Lactadherin and procoagulant activities of red blood cells in cyclosporine induced thrombosis

Background The side effects of cyclosporine therapy include thromboembolic complications. However, the mechanisms underlying the hypercoagulable state induced by cyclosporine are not fully understood. Cyclosporine binds to red blood cells （RBCs） with a high affinity in circulation and alters the membranes of RBCs. Therefore, we propose that such alterations in RBCs membranes play a role in cyclosporine-induced coagulopathy and this disorder may be rectified by lactadherin, a phosphatidylserine binding protein. Methods RBCs from healthy adults were treated with various concentrations of cyclosporine. Procoagulant activity of the RBC membrane was measured by the single stage recalcification time and confirmed by detection of tenase and thrombin assembly through enzymatic assays. Inhibition assays of coagulation were carried out in the presence of lactadherin, annexin V or antitissue factor. Phosphatidylserine exposure was detected by flow cytometry and confocal microscopy through binding with fluorescein isothiocyanate （FITC）-Iabeled lactadherin as well as FITC annexin V. Results RBCs treated with cyclosporine demonstrated increased procoagulant activity. Cyclosporine treatment markedly shortened the clotting time of RBCs （（305±10） seconds vs （366±15） seconds） and increased the generation of intrinsic factor Xase （（7.68±0.99） nmol/L vs （2.86±0.11） nmol/L） and thrombin （（15.83±1.37） nmol/L vs （4.88±0.13） nmol/L）. Flow cytometry and confocal microscopy indicated that cyclosporine treatment induced an increased expression of phosphatidylserine on the RBC membrane. Lactadherin was more sensitive in detecting phosphatidylserine exposure of the RBC membrane than annexin V. The modulating effect of procoagulant activity was concomitant with and dependent on phosphatidylserine exposure. Blocking of phosphatidylserine with lactadherin effectively inhibited over 90% of FXa generation and prothrombinase activity and prolonged coagulation time. Conclusions Procoagulant properties

Aqueous zinc-ion batteries （AZIBs） are fundamentally challenged by the instability of the electrode/electrolyte interface,predominantly due to irreversible zinc （Zn） deposition and hydrogen ***,the intricate mechanisms behind the electrochemical discrepancies induced by interfacial Zn2+-solvation and deposition behavior demand comprehensive *** molecules endowed with special functional groups （such as hydroxyl,carboxyl,etc.） have the potential to significantly optimize the solvation structure of Zn2+and regulate the interfacial electric double layer （EDL）.By increasing nucleation overpotential and decreasing interfacial free energy,these functional groups facilitate a lower critical nucleation radius,thereby forming an asymptotic nucleation model to promote uniform Zn ***,this study presents a pioneering approach by introducing trace amounts of n-butanol as solvation regulators to engineer the homogenized Zn （H-Zn） anode with a uniform and dense *** interfacial reaction and structure evolution are explored by in/ex-situ experimental techniques,indicating that the H-Zn anode exhibits dendrite-free growth,no by-products,and weak hydrogen evolution,in sharp contrast to the bare ***,the H-Zn anode achieves a remarkable Zn utilization rate of approximately 20% and simultaneously sustains a prolonged cycle life exceeding 500 ***,the H-Zn//NH4V4O10（NVO） full battery showcases exceptional cycle stability,retaining 95.04%capacity retention after 400 cycles at a large current density of 5 A *** study enlightens solvation-regulated additives to develop Zn anode with superior utilization efficiency and extended operational lifespan.

Selective modification of peptides has always been a challenging issue. The selective alkylation of peptides can alter the biological activity and physical properties of molecules, and it has gained significant research interest. We herein disclose an intriguing gold-catalyzed alkylation of peptides using bench-stable unactivated alkyl bromides under photoredox catalysis. A wide range of structurally diverse primary, secondary and tertiary alkyl bromides serve as effective coupling partners to precisely connect with the α-C(sp3)–H in glycine residue of peptides. This protocol demonstrates an exceptionally broad scope for alkyl halides and excellent tolerance for a wide range of useful functional groups. A series of relay transformations to construct cyclic peptides, to proceed click reaction and to realize peptide fluorescence labeling further enhance its synthetic practicality. In addition, mechanistic studies and density functional theory(DFT) calculations reveal an inner-sphere and subsequent outersphere single electron transfer(SET) mechanism.

