the IAC was reduced from 60 to 20 mg N/L,while the nitrate production ratio increased from 13.87%±0.90%to 26.50%±3.76%.NOB activity increased to1,133.86 mg N/m^(2)/day after the IAC decreased,approximately 4-fold,indicating that the IAC plays an important inhibitory role in *** rate-limiting step in the mature biofilm of the PN/A system is the nitritation process and is not shifted by the *** analysis of the microbial community structure in the biofilm indicates that the IAC was the dominant factor in changes in community *** and *** were the main anammox bacteria,and Nitrosomonas and Nitrospira were the main AOB and NOB,*** did not affect the difference in growth between *** and ***,modulating the IAC promoted the PN/A process with efficient nitrogen removal performance at medium to low ammonium concentrations.
