Palmitoleic acid on top of HfD ameliorates insulin resistance independent of diacylglycerols and alters gut microbiota in C57BL/6j mice

With the prevalence of obesity and obesity-related metabolic syndrome,such as insulin resistance in recent years,it is urgent to explore effective interventions to prevent the progression of obesity-related metabolic *** acid is a monounsaturated fatty acid that is available from dietary sources,mainly derived from marine products.P almitoleic acid plays a positive role in maintaining glucose homeostasis and reducing ***,it is still unknow the mechanism of palmitoleic acid in ameliorating insulin ***,we investigated the effects of palmitoleic acid on chow diet(CD)-fed and high-fat diet(HfD)-fed mice,which were fed CD or HfD for 12 weeks before *** administrated mice with BSA(control),oleic acid,or palmitoleic acid for 6 weeks on top of CD or HfD *** found that palmitoleic acid only improved glucose homeostasis in HfD-fed obese mice by increasing glucose clearance and reducing *** study explored that palmitoleic acid changed the composition of gut microbiota by decreasing firmicutes population and increasing Bacteroidetes *** colon,palmitoleic acid increased intestinal tight junction integrity and reduced ***,palmitoleic acid decreased macrophage infiltration in liver and adipose tissue and increase glucose uptake in adipose ***(DAG)in tissue(for example,liver)is found to positively correlated with *** enhanced the levels of DAGs in liver but not in adipose tissue in this *** acid did not reverse the high DAG levels induced by HfD in ***,in HfD-fed mice,palmitoleic acid reduced insulin resistance by an independent-manner of *** might be associated with the beneficial effects of palmitoleic acid on altering the gut microbiota composition,improving of intestinal barrier function,and downregulating the inflammation in colon,liver,and adipose tissue.

Gamma-aminobutyric acid(GABA),widely existing in different organisms,is rapidly accumulated in plants in response to environmental *** main biosynthesis and degradation pathways of GABA constitute the GABA shunt,which is tied to the tricarboxylic acid(TCA)*** transaminase(GABA-T)and succinate semialdehyde dehydrogenase(SSADH)are two essential enzymes for the GABA degradation *** there are abundant studies on GABA shunt in higher plants at the physiological and genetic levels,research on its role in microalgae remains *** study aimed at exploring the function of GABA-T and SSADH genes in Isochrysis zhanjiangensis,an important diet microalga,under different *** cloned two GABA-T genes,IzGABA-T1 and IzGABA-T2,and one SSADH gene IzSSADH from Isochrysis zhanjiangensis and conducted heterologous expression *** results showed that the overexpression of IzGABA-T1 or IzGABA-T2 enhanced the survival rates of yeast transformants under heat or NaCl stress,while the overexpression of IzSSADH improved yeast tolerance to NaCl stress but had no obvious effect on heat ***,the results of quantitative real-time polymerase chain reaction(qPCR)showed that IzGABA-T1 transcription increased in the HT(salinity 25,35℃)and LS(salinity 15,25℃)*** 24 h,the IzGABA-T2 transcriptions increased in the HT,LS,and HS(salinity 35,25℃)groups,but their transcription levels decreased in all groups at 48 *** transcription increased in the LS *** results suggest that IzGABA-T1,IzGABA-T2,and IzSSADH are associated with temperature and salinity stresses and possess a certain preference for different stresses.

Exploring device physics of perovskite solar cell via machine learning with limited samples

Perovskite solar cells(PsCs)have developed tremendously over the past ***,the key factors influencing the power conversion efficiency(PCE)of PSCs remain incompletely understood,due to the complexity and coupling of these structural and compositional *** this research,we demon-strate an effective approach to optimize PSCs performance via machine learning(ML).To address chal-lenges posed by limited samples,we propose a feature mask(fM)method,which augments training samples through feature transformation rather than synthetic *** this approach,squeeze-and-excitation residual network(SEResNet)model achieves an accuracy with a root-mean-square-error(RMSE)of 0.833%and a Pearson's correlation coefficient(r)of ***,we employ the permu-tation importance(PI)algorithm to investigate key features for ***,we predict PCE through high-throughput screenings,in which we study the relationship between PCE and chemical *** that,we conduct experiments to validate the consistency between predicted results by ML and experimental *** this work,ML demonstrates the capability to predict device performance,extract key parameters from complex systems,and accelerate the transition from laboratory findings to commercialapplications.