The Triple Ionosphere Photo Meter （TRIPM） is a far ultraviolet nadir-view photometer that is onboard the secondgeneration,polar-orbiting Chinese meteorological satellite Feng-Yun 3E （FY-3E）,which was launched on July 5,*** has three channels,and each is used to measure different types of emissions:（1） 135.6 nm airglow produced by the recombination of the O+ions and electrons in the absence of sunlight （nighttime channel）;（2） 135.6 nm airglow from the OI when sunlight is present （daytime/dusk-dawn 135.6 nm channel）;and （3） Airglow in the N2LBH of the TRIPM （daytime/dusk-dawn N2LBH channel）.FY-3E is a satellite operating in the early morning ***,the data are mostly distributed on the dawndusk side or at times when the solar zenith angle is relatively *** this study,combined with simulation calculations,the data are *** further explore the application value of the data products,a model is used to separate the ionospheric radiative recombination contribution from the thermosphere photoelectron impact excitation contribution in the data from the daytime/dusk-dawn 135.6 nm *** approach not only improves the accuracy of O/N2derived from the 135.6 nm channel and N2LBH channel but also provides the ionospheric 135.6 nm radiance at the same time as the *** results show that the 135.6 nm radiance originating from the combination of O+and electrons extracted from the measurement data exhibits similar equatorial anomaly peak structure characteristics and seasonal variations as the corresponding GPS total electron content （TEC） *** the other hand,the separated 135.6 nm radiance generated from the thermosphere photoelectron collision excitation does not contain radiative recombination contributions;thus,the ionosphere influence is greatly avoided when the O/N2is obtained.

Based on the survey and research on rural households in Changshu City and Jiangdu City of Jiangsu Province, this paper researches the microeconomic subject-commercial decision-making and production behaviours of the rural households, under the circumstance of development of agricultural product market in different regions. We conduct random sampling questionnaire survey on the rural households in 7 villages of Changshu City and Jiangdu City, Jiangsu Province, and obtain data, in order to judge and verify the theory, explore the relationship between market development of agricultural products, behaviours of rural households and income of rural households, and establish the econometric model of impact of market of agricultural products on income per capita of rural households. The research shows that there is difference in the level of socio-economic development in different regions of Jiangsu Province, and the effect of the same factors in different regions is not quite similar; the income structure of rural households and marketization development have great impact on total income per capita of rural households; the planting structure has significant impact on agricultural income per capita. Then the countermeasures and suggestions are put forward as follows: strengthen rural marketization building and broaden sales channel; reinforce information-based building and promote transparency and openness of rural market information; adjust the production structure of rural households rationally, coordinate agricultural production activities and non-agricultural activities of rural households, and balance the planting of cash crops and grain crops.

A reduction in adult neurogenesis is associated with behavioral abnormalities in patients with Alzheimer's ***,enhancing adult neurogenesis represents a promising therapeutic approach for mitigating disease symptoms and ***,nonpharmacological interventions aimed at inducing adult neurogenesis are currently *** individual non-pharmacological interventions,such as aerobic exercise,acousto-optic stimulation,and olfactory stimulation,have shown limited capacity to improve neurogenesis and cognitive function in patients with Alzheimer's disease,the therapeutic effect of a strategy that combines these interventions has not been fully *** this study,we observed an age-dependent decrease in adult neurogenesis and a concurrent increase in amyloid-beta accumulation in the hippocampus of amyloid precursor protein/presenilin 1 mice aged 2-8 *** deposition became evident at 4 months,while neurogenesis declined by 6 months,further deteriorating as the disease ***,following a 4-week multifactor stimulation protocol,which encompassed treadmill running(46 min/d,10 m/min,6 days per week),40 Hz acousto-optic stimulation(1 hour/day,6 days/week),and olfactory stimulation(1 hour/day,6 days/week),we found a significant increase in the number of newborn cells(5'-bromo-2'-deoxyuridine-positive cells),immature neurons(doublecortin-positive cells),newborn immature neurons(5'-bromo-2'-deoxyuridine-positive/doublecortin-positive cells),and newborn astrocytes(5'-bromo-2'-deoxyuridine-positive/glial fibrillary acidic protein-positive cells).Additionally,the amyloid-beta load in the hippocampus *** findings suggest that multifactor stimulation can enhance adult hippocampal neurogenesis and mitigate amyloid-beta neuropathology in amyloid precursor protein/presenilin 1 ***,cognitive abilities were improved,and depressive symptoms were alleviated in amyloid precursor protein/presenilin 1 mice
