Correlative factors of poor prognosis and abnormal cellular immune function in patients with Alzheimer’s disease

A chiral fluorescent probe for molecular recognition of basic amino acids in solutions and cells

A novel and readily available binaphthyl-based fluorescent probe（S）-1 was designed and synthesized.（S）-1 can be used to not only chemoselectively discriminate 3 basic amino acids out of common amino acids, but also enantioselectively recognize histidine. Encouragingly, enantioselective imaging of histidine in cells was achieved for the first time by the probe（S）-1. These performances endowed it potential application in the chiral analysis of basic amino acids in asymmetric synthesis and cell imaging for diagnosis of diseases caused by racemization of histidine. Nuclear magnetic resonance（NMR） and mass spectrometry investigations suggested that different reaction extent of（S）-1 with L/D-histidine and different product structures generated the observed enantioselective fluorescent response. The molecular structures and thermodynamic stability of the complexes, formed from（S）-1 + Zn2+and enantiomers of histidine, were calculated by Gaussian 16 based on density functional theory（DFT） to validate the above action mechanism.

Neurological abnormalities identified via neuroimaging are common in patients with Alzheimer’s ***,it is not yet possible to easily detect these abnormalities using head computed tomography in the early stages of the *** this review,we evaluated the ways in which modern imaging techniques such as positron emission computed tomography,single photon emission tomography,magnetic resonance spectrum imaging,structural magnetic resonance imaging,magnetic resonance diffusion tensor imaging,magnetic resonance perfusion weighted imaging,magnetic resonance sensitive weighted imaging,and functional magnetic resonance imaging have revealed specific changes not only in brain structure,but also in brain function in Alzheimer’s disease *** reviewed literature indicated that decreased fluorodeoxyglucose metabolism in the temporal and parietal lobes of Alzheimer’s disease patients is frequently observed via positron emission computed ***,patients with Alzheimer’s disease often show a decreased N-acetylaspartic acid/creatine ratio and an increased myoinositol/creatine ratio revealed via magnetic resonance *** of the entorhinal cortex,hippocampus,and posterior cingulate gyrus can be detected early using structural magnetic resonance *** resonance sensitive weighted imaging can show small bleeds and abnormal iron ***-related functional magnetic resonance imaging can display brain function activity through cerebral blood *** functional magnetic resonance imaging can display the functional connection between brain neural *** are helpful for the differential diagnosis and experimental study of Alzheimer’s disease,and are valuable for exploring the pathogenesis of Alzheimer’s disease.

Glutamine supplementation attenuates intestinal apoptosis by inducing heat shock protein 70 in heatstroke rats

BACKGROUND:Heatstroke is the most hazardous heat-related illness and has a high fatality *** investigated whether glutamine supplementation could have a protective effect on heatstroke ***:Twenty-five 12-week-old male Wistar rats(weight 305±16 g)were randomly divided into a control group(n=5),heatstroke(HS)group(n=10),and heatstroke+glutamine(HSG)group(n=10).Seven days before heat exposure,glutamine(0.4 g/[kg·d])was administered to the rats in the HSG group by gavage every *** hours after heat exposure,serum samples were collected to detect white blood cells,coagulation indicators,blood biochemical indicators,and inflammatory cytokines in the *** small intestine tissue was stained to analyze pathological structural changes and ***,immunohistochemistry and Western blotting were used to analyze the expression levels of heat shock protein 70(HSP70).Multiple comparisons were analyzed by using one-way analysis of variance,and the Bonferroni test was conducted for the post hoc ***:After heat exposure,the core temperature of the HS group(40.65±0.31°C)was higher than the criterion of heatstroke,whereas the core temperature of the HSG group(39.45±0.14°C)was lower than the *** supplementation restored the increased white blood cells,coagulation indicators,blood biochemical indicators,and inflammatory cytokines that were induced by heatstroke to normal *** intestinal mucosa was injured,and the structure of tight junctions was damaged in the HS group;however,the structure of intestinal mucosal epithelial cells was stable in the HSG *** supplementation alleviated intestinal apoptosis and up-regulated HSP70 ***:Glutamine supplementation may alleviate intestinal apoptosis by inducing the expression of HSP70 and have a protective effect on heatstroke rats.

Downregulation of signal transduction and STAT3 expression exacerbates oxidative stress mediated by NLRP3 inflammasome

Activated nucleotide binding to the oligonucleotide receptor protein 3（NLRP3） inflammasome is possibly involved in the pathogenesis of Alzheimer＇s disease through oxidative stress and neurogenic inflammation. Low expression of the signal transducer and activator of transcription 3（STAT3） gene may promote the occurrence of neurodegenerative diseases to some extent. To clarify the roles of the NLRP3 inflammasome and STAT3 expression in oxidative stress,（1） SHSY5 Y cells were incubated with 1 mM H2 O2 to induce oxidative stress injury, and the expression of human-cell-specific signal transduction, STAT3-shRNA silencing signal transduction and STAT3 were detected. Cells were pretreated with Ca2＋ chelator BAPATA-AM（0.1 mM） for 30 minutes as a control.（2） Western blot assay was used to analyze the expression of caspase-1, NLRP3, signal transduction and STAT3. Enzyme-linked immunosorbent assay was used to analyze interleukin-1β levels. Flow cytometry was carried out to calculate the number of apoptotic cells. We found that H2 O2 treatment activated NLRP3 inflammasomes and decreased phosphorylation of signal transduction and STAT3 serine 727. BAPTA-AM pretreatment abolished the H2 O2-induced activation of NLRP3 inflammasomes, caspase-1 expression, interleukin-1β expression and apoptosis in SHSY5 Y cells, and had no effect in cells with downregulated STAT3 expression by RNAi. The findings suggest that downregulation of signal transduction and STAT3 expression may enhance the oxidative stress mediated by NLRP3, which may not depend on the Ca2^＋ signaling pathway.

