Endoplasmic reticulum(ER) viscosity has emerged as a new potential biomarker for hepatic fibrosis(HF). Herein, we report the first ER-targeting viscosity-responsive near-infrared(NIR) fluorescent probe HBT-PP for investigations of HF *** twisted intramolecular charge transfer(TICT) and the excited state intramolecular proton transfer(ESIPT) are combined to endow HBT-PP with the selective response towards viscosity(～62-fold) and a super large stokes shift(～320 nm). HBT-PP mainly targets organelle ER and is able to image the drug-or alcohol-induced viscosity elevation in living cells. Significantly,HBT-PP was successfully applied to visualize the gender difference in HF and the protective role of estrogen in restraining HF progression. Of note, after 10 min of stain with HBT-PP, HF lesions showed a stage-based response, indicating the potential of this probe in fast and early diagnosis of HF. These results clearly demonstrated the promising application of HBT-PP as a new tool for exploring ER viscosity-involved physiological and pathological processes.

Ambient mass spectrometry imaging (MSI) enables hundreds of analytes in tissue sections to be directly mapped at atmospheric pressure with minimal sample preparation. This field is currently experiencing rapid growth, with numerous reported ambient ionization techniques resulting in a "hundred flowers bloom" situation. Nanospray desorption electrospray ionization (nano-DESI), developed by the Laskin group in 2010, is a widely used liquid-extraction-based ambient ionization technique that was first used for mass spectrometry imaging of tissue in 2012. The nano-DESI probe comprises a primary capillary and a nanospray capillary, with the latter efficiently transferring analyte-containing droplets via a tiny liquid bridge formed between the probe and sample surface, thereby enabling nanoelectrospray ionization (nano-ESI) in front of the inlet of a mass spectrometer. The advantages of nano-DESI MSI include minimal sample preparation, high spatial resolution, and high sensitivity. These features are well-suited for imaging various sample types, including frozen tissue sections, microbial communities, and environmental samples. A PubMed-database search using the "nano-DESI" keyword revealed 72 related articles in the 2010-2024 period, with 34 of them published between 2021 and 2024, which indicates that nano-DESI has rapidly developed as an ambient ionization technique over recent years. Herein, we briefly introduce key nano-DESI-MSI research progress reported in the past three years with the aim of better understanding and facilitating the use of this technology. We first discuss advances in ion-source development. Since no commercial nano-DESI source exists, designing and constructing ion sources remain technical challenges that limit its development. Lithium dendrite formation poses a significant safety hazard in lithium-ion batteries（LIBs）, severely impeding their widespread adoption and further technological advancements. Here, we present a novel quaternized polybenzimidazolium（q-PBI） to armor polyimide（PI） nanofiber separator（q-PBI@PI）, in which the q-PBI containing lithiophilicity and anionphilicity sites serves as the multifunctional nano-layers to suppress the Li dendrite and reinforce the mechanical properties of PI nanofiber ***, the q-PBI@PI separator displays an unprecedented mechanical strength of 82.8 MPa, significantly outperforming previously reported PI nanofiber membranes（typically below 50.0 MPa）. Besides, we demonstrate that the q-PBI@PI separator simultaneously possesses low nucleation overpotential（59 mV） and exceptional average coulombic efficiency due to the suppression of Li dendrite. Importantly, q-PBI@PI-based Li||Li battery enables a stable Li plating-stripping at 1.0 m A cm-2for600 h. Attributing to these exceptional merits, the armored PI separator-based LIBs display remarkable specific capability and capacity retention. The density function theory calculations reveal that q-PBI is conducive to the desolvation of Li+and can immobilize Li+and PF6-ions near affinity sites of PI nanofibers for inhibiting the formation of Li dendrites. Our work paves a new avenue for the design of next-generation high-performing separators in LIBs.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Reconstructing forest and grassland cover changes in China over the past millennium

Historical forest and grassland cover changes not only are critical indicators for quantifying ecological and environmental change processes but also serve as fundamental data for long-term climate change simulations and terrestrial ecosystem carbon emission assessments. However, because of limitations in historical data, quantitative estimations and spatially gridded reconstructions of these changes remain challenging, necessitating further methodological exploration. This study focused on China's present-day land area over the past millennium, objectively capturing the characteristics and drivers of forest and grassland cover changes. On this basis, using the forest transition theory and the space-for-time substitution method, we depicted the historical deforestation process as an inverted “S” curve and developed a model to reconstruct historical forest area changes based on the functional relationship between the forest area and population size dynamics. Subsequently, a gridded forest allocation model was established on the basis of deforestation tendencies. For the grassland cover, we implemented regionspecific methods, such as the cropland area deduction method and the habitat constraint method, to quantitatively reconstruct historical changes. Consequently, we obtained provincial forest and grassland area changes over the past millennium and mapped 10-km-resolution gridded data of forest and grassland cover. The results indicated the following.(1) The methods developed using population data as a proxy objectively reproduced the spatiotemporal evolution of forest and grassland cover in China over the past millennium. These feasible methods offer a novel pathway for the quantitative reconstruction of historical forest and grassland cover changes.(2) The data indicated that China's forest area generally decreased over the past millennium, characterized by a “decrease-then-increase” pattern. Insight into the growth behavior and electronic regulation of platinum （Pt） on various transition metal supports is paramount in developing high-performing electrocatalysts for hydrogen evolution reactions （HER）.Herein,we studied the influence of molybdenum-based supports （MoX,X=C,N,P,and S） on the growth behavior and electronic regulation of *** found that the formation energy variations between Pt single atom and clusters on MoX supports play a pivotal role in the growth behavior of ***,the electronic regulation of Pt induced by metal-support interaction may reflect the valence changes of Pt in Pt-MoX/*** Pt-MoC/C catalyst with a moderate valence state of Pt exhibits the best HER activity with an overpotential of 12.0 mV at 10 mA cm-2and a mass activity of 27.1 A mgPt-1,12.3 times as high as that of commercial 20 wt% Pt/*** work provides constructive guidance for the design of high-performance HER catalysts.

A fire control system of artillery includes a photo-tracker, a computing subsystem and a servomechanism. The photo-tracker outputs moving characteristics of the target during the tracking process, and basing upon this, the computing subsystem makes the computation and outputs shooting elements, and the servo-mechanism controls this piece of artillery to point to the foregoing position of its target in terms of the latter elements.
