Aerobic Cr(VI) Reduction by an Indigenous Soil Isolate Bacillus thuringiensis BRC-ZYR2

Chromium(Cr) may cause losses in the yield of field plant, which is one of the favorite habitats of Bacillus thuringiensis(Bt). The purposes of our study were to assess the Cr(VI)-resistance and Cr(VI)-reducing abilities of an indigenous soil isolate of Bt and to determine the factors governing Cr(VI) reduction. Towards this end a novel dichromate-reducing Bt BRC-ZYR2, characterized with insecticidal crystal proteins(ICPs), was isolated from a uranium deposit. Minimum inhibitory concentrations(MICs) of Cr(VI) were determined by broth dilution method and the concentrations of Cr(VI) and total Cr in the supernatant were quantified colorimetrically using 1,5-diphenylcarbazide(DPC) reagent and a mixture of sulfuric-nitric acids, respectively. The isolate contained five ICP genes(cry1Ba, cry1 Bb, cry1Be/cry1 Bf, cry9 Ca and cry9Da) and exhibited a high level of Cr(VI) resistance with MICs of 150 mg L-1at pH 7.0 and 30?C, and 500 mg L-1under optimal conditions(pH 9.0 and 40?C). The total Cr concentration was similar to initial concentration of Cr(VI) under the optimal condition, suggesting that the essential removal of the Cr(VI) was dependent on Bt reduction. Under optimal conditions, the initial Cr(VI) concentrations from 25 to 75 mg L-1significantly decreased in 24 h after incubation. Addition of Mn2+, Co2+, Mo2+and Cu2+activated Bt-mediated Cr(VI) reduction, while Zn2+, Ni2+and glucose were found to inhibit the reduction. Our results indicated that this isolate could be a promising biopesticide with the potential for both insect biocontrol and Cr bioremediation in the field.

Analysis on small-earthquake composite fault plane solutions from 1961 to 1999 in Xinfengjiang reservoir area

To understand the earthquake characteristics in Xinfengjiang (XFJ for short) reservoir area, we collected the small earthquakes occurred in the area from 1961 to 1999. We segmented this 40-year period, parted the research region and calculated the composite fault plane solution of each block, disscussed the effect characteristics of stress field of water pressure using Mohrs stress circle. The final result shows that the main rupture pattern was very different before and after the M = 6.1 main shock, changing from strike slip to normal rupture. The maximum principal stress axes of composite fault plane solutions are characterized by synchronous change with water level.

Evaluation of harmonious development for economy-environment system in coastal cities: a case study of Tianjin Municipality

With the aim of the harmonious development of economy-environment system in coastal cities in China. an index ,system used to evaluate the economy-environment system is built up in this paper, which includes four aspects： economy, environment, resources, and ocean industry. Based on the analysis on present condition and future trends of economic development in Tianjin and the quantification of various evaluation indices, the aathor applies integrated index valuation model to valuate the harmonious development af economy-environment of Tianjin. The results show that the coordinated degree of economy-environment would drop down in the future, from 0.95（superior level of harmonious development） in 2000 to 0.59（inferior level of harmonious development） in 2015. under the circumstance of the current economic development mode. The level of comprehensive development of Tianjin also presents to descend. Based on the analyzing of status and future trends of environment-economy coordinated development, the paper puts forward the countermeasures such as industry, structure adjustment, increasing the level of environmental protection investment, strengthening the enforcement of en vironmental policies to improve the coordinated development of environment-economy in Tianjin municipality.

Palmitate induces fat accumulation by activating C/EBPβ-mediated G0S2 expression in HepG2 cells

