Melatonin is a pleiotropic molecule that,after a short-term sleep deprivation,promotes the proliferation of neural stem cells in the adult ***,this effect has not been observed in long-term sleep *** precise mechanism exerted by melatonin on the modulation of neural stem cells is not entirely elucidated,but evidence indicates that epigenetic regulators may be involved in this *** this study,we investigated the effect of melatonin treatment during a 96-hour sleep deprivation and analyzed the expression of epigenetic modulators predicted by computational text mining and keyword *** results showed that the administration of melatonin under sleep-deprived conditions increased the MECP2 expression and reduced the SIRT1 expression in the dentate *** observed that let-7 b,mir-132,and mir-124 were highly expressed in the dentate gyrus after melatonin administration,but they were not modified by sleep *** addition,we found more Sox2^+/5-bromo-2’-deoxyuridine(BrdU)^+cells in the subgranular zone of the sleep-deprived group treated with melatonin than in the untreated *** findings may support the notion that melatonin modifies the expression of epigenetic mediators that,in turn,regulate the proliferation of neural progenitor cells in the adult dentate gyrus under long-term sleep-deprived *** procedures performed in this study were approved by the Animal Ethics Committee of the University of Guadalajara,Mexico(approval ***-16610)on January 2,2016.

Neurodegenerative diseases are a heterogeneous group of maladies, characterized by progressive loss of neurons. These diseases involve an intricate pattern of cross-talk between different types of cells to maintain specific signaling pathways. A component of such intercellular cross-talk is the exchange of various types of extracellular vesicles (EVs). Exosomes are a subset of EVs, which are increasingly being known for the role they play in the pathogenesis and progression of neurodegenerative diseases, e.g., synucleinopathies and tauopathies. The ability of the central nervous system exosomes to cross the blood-brain barrier into blood has generated enthusiasm in their study as potential biomarkers. However, the lack of standardized, efficient, and ultra-sensitive methods for the isolation and detection of brain-derived exosomes has hampered the development of effective biomarkers. Exosomes mirror heterogeneous biological changes that occur during the progression of these incurable illnesses, potentially offering a more comprehensive outlook of neurodegenerative disease diagnosis, progression and treatment. In this review, we aim to discuss the challenges and opportunities of peripheral biofluid-based brain-exosomes in the diagnosis and biomarker discovery of Alzheimer’s and Parkinson’s diseases. In the later part, we discuss the traditional and emerging methods used for the isolation of exosomes and compare their advantages and disadvantages in clinical settings.

Morphology and viability of adult fasciola gigantica(giant liver flukes) from Philippine carabaos(Bubalus bubalis) upon in vitro exposure to lead

Objective: To evaluate the effects of lead in the morphology and viability of fasciola gigantica(f. gigantica)(giant liver fluke) isolated from infected livers of carabaos in vitro using the following concentrations of lead: 0, 100, 150 and 200 mg/L. Methods: In vitro viability and motility assay was conducted to evaluate the ef ects of lead using 1% methylene blue as the vital dye for assessment of the l ukes' viability. Results: Results indicate that f. gigantica can tolerate lead exposure as high as 200 mg/L with visible morphological variations. Upon exposure to lead, liver l ukes tend to curl and excrete black precipitates as a sign of physiological stress response. furthermore, the lethal concentration(LC50) of lead against f. gigantica in vitro was 160 mg/L. Conclusions: In conclusion, tolerance of liver flukes to high levels of lead suggests its potential as a possible biomarker of environmental pollution.

The ingest of diets with high content of fats and carbohydrates, low or no physical exercise and a stressful routine are part of the everyday lifestyle of most people in the western world. These conditions are triggers for different diseases with complex interactions between the host genetics, the metabolism, the immune system and the microbiota, including inflammatory bowel diseases(IBD), obesity and diabetes. The incidence of these disorders is growing worldwide; therefore, new strategies for its study are needed. Nowadays, the majority of researches are in use of murine models for understand the genetics, physiopathology and interaction between cells and signaling pathways to find therapeutic solutions to these diseases. The zebrafish, a little tropical water fish, shares 70% of our genes and conserves anatomic and physiological characteristics, as well as metabolical pathways, with mammals, and is rising as a new complementary model for the study of metabolic and inflammatory diseases. Its high fecundity, fast development, transparency, versatility and low cost of maintenance makes the zebrafish an interesting option for new researches. In this review, we offer a discussion of the existing genetic and induced zebrafish models of two important Western diseases that have a strong inflammatory component, the IBD and the obesity.

The Potential of Compact Libs System with Multi-Pulse Nd:YAG Laser for Bacteria Identification

This work shows the potential of a very compact, portable Laser Induced-Breakdown Spectroscopy (LIBS) system that includes low cost and compact Nd:YAG laser, intended for use in fieldwork identification of bacteria. This device allows for the generation of 2 - 4 pulses per laser shot, with a maximum energy of 120 mJ per pulse. Each pulse has a duration of 60 ns, and the total duration of the train of pulses is 180 μs. The device dimensions including the power supply are 50 × 20 × 140 cm, which indicates that can be utilized as a portable LIBS system in field conditions. The spectra captured on two different bacteria shows the capability of identification.
