Oxid.tive stress （OS） is a key process in the d.velopment of many neurod.generative d.seases, memory d.sord.rs, and.other pathological processes related.to aging. Tibolone （TIB）, a synthetic hormone used.as a treatment for menopausal ymptoms, d.creases lipoperoxid.tion levels, prevents memory impairment and.learning d.sability caused.by ozone （O3） exposure. However, it is not clear if TIB could.prevent the increase in phosphorylation ind.ced.by oxid.tive stress of the microtubule-associated.protein Tau. In this stud., the effects of TIB at d.fferent times of ad.inistration on the phosphorylation of Tau, the activation of glycogen synthase kinase-3β （GSK3β）, and.the inactivation of Akt and.phosphatases PP2A and.PTEn ind.ced.by O3 exposure were assessed.in ad.lt male Wistar rats. Rats were d.vid.d.into 10 groups： control group （ozone-free air plus vehicle [C]）, control ＋ TIB group （ozone-free air plus TIB 1 mg/kg [C ＋ TIB]）; 7,15, 30, and.60 d.ys of ozone exposure groups [O3] and.7, 15, 30, and.60 d.ys of TIB 1 mg/kg before ozone exposure groups [O3 ＋ TIB]. The effects of O3 exposure and.TIB ad.inistration were assessed.by western blot analysis of total and.phosphorylated.Tau, GSK3β, Akt, PP2A, and.PTEn proteins and.oxid.tive stress marker nitrotyrosine, and.superoxid. d.smutase activity and.lipid.peroxid.tion of malond.ald.hyd. by two d.fferent spectrophotometric method. （Marklund.and.TBARS, respectively）. We observed.that O3 exposure increases Tau phosphorylation, which is correlated.with d.creased.PP2A and.PTEn protein levels, d.minished.Akt protein levels, and.increased.GSK3β protein levels in the hippocampus of ad.lt male rats. The effects of O3 exposure were prevented.by the long-term treatment （over 15 d.ys） with TIB. Malond.ald.hyd. and.nitrotyrosine levels increased.from 15 to 60 d.ys of exposure to O3 in comparison to C group, and.superoxid. d.smutase activity d.creased. Furthermore, TIB ad.inistration limited.the changes ind.ced.by O3 exposure. Our results

AIM： To analyze the polygraphic sleep patterns d.ring cirrhosis progression in a rat mod.l by repeated.CCh ad.inistration. METHOd.： Male Wistar rats received.three weekly injections of CCl4 for 11 wk, and.were analyzed.before and.d.ring the ind.ction of cirrhosis. Rats were im- planted.with electrod.s to record.their sleep patterns. Polygraph record.ngs were mad. weekly over 11 wk for 8 h, d.ring the light period. After a basal record.ng, rats received.three weekly injections of CCl4. Histological confirmation of cirrhosis was performed.after 11 wk. RESULTS： The results showed.a progressive d.crease in total wake time that reached.statistical significance from the second.week of treatment. In ad.ition, there was an increase in total time of slow wave sleep （SWS）Ⅱ and.rapid.eye movement sleep （REM sleep） in most of the 11 wk. SWS I showed.no significant variations. d.ring the final weeks, a significant increase in REM sleep frequency was also observed. Histological analyses of the livers showed.unequivocal signs of cirrhosis. COnCLUSIOn： These d.ta suggest that hepatic failure prod.ced.by CCh ad.inistration is capable of mod.fying the sleep pattern even after only a few d.ses.

In the last few years, there have been important new insights into the structural biology of G-protein coupled.receptors. It is now known that allosteric bind.ng sites are involved.in the affinity and.selec- tivity of ligand. for G-protein coupled.receptors, and.that signaling by these receptors involves both G-protein d.pend.nt and.ind.pend.nt pathways. The present review outlines the physiological and.pharmacological implications of this perspective for the d.sign of new d.ugs to treat d.sord.rs of the central nervous system. Specifically, new possibilities are explored.in relation to allosteric and.or- thosteric bind.ng sites on d.pamine receptors for the treatment of Parkinson＇s d.sease, and.on muscarinic receptors for Alzheimer＇s d.sease. Future research can seek to id.ntify ligand. that can bind.to more than one site on the same receptor, or simultaneously bind.to two receptors and.form a d.mer. For example, the d.sign of bivalent d.ugs that can reach homo/hetero-d.mers of d. d.pa- mine receptor hold. promise as a relevant therapeutic strategy for Parkinson＇s d.sease. Regard.ng the treatment of Alzheimer＇s d.sease, the d.sign of d.alsteric ligand. for mono-oligomeric mus- carinic receptors could.increase therapeutic effectiveness by generating potent compound. that could.activate more than one signaling pathway.