AIM To determine the role of G0/G1 switch gene 2(G0 S2) and its transcriptional regulation in palmitate-induced hepatic lipid *** Hep G2 cells were treated with palmitate,or palmitate in combination with CCAAT/enhancer binding protein(C/EBP)β si RNA or G0 S2 si RNA. The m RNA expression of C/EBPβ,peroxisome proliferator-activated receptor(PPAR)γ and PPARγ target genes(G0 S2,GPR81,GPR109 A and Adipoq) was examined by q PCR. The protein expression of C/EBPβ,PPARγ,and G0 S2 was determined by Western blotting. lipid accumulation was detected with Oil Red O staining and quantified by absorbance value of the extracted Oil Red O dye. lipolysis was evaluated by measuring the amount of glycerol released into the medium. RESULTS Palmitate caused a dose-dependent increase in lipid accumulation and a dose-dependent decrease in lipolysis in Hep G2 cells. In addition,palmitate increased the m RNA expression of C/EBPβ,PPARγ,and PPARγ target genes(G0 S2,GPR81,GPR109 A,and Adipoq) and the protein expression of C/EBPβ,PPARγ,and G0 S2 in a dose-dependent manner. Knockdown of C/EBPβ decreased palmitate-induced PPARγ and its target genes(G0 S2,GPR81,GPR109 A,and Adipoq) m RNA expression and palmitate-induced PPARγ and G0 S2 protein expression in Hep G2 cells. Knockdown of C/EBPβ also attenuated lipid accumulation and augmented lipolysis in palmitate-treated Hep G2 cells. G0 S2 knockdown attenuated lipid accumulation and augmented lipolysis,while G0 S2 knockdown had no effects on the m RNA expression of C/EBPβ,PPARγ,and PPARγ target genes(GPR81,GPR109 A and Adipoq) in palmitate-treated Hep G2 cells. CONCLUSION Palmitate can induce lipid accumulation in Hep G2 cells by activating C/EBPβ-mediated G0 S2 expression.

Inhibiting 5-hydroxytryptamine receptor 3 alleviates pathological changes of a mouse model of Alzheimer's disease

Extracellular amyloid beta(Aβ) plaques are main pathological feature of Alzheimer’s ***,the specific type of neuro ns that produce Aβ peptides in the initial stage of Alzheimer’s disease are *** this study,we found that 5-hydroxytryptamin receptor 3A subunit(HTR3A) was highly expressed in the brain tissue of transgenic amyloid precursor protein and presenilin-1 mice(an Alzheimer’s disease model) and patients with Alzheimer’s *** investigate whether HTR3A-positive interneurons are associated with the production of Aβ plaques,we performed double immunostaining and found that HTR3A-positive interneurons were clustered around Aβ plaques in the mouse *** amyloid precursor protein-positive or β-site amyloid precursor protein cleaving enzyme-1-positive neurites near Aβ plaques were co-localized with HTR3A *** results suggest that HTR3A-positive interneurons may partially contribute to the generation of Aβ *** treated 5.0-5.5-month-old model mice with tro pisetron,a HTR3 antagonist,for 8 consecutive *** found that the cognitive deficit of mice was partially reversed,Aβ plaques and neuroinflammation we re remarkably reduced,the expression of HTR3 was remarkably decreased and the calcineurin/nuclear factor of activated T-cell 4 signaling pathway was inhibited in treated model *** findings suggest that HTR3A interneurons partly contribute to generation of Aβ peptide at the initial stage of Alzheimer’s disease and inhibiting HTR3 partly reve rses the pathological changes of Alzheimer’s disease.

Moving object detection method based on complementary multi resolution background models

A novel moving object detection method was proposed in order to adapt the difficulties caused by intermittent object motion,thermal and dynamic background *** groups of complementary Gaussian mixture models were *** ghost and real static object could be classified by comparing the similarity of the edge images *** each group,the multi resolution Gaussian mixture models were used and dual thresholds were applied in every resolution in order to get a complete object mask without much *** computational color model was also used to depress illustration variations and light *** proposed method was verified by the public test sequences provided by the IEEE Change Detection Workshop and compared with three state-of-the-art *** results demonstrate that the proposed method is better than others for all of the evaluation parameters in intermittent object motion *** and two in the seven evaluation parameters are better than the others in thermal and dynamic background sequences,*** proposed method shows a relatively good performance,especially for the intermittent object motion sequences.

Vascular Damages in Rats Immunized by α_1-Adrenoceptor Peptides

Autoantibodies against the α1-adrenoceptor which had agonist activity as norepinephrine might play roles in the progression of hypertension, but whether the autoantibodies could induce vascular remodeling as norepinephrine is not clear. In this paper, the models with antibodies against the α1-adrenoceptor were made by immunizing Wistar rats with the synthesized the second extracellular loop of α1-adrenoceptor peptides. The homo-age male Wistar rats received BSA in the same immunizing manner and male spontaneous hypertensive rats （SHR） were used as control. All the rats were raised for one year. The blood pressure and morphological changes of arteries were measured. In the end, despite the systolic blood pressure of immunized rats had no difference with normal control, the media thickness of aortas and ratio of media to lumen in the third-order arteries of mesenteric vasculature were increased in immunized rats. The observation with electron microscope showed that the mitochondria of vascular smooth muscle cells （VSMCs） had notable hyperplasia, and the interstitial collagen fibril was increased too. The effects of purified antibodies against α1-adrenoceptor on the proliferation of cultured VSMCs, and the expressions of c-jun, c-fos and α1-adrenoceptor were detected. The results showed that the antibodies could promote the proliferation of cultured VSMCs, and enhance the expression of c-jun both in vitro and in vivo. So we concluded that antibodies against the α1-adrenoceptor could contribute to vascular damages in rats by stimulating the growth of VSMCs which might be caused by the increased c-jun expression, and might play particular roles in the pathological changes of hypertension.