Innovative Mn3-xO4-y@NCA design:Leveraging Mn/O vacancies and amorphous architecture for enhanced sodium-ion storage

Manganese-based oxide electrode materials suffer from severe jahn-Teller（j-T） distortion,leading to severe cycle instability in sodium ion ***,it is difficult to adjust the electron at d orbitals exactly to a low spin state to eliminate orbital degeneracy and suppress j-T distortion *** article constructed concentration-controllable Mn/O coupled vacancy and amorphous network in Mn3O4and coated it with nitrogen-doped carbon aerogel(Mn3-xO4-y@NCA).The existence of Mn/O vacancies has been confirmed by scanning transmission electron microscopy（STEM） and positron annihilation lifetime spectroscopy（PALS）.Atomic absorption spectroscopy（AAS） and X-ray photoelectron spectroscopy（XPS） determine the most optimal ratio of Mn/O vacancies for sodium ion storage is 1:*** functional theory（DfT） calculations prove that Mn/O coupled vacancies with the ratio of 1:2could exactly induce a low spin states and a d4electron configuration of Mn,suppressing the j-T distortion *** abundant amorphous regions can shorten the transport distance of sodium ions,increase the electrochemically active sites and improve the pseudocapacitance *** the synergetic effect of Mn/O coupled vacancies and amorphous regions,Mn3-xO4-y@NCA exhibits an energy density of 37.5 W h kg-1and an ultra-high power density of 563 W kg-1in an asymmetric *** sodium-ion batteries,it demonstrates high reversible capacity and exceptional cycling *** research presents a new method to improve the Na+storage performance in manganese-based oxide,which is expected to be generalized to other structural distortion.

Water quality and habitat drive phytoplankton taxonomic and functional group patterns in the Yangtze River

Background Although phytoplankton are important primary producers in food webs, they are relatively less studied in large rivers compared to other types of systems. To fill this research gap, we studied phytoplankton taxonomic and functional composition and their relationships with water quality, habitat, climate, and land use across 30 river sections in the middle and lower reaches of the Yangtze River during 2017–*** Major observed phytoplankton groups were cyanobacteria, bacillariophyta, and chlorophyta. Phytoplankton total abundance, total biomass, and species richness significantly decreased in the dry season compared to the wet season, with the species and functional composition differing significantly between seasons. Phytoplankton species differences between seasons were mainly contributed by Oscillatoria sp., Pseudanabaena sp., and Melosira granulata. The dfferences in phytoplankton functional groups between seasons were mainly contributed by P（including Closterium sp., Melosira sp.）, Lo（including Merismopedia sp., Peridinium sp., Ceratium sp., and Gymnodinium sp.）, and j（including Pediastrum sp., Tetraedron sp., Crucigenia sp., Scenedesmus sp., and Coelastrum sp.）. The variance partitioning showed that water quality（NO3-N, total suspended solids, turbidity） and habitat（water flow, river bank and river channel conditions） were critical factors in shaping phytoplankton patterns, followed by climate and land *** Results indicated that there was significant seasonal variation of phytoplankton in the Yangtze River, with water quality and habitat primarily driving phytoplankton patterns. Our study contributes to the understanding of natural and anthropogenic factors that drive seasonal successional processes of phytoplankton in the Yangtze River. These findings have important implications for environmental management as well as towards the ecological restoration of large rivers.

As a precise and versatile tool for genome manipulation,the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)platform holds promise for modifying fish traits of *** the aim of reducing transgene introgression and controlling reproduction,upscaled disease resistance and reproductive intervention in catfish species have been studied to lower the potential environmental risks of the introgression of escapees as transgenic *** advantage of the CRISPR/Cas9-mediated system,we succeeded in integrating the cathelicidin gene(As-Cath)from an alligator(Alligator sinensis)into the target luteinizing hormone(lh)locus of channel catfish(Ictalurus punctatus)using two delivery systems assisted by double-stranded DNA(dsDNA)and single-stranded oligodeoxynucleotides(ssODNs),*** this study,high knock in(KI)efficiency(22.38%,64/286)but low ontarget events was achieved using the ssODN strategy,whereas adopting a dsDNA as the donor template led to an efficient on-target KI(10.80%,23/213).The on-target KI of As-Cath was instrumental in establishing the lh knockout(LH^(–)_As-Cath^(+))catfish line,which displayed heightened disease resistance and reduced fecundity compared with the wild-type(WT)sibling ***,administration of human chorionic gonadotropin(HCG)and luteinizing hormone-releasing hormone analogue(LHRHa)can restore the reproduction of the transgenic fish ***,we replaced the lh gene with an alligator cathelicidin transgene and then administered hormone therapy to move towards complete reproductive control of diseaseresistant transgenic catfish in an environmentally responsible *** strategy not only effectively improves consumer-valued traits but also guards against unwanted introgression,providing a breakthrough in aquaculture genetics to confine fish reproduction and prevent the establishment of transgenic or domestic genotypes in the natural environment.

Capsaicin alleviates the hepatic clock gene disruption and gut microbiota dysbiosis in circadian rhythm disorder mouse model
