Monogenic f.atures of.urolithiasis: A comprehensive review

Objective: Urolithiasis f.rmation has been attributed to environmental and dietary f.ctors. However, evidence is accumulating that genetic background can contribute to urolithiasis f.rmation. Advancements in the identif.cation of.monogenic causes using high-throughput sequencing technologies have shown that urolithiasis has a strong heritable ***: This review describes monogenic f.ctors implicated in a genetic predisposition to urolithiasis. Peer-reviewed journals were evaluated by a PubMed search until July 2023 to summarize disorders associated with monogenic traits, and discuss clinical implications of.identif.cation of.patients genetically susceptible to urolithiasis ***: Given that more than 80% of.urolithiases cases are associated with calcium accumulation, studies have f.cused mainly on monogenetic contributors to hypercalciuric urolithiases, leading to the identif.cation of.receptors, channels, and transporters involved in the regulation of.calcium renal tubular reabsorption. Nevertheless, available candidate genes and linkage methods have a low resolution f.r evaluation of.the ef.ects of.genetic components versus those of.environmental, dietary, and hormonal f.ctors, and genotypes remain undetermined in the majority of.urolithiasis ***: The pathophysiology underlying urolithiasis f.rmation is complex and multif.ctorial, but evidence strongly suggests the existence of.numerous monogenic causes of.urolithiasis in humans.

Background:A multitude of.mouse models are utilized to emulate and study intestinal inf.ammation.T-cell receptor alpha chain(TCRα)-def.cient mice are used as a model of.spontaneous colitis that has similarities with human ulcerative ***,colitis is triggered late in the lif. of.the mouse(age:4-5 months),and inf.ammation does not develop at the same time in dif.erent mice.A previously conducted study reported that the administration of.the drug piroxicam triggered predictable and early colitis in TCRα-def.cient mice at the age of.6-8 ***,a detailed characterization of.ensuing inf.ammation was not ***:We conducted an in-depth examination of.piroxicam-triggered colitis in TCRα-def.cient mice,with emphasis on spatial histopathologic changes and analysis of.expression of.inf.ammatory ***,we tested amelioration of.colitis with ***:We conf.rmed that piroxicam induced a time-prescribed colitis and did so in the proximal colon as well as the cecum of.TCRα-def.cient *** adminis-tration was observed to induce epithelial hyperplasia,goblet cell loss,and leukocyte inf.ltration with occasional ulceration.A Swiss roll technique was used to examine the colon and cecum in its ***,we observed that inf.ammation was mul-tif.cal segmental,with areas of.tissue damage in between healthy *** addition,we observed variability in the severity of.inf.ammation among replicate animals and treatments,and that the administration of.dexamethasone only partially ameliorated inf.ammation in the proximal ***:Piroxicam consistently induced multif.cal segmental colitis in the proxi-mal colon and cecum,although the degree of.inf.ammation was reduced in the ***,spatial variability in inf.ammation in the large intestine and the inter-replicate variation in the severity of.inf.ammation must be taken into consideration when utilizing this murine model of.synchronized colitis.

Background: Soybean meal is an excellent and cost-ef.ective protein source; however, its usage is limited in the piglet due to the presence of.anti-nutritional f.ctors and the antigens glycinin and β-conglycinin. The objective of.the current study was to screen and select f.r bacteria that can be ef.iciently adopted to f.rment soybean meal in order to solve this ***: Bacteria were isolated f.om f.rmented soy f.ods and the grass carp intestine, and strains selected f.r high protease, cellulase and amylase activities. The isolated bacteria were characterized as Bacillus cereus, Bacillus subtilis and Bacilus amyloliquef.cien, respectively. f.rmentation with f.od-derived Isolate-2 and f.sh-derived f.9 increased crude protein content by 5.32% and 8.27%, respectively; improved the amino acid prof.le by increasing certain essential amino acids, broke down larger soy protein to 35 k Da and under, eliminated antigenicity against glycinin and β-conglycinin, and removed raf.inose and stachyose in the soybean meal f.llowing a 24-h ***: Our results suggest these two B. amyloliquef.ciens bacteria can ef.iciently solid state f.rment soybean meal and ultimately produce a more utilizable f.od source f.r growing healthy piglets.

Tumor metastases present an of.en dif.icult to treat therapeutic challenge f.r cancer patients and their *** recent paper by the Kaplan group(Kaczanowska et al.)exploited the natural capacity of.monocytes or a subset of.monocytes to migrate to tumor sites and pre-metastatic niches to deliver IL-12 in a novel anticancer approach designed to stimulate local immune responses.1 This raises the possibility to simultaneously f.ght cancer on at least two critical f.onts,(1)elimination of.the primary tumor and(2)inhibition of.newly developing pre-metastatic lesions.

Insights into erlotinib action in pancreatic cancer cells using a combined experimental and mathematical approach

AIM:To gain insights into the molecular action of.erlotinib in pancreatic cancer (PC) cells. METHODS:Two PC cell lines, BxPC-3 and Capan-1, were treated with various concentrations of.erlotinib, the specif.c mitogen-activated protein kinase kinase (MEK) inhibitor U0126, and protein kinase B (AKT) inhibitor XIV. DNA synthesis was measured by 5-bromo-2'-deoxyuridine (BrdU) assays. Expression and phosphorylation of.the epidermal growth f.ctor receptor (EGf.) and downstream signaling molecules were quantif.ed by Western blot analysis. The data were processed to calibrate a mathematical model, based on ordinary dif.erential equations, describing the EGf.mediated signal transduction. RESULTS:Erlotinib signif.cantly inhibited BrdU incorporation in BxPC-3 cells at a concentration of.1 mol/L, whereas Capan-1 cells were much more resistant. In both cell lines, MEK inhibitor U0126 and erlotinib attenuated DNA synthesis in a cumulative manner, whereas the AKT pathway-specif.c inhibitor did not enhance the ef.ects of.erlotinib. While basal phosphorylation of.EGf. and extracellular signal-regulated kinase (ERK) did not dif.er much between the two cell lines, BxPC-3 cells displayed a more than f.ve-times higher basal phospho-AKT level than Capan-1 cells. Epidermal growth f.ctor (EGf. at 10 ng/mL induced the phosphorylation of.EGf., AKT and ERK in both cell lines with similar kinetics. In BxPC-3 cells, higher levels of.phospho-AKT and phospho-ERK (normalized to the total protein levels) were observed. Independent of.the cell line, erlotinib ef.iciently inhibited phosphorylation of.EGf., AKT and ERK. The mathematical model successf.lly simulated the experimental f.ndings and provided predictions regarding phosphoprotein levels that could be verif.ed experimentally. CONCLUSION:Our data suggest basal AKT phosphorylation and the degree of.EGf.induced activation of.AKT and ERK as molecular determinants of.erlotinib ef.iciency in PC cells.

All colorectal surgeons are f.ced f.om time to time with anastomotic leakage af.er colorectal surgery. This complication has been studied extensively without a signif.cant reduction of.incidence over the last 30 years. New techniques of.prevention, by innovative anastomotic techniques should improve results in the f.ture, but standardization and "teachability" should be guaranteed. Risk scoring enables intra-operative decision-making whether to restore continuity or deviate. Early detection can lead to reduction in delay of.diagnosis as long as a standard system is used. f.r treatment options, no f.rm evidence is available, but f.ture studies could f.cus on repair and saving of.the anastomosis on the one hand or anastomotical breakdown and def.nitive colostomy on the other hand.