Adipocytes and immune cells are vital for the development of adipose ***-pokines secreted by adipocytes regulate adipogenesis and body *** is one of the ***,the function and mechanism of chemerin in adipose tissue are not fully *** with wild type(WT)mice,Rarres2^(-/-)mice gained weight and significantly increased fat distribution in subcutaneous adipose tissue(SAT),rather than visceral adipose tissue(VAT)on high fat diet(HFD).PPARg and C/EBPa,the master regulators of adipogenesis,were up-regulated in SAT and down-regulated in VAT in Rarres2^(-/-)mice comparing with WT *** of chemerin deficiency or not,the ratio of adipocyte-progenitors to total cells and the differentiation capacity of adipocyte-progenitors were similar in SAT and VAT,but macrophage infiltration in VAT was more severe than in SAT in Rarres2^(-/-)***,CD45þimmune cells supernatant from Rarres2^(-/-)SAT pro-moted the differentiation of adipocyte-progenitors and 3T3-L1 *** array assay of CD45þimmune cells supernatant revealed that metalloproteinase inhibitor 1(TIMP1),an in-hibitor of adipogenesis,was reduced in Rarres2^(-/-)SAT,but increased in Rarres2^(-/-)*** we specifically knocked down chemerin in SAT,TIMP1 was down-regulated and adipogenesis was promoted with reducing infiltration of *** present study demonstrates that the effects of chemerin on adipose tissue is depot different,and specific knock down chemerin in SAT promote adipogenesis and improve glucose tolerance test(GTT)and insulin tolerance test(ITT).This suggests a potential therapeutic target for chemerin in the treatment of obesity related metabolic disorder.

Tunable amplified spontaneous emissions by dimensional-controlled microcrystal synthesis

The excited-state intramolecular proton transfer molecules based on chalcone （E）-3-（4＇-dimethylami- nophenyl）-l-（4＇-fluoro-2＇-hydroxyphenyl）-2-propen-l-one （DMF-HPPO） have been synthesized under microwave irradiation. One-dimensional （1D） microwires and 2D microdisks of DMF-HPPO have been selectively prepared by controlling the solution polarity. XRD results revealed the two microcrystals exhibit distinct diffraction patterns, which indicates that they have belonged to different crystalline nature. The microcrystals demonstrate shape-dependent amplified spontaneous emissions （ASE） that the emission of ID microwire is central around 618 nm and the 2D microdisk emits fluorescence at 650 nm. This result reveals the controlled synthesis of two microcrystals and their consequent multicolor amplified spontaneous emission, providing considerable promise for the development and application of new opto-electronic devices.

Calcium-handling abnormalities underlying atrial arrhythmogenesis in a Fontan operation canine model

Background Atrial tachyarrhythmia (AT) is a common complication in patients who have undergone a Fontan operation. In this study, we investigated whether abnormal Ca2+ handling contributes to the Fontan operation-related atrial arrhythmo-genic substrate. Methods Mongrel dogs were randomly assigned to sham and Fontan groups. The Fontan operation model was developed by performing an atriopulmonary anastomosis. After 14 days, an electrophysiological study was performed to evaluate the AT vulnerability. Ca2+ handling properties were measured by loading atrial cardiomyocytes (CMs) with fura-2 AM. The L-type Ca2+ (ICa?L) and Na+–Ca2+ exchanger (INCX) currents of the CMs were recorded by the whole-cell patch-clamp technique. The key Ca2+ handling proteins expression was assessed by Western blotting. Results The AT inducibility was higher in the Fontan group than in the sham group (85.71 vs. 14.29%, P litude and sarcoplasmic reticulum (SR) Ca2+ content, but in enhanced diastolic intracellular Ca2+ concentration and SR Ca2+ leak in the atrial CMs. The spontaneous CaT events, triggered ectopic activity and INCX density were increased, but ICa?L density was reduced in CMs from the Fontan atria (all Plin-dependent protein kinase II expres-sion and function were markedly enhanced in the Fontan atria. Conclusion The Fontan operation caused atrial CM Ca2+ handling abnormalities that produced arrhythmogenic-triggered activity and increased vulnerability to AT in experimental Fontan dogs.
