Glycoprotein fucosylation is increased in seminal plasma of subfertile men

Fucose, the monosaccharide frequent in N- and O-glycans, is a part of Lewis-type antigens that are known to mediate direct sperm binding to the zona pellucida. Such interaction was found to be inhibited in vitro by fucose-containing oligo- and polysaccharides, as well as neoglycoproteins. The objective of this study was to screen seminal plasma proteins of infertile/subfertile men for the content and density of fucosylated glycoepitopes, and compare them to samples of fertile normozoospermic subjects. Seminal proteins were separated in polyacrylamide gel electrophoresis and blotted onto nitrocellulose membrane and probed with fucose-specific Aleuria aurantia lectin （AAL）. Twelve electrophoretic bands were selected for quantitative densitometric analysis. It was found that the content, and especially the density of fucosylated glycans, were higher in glycoproteins present in seminal plasma of subfertile men. No profound differences in fucosylation density were found among the groups of normozoospermic, oligozoospermic, asthenozoospermic, and oligoasthenozoospermic subfertile men. According to the antibody probing, AAL-reactive bands can be attributed to male reproductive tract glycoproteins, including prostate-specific antigen, prostatic acid phosphatase, glycodelin and chorionic gonadotropin. Fibronectin, a1-acid glycoprotein, a1-antitrypsin, immunoglobulin G and antithrombin III may also contribute to this high fucosylation. It is suggested that the abundant fucosylated glycans in the sperm environment could interfere with the sperm surface and disturb the normal course of the fertilization cascade.

Polarization effect in tip-enhanced infrared nanospect- roscopy studies of the selective Y5 receptor antagonist Lu AA33810

A novel approach of combining conventional infrared spectroscopy （IR） and atomic force microscopy （AFm） is presented to better understand the behavior of a drug adsorbed on a metal substrate at the nanoscale level. Tip-enhanced infrared nanospectroscopy （TEIRA） was used for the first time to investigate Lu AA33810, a selective brain-penetrating Y5 receptor antagonist, after immobilization on gold nanopartides （GNPs）. Here, a gold coated AFm tip and gold substrate were used to obtain the near-field electromagnetic field trapping effect. Because of the huge signal enhancement, it was possible to obtain the spectral information regarding the self-assembled monolayer of the investigated molecule. The effect of two orthogonal polarizations （p- and s-polarization modulations） of the excitation laser beam on the spectral patterns is also discussed. The results show that there is a strong relationship between the state of polarization of the incident radiation and the relative infrared band intensities. Another factor affecting the observed spectral differences is the topology of the metal substrate, which may result in the induction of a cross-polarization effect. The performed analysis indicates that the C--C bond from the cyclohexyl group is oriented almost parallel to the metal surface. Conversely, the p- and s-polarized spectral variations suggest that the O=S---O angle is high enough to enable the simultaneous interaction of both oxygen atoms with the GNPs.

Chemically binding Eu^3+ onto CdS semiconductor nanoparticle surface

Europium ions were chemically bound to CdS nanoparticles surface by diethylenetri-aminepentaacetate （DTPA, 1） in a two-step synthetic route. First 1 was applied to chelate with cadmium on the surface of cadmium-rich CdS nanoparticles and act as a capping agent. Further, the purified 1-capped particles were used to bind with Eu^3＋. The purified and redispersed particles were characterized by photoluminescence spectroscopy, TEm and SEm. It was observed that Eu^3＋ on the nanoparticle surface significantly increased the band gap emission and decreased the surface emission intensity of the CdS nanoparticles.

Decreased serum prohepcidin concentration in patients with polycythemia vera

Objective： Iron deficiency is a common complication in patients with polycythemia vera （PV）. Hepcidin is a principal regulator of iron homeostasis. The aim of our study was to assess prohepcidin, a hepcidin precursor, and other iron status pa- rameters in the serum of PV patients. methods： The study was performed in 60 patients （F/m 26/34） aged 38-84 （66＋ 10） years. The control group consisted of 20 healthy volunteers, age and sex matched. The following parameters were determined in blood serum samples： prohepcidin concentration, iron content, unsaturated iron binding capacity （UIBC）, total iron binding capacity （TIBC）, transferrin saturation （TfS）, and concentrations of ferritin and soluble transferrin receptor （sTfR）. Results： All PV patients showed significantly lower levels of prohepcidin, higher levels of sTIR and TIBC compared to the control group. 40% of the patients from the study group showed concentrations of ferritin below the normal range and significantly lower levels of serum iron and TfS, and significantly higher levels of sTIR, UIBC and TIBC in comparison with the rest of the study group. In this group of patients, prohepcidin concentrations were significantly lower than those in other patients. Conclusion： The results indicate that PV patients suffer from iron metabolism disorders. The decreased serum level ofprohepcidin in PV patients may be a result of iron deficiency.