Temperature Stress at Grain Filling Stage Mediates Expression of Three Isoform Genes Encoding Starch Branching Enzymes in Rice Endosperm

An early-maturity indica rice variety Zhefu 49, whose grain quality and starch structure are sensitive to environmental temperature, was subjected to different temperatures （32℃ for high temperature and 22℃ for optimum temperature） at the grain filling stage in plant growth chambers, and the different expressions of three isoform genes （SBEI, SBEIII and SBE/V） encoding starch branching enzyme （SBE） in the endosperms were studied by the real-time fluorescence quantitative PCR （FQ-PCR） method. Effects of high temperature on the SBE expression in developing rice endosperrns were isoform-dependent. High temperature significantly down-regulated the expressions of SBEI and SBEIII, while up-regulated the expression of SBEIV. Compared with SBEIV and SBEIII, the expression of SBEI gene in Zhefu 49 rice endosperms was more sensitive to temperature variation at the grain filling stage. This study indicates that changes in weather/climate conditions especially temperature stress influence rice grain formation and its quality as evidenced by isoform expression.

Training Generative Adversarial Networks with Adaptive Composite Gradient

The wide applications of Generative adversarial networks benefit from the successful training methods,guaranteeing that an object function converges to the local ***,designing an efficient and competitive training method is still a challenging task due to the cyclic behaviors of some gradient-based ways and the expensive computational cost of acquiring the Hessian *** address this problem,we proposed the Adaptive Composite Gradients(ACG)method,linearly convergent in bilinear games under suitable *** analysis and toy-function experiments both suggest that our approach alleviates the cyclic behaviors and converges faster than recently proposed SOTA *** convergence speed of the ACG is improved by 33%than other *** ACG method is a novel Semi-Gradient-Free algorithm that can reduce the computational cost of gradient and Hessian by utilizing the predictive information in future *** mixture of Gaussians experiments and real-world digital image generative experiments show that our ACG method outperforms several existing technologies,illustrating the superiority and efficacy of our method.

Stomatal regulation is critical for mangroves to survive in the hyper-saline intertidal zone where water stress is severe and water availability is highly ***,very little is known about the stomatal sensitivity to vapour pressure deficit(VPD)in mangroves,and its co-ordination with stomatal morphology and leaf hydraulic *** measured the stomatal response to a step increase in VPD in situ,stomatal anatomy,leaf hydraulic vulnerability and pressure-volume traits in nine true mangrove species of five families and collected the data of genome *** aimed to answer two questions:(1)Does stomatal morphology influence stomatal dynamics in response to a high VPD in mangroves?with a consideration of possible influence of genome size on stomatal morphology;and(2)do leaf hydraulic traits influence stomatal sensitivity to VPD in mangroves?We found that the stomata of mangrove plants were highly sensitive to a step rise in VPD and the stomatal responses were directly affected by stomatal anatomy and hydraulic ***,denser stomata was correlated with faster stomatal closure at high VPD across the species of Rhizophoraceae,and stomata size negatively and vein density positively correlated with genome *** negative leaf osmotic pressure at the full turgor(πo)was related to higher operating steady-state stomatal conductance(gs);and a higher leaf capacitance(Cleaf)and more embolism resistant leaf xylem were associated with slower stomatal responses to an increase in *** addition,stomatal responsiveness to VPD was indirectly affected by leaf morphological traits,which were affected by site salinity and consequently leaf water *** results demonstrate that mangroves display a unique relationship between genome size,stomatal size and vein packing,and that stomatal responsiveness to VPD is regulated by leaf hydraulic traits and stomatal *** work provides a quantitative framework to better understand of stomatal regulation in mangroves

Efficient and fair PPO-based integrated scheduling method for multiple tasks of SATech-01 satellite

SATech-01 is an experimental satellite for space science exploration and on-orbit demonstration of advanced *** satellite is equipped with 16 experimental payloads and supports multiple working modes to meet the observation requirements of various *** to the limitation of platform power supply and data storage systems,proposing reasonable mission planning schemes to improve scientific revenue of the payloads becomes a critical *** this article,we formulate the integrated task scheduling of SATech-01 as a multi-objective optimization problem and propose a novel Fair Integrated Scheduling with Proximal Policy Optimization(FIS-PPO)algorithm to solve *** use multiple decision heads to generate decisions for each task and design the action mask to ensure the schedule meeting the platform *** results show that FIS-PPO could push the capability of the platform to the limit and improve the overall observation efficiency by 31.5%compared to rule-based plans currently ***,fairness is considered in the reward design and our method achieves much better performance in terms of equal task *** of its low computational complexity,our task scheduling algorithm has the potential to be directly deployed on board for real-time task scheduling in future space projects.