Fine root morphology and soil properties under influence of different tree stands along an altitudinal climosequence in the Carpathian mountains

In an era of climate change, understanding the factors that impact root systems can improve our understanding of carbon cycling in forest ecosystems. The study objective was to determine the impact of climatic conditions on the biomass and morphology of roots of different tree species along an elevation gradient, and consequently on properties of montane forest soils. The study plots were established at three different elevations(600, 800 and1000 m a.s.l.) along a slope with an inclination of 15°. The research plots were located in a beech stand(Fagus sylvatica L.) and fir stand(Abies alba mill.). Soil samples were collected from each study plot, for which basic physical and chemical properties were determined. Additionally, we determined the morphology, production and decomposition rate of fine roots. Our analyses confirmed the significance of climatic conditions in the formation of soil properties, in particular the amount of accumulated carbon and nitrogen content. A decrease of root biomass and reduced root growth were recorded with increasing elevation. The characteristics of roots were linked with the properties of the studied mountain soils, in particular p H, alkaline cation content and content of selected micronutrients. Limitation of root growth in higher elevations affected both study species. Additional research into the formation of tree root morphology is needed, especially in mountainous regions where changes may occur more dynamically. This will provide a better understanding of how stands can cope with different types of environmental stress.

Deadwood affects the soil organic matter fractions and enzyme activity of soils in altitude gradient of temperate forests

The main objective of our study has been to determine the role of deadwood in the shaping of the amount of soil organic matter fractions in mountain forest *** this purpose,a climosequence approach comprising north(N)and south(S)exposure along the altitudinal gradient(600,800,1000 and 1200 m a.s.l.)was set *** comparing the properties of decomposing deadwood and those of the soils located directly beneath the decaying wood we drew conclusions about the role of deadwood in the shaping of soil organic matter fractions and soil carbon storage in different climate *** basic properties,enzymatic activity and fractions of soil organic matter(SOm)were determined in deadwood and affected directly by the components released from decaying *** decomposed deadwood impacts soil organic matter stabilization more strongly than the less decayed deadwood and the light fraction of SOm is more sensitive to deadwood effects than the heavy fraction regardless of the location in the altitude *** in SOm mineral-associated fraction C content is more pronounced in soils under the influence of deadwood located in lower locations of warmer *** released from decaying wood stimulate the enzymatic activity of soils that are within the range of deadwood influence.

Spatiotemporal trends of Pm_(2.5) and its major chemical components at urban sites in Canada

To evaluate the effectiveness of emission control regulations designed for reducing air pollution, chemically resolved Pm_(2.5) data have been collected across Canada through the National Air Pollution Surveillance network in the past decade. 24-hr time integrated Pm_(2.5) collected at seven urban and two rural sites during 2010-2016 were analyzed to characterize geographical and seasonal patterns and associated potential causes. Site-specific seven-year mean gravimetric Pm_(2.5) mass concentrations ranged from 5.7 to 9.6 μg/m^(3). Seven-year mean concentrations of SO_(4)^(2-), NO_(3)^(-), NH_(4)^(+), organic carbon(OC), and elemental carbon(EC) were in the range of 0.68 to 1.6, 0.21 to 1.5, 0.27 to 0.71, 1.1 to 1.9, and 0.37 to 0.71 μg/m^(3), accounting for 10.8%-18.1%, 3.7%-16.7%, 4.7%-7.4%, 18.4%-21.0%, and 6.4%-10.6%, respectively, of gravimetric Pm_(2.5) mass. Pm_(2.5) and its five major chemical components showed higher concentrations in southeastern Canada and lower values in Atlantic Canada, with the seven-year mean ratios between the two regions being on the order of 1.7 for Pm_(2.5) and 1.8-7.1 for its chemical components. When comparing the concentrations between urban and rural sites within the same region, those of SO_(4)^(2-) and NH_(4)^(+) were comparable, while those of NO_(3)^(-), OC, and EC were around 20%, 40%-50%, and 70%-80%, respectively, higher at urban than rural sites, indicating the regional scale impacts of SO_(4)^(2-) and NH_(4)^(+) and effects of local sources on OC and EC. monthly variations generally showed summertime peaks for SO_(4)^(2-) and wintertime peaks for NO_(3)^(-), but those of NH_(4)^(+), OC, and EC exhibited different seasonality at different locations.
