Cementum is the outer-, mineralized-tissue covering the tooth root and an essential part of the system of periodontal tissue that anchors the tooth to the bone. Periodontal disease results from the destructive behavior of the host elicited by an infectious biofilm adhering to the tooth root and left untreated, may lead to tooth loss. We describe a novel protocol for identifying peptide sequences from native proteins with the potential to repair damaged dental tissues by controlling hydroxyapatite biomineralization. Using amelogenin as a case study and a bioinformatics scoring matrix, we identified regions within amelogenin that are shared with a set of hydroxyapatite-binding peptides （HABPs） previously selected by phage display. One 22-amino acid long peptide regions referred to as amelogenin-derived peptide 5 （ADP5） was shown to facilitate cell-free formation of a cementum-like hydroxyapatite mineral layer on demineralized human root dentin that, in turn, supported attachment of periodontal ligament cells in vitro. Our findings have several implications in peptide-assisted mineral formation that mimic biomineralization. By further elaborating the mechanism for protein control over the biomineral formed, we afford new insights into the evolution of protein-mineral interactions. By exploiting small peptide domains of native proteins, our understanding of structure-function relationships of biomineralizing proteins can be extended and these peptides can be utilized to engineer mineral formation. Finally, the cementomimetic layer formed by ADP5 has the potential clinical application to repair diseased root surfaces so as to promote the regeneration of periodontal tissues and thereby reduce the morbiditv associated with tooth loss.

During the Late Cretaceous in the eastern Mediterranean, the northern branch of the southern Neotethys was closed by multiple northward subductions. Of these, the most northerly located subduction created the Baskil continental arc at around 82–84 Ma. The more southerly and intra-oceanic subduction, on the other hand, produced an arc-basin system,the Yüksekova Complex, as early as the late Cenomanian–early Turonian. The abundant and relatively well-studied basaltic rocks of this complex were intruded by dykes, sills and small stocks of felsic–intermediate rocks, not previously studied in detail. The intrusives collected from five different localities in the elazig region of eastern Turkey are all subalkaline, with low Nb/Y values. Most of them have been chemically classified as rhyodacites/dacites, whereas a small number appear to be andesites. In normal mid-ocean-ridge basalt(N-MORB)-normalised plots, the intrusives are characterised by relative enrichments in Th and La over Nb, Zr, Hf, Ti and high field strength elements(HRees), indicating their derivation from a subduction-modified source. While their relatively high, positive εN d(i) values(+6.4 and +7.2) might suggest a depleted mantle source for their ultimate origin, somewhat radiogenic Pb values indicate a sedimentary contribution to the source of the rocks. The overall geochemical characteristics indicate their generation in an oceanic arc setting. The zircon U-Pb Laser ablation-inductively coupled plasma-mass spectrometry(LA-ICP-MS) data obtained from five felsic-intermediate rock samples yielded intrusion dates of 80–88 Ma. This suggests that the elazig oceanic arc-related intrusives are slightly younger than those of the Yüksekova arc-basin system, but coeval with the Baskil continental arc. However, the felsic–intermediate intrusives show different geochemical characteristics(oceanic arc-type, with a lack of crustal contamination)to those of the Baskil continental arc. A sediment sample was collected from a deep-sea hydrothermal vent field located at a depth of 2 951 m on the Southwest Indian Ridge. Phylogenetic analyses were performed on the prokaryotic community using polymerase chain reaction（PCR） amplification of the 16 S rRNA and nifH genes. Within the Archaea, the dominant clones were from marine benthic group e（MBGe） and marine group I（MGI） belonging to the phyla euryarchaeota and Thaumarchaeota, respectively. More than half of the bacterial clones belonged to the Proteobacteria, and most fell within the Gammaproteobacteria. No epsilonproteobacterial sequence was observed. Additional phyla were detected including the Actinobacteria, Bacteroidetes, Planctomycetes, Acidobacteria, Nitrospirae, Chloroflexi, Chlorobi, Chlamydiae, Verrucomicrobia, and candidate divisions OD1, OP11, WS3 and TM6, confirming their existence in hydrothermal vent environments. The detection of nifH gene suggests that biological nitrogen fixation may occur in the hydrothermal vent field of the Southwest Indian Ridge. Phylogenetic analysis indicated that only Clusters I and III NifH were present. This is consistent with the phylogenetic analysis of the microbial 16 S rRNA genes, indicating that Bacteria play the main role in nitrogen fixation in this hydrothermal vent environment.

Plant mitochondrial genomes （mtDNAs） are large and undergo frequent recombination events. A common phenotype that emerges as a consequence of altered mtDNA structure is cytoplasmic-male sterility （CMS）. The molecular basis for CMS remains unclear, but it seems logical that altered respiration activities would result in reduced pollen production. Analysis of tobacco （Nicotiana tabacum） mtDNAs indicated that CMS-associated loci often contain fragments of known organellar genes. These may assemble with organellar complexes and thereby interfere with normal respiratory functions. Here, we analyzed whether the expression of truncated fragments of mitochondrial genes （i.e. atp4, cox1 and rps3） may induce male sterility by limiting the biogenesis of the respiratory machinery, cDNA fragments corresponding to atp4f, coxlfand rps3f were cloned in-frame to a mitochondrial localization signal and a C-termini HA-tag under a tapetum-specific promoter and introduced to tobacco plants by Agrobacterium-mediated transformation. The constructs were then analyzed for their effect on mitochondrial activity and pollen fertility. Atp4f, Coxlf and Rps3f plants demonstrated male sterility phenotypes, which were tightly correlated with the expression of the recombinant fragments in the floral meristem. Fractionation of native organellar extracts showed that the recombinant ATP4f-HA, COXlf-HA and RPS3f-HA proteins are found in large membrane-associated particles. Analysis of the respiratory activities and protein profiles indicated that organellar complex I was altered in Atp4f, Coxlfand Rps3f plants.

AIM: To examine whether hepatitis C virus (HCV)-infected patients who carry hypercoagulable mutationssuffer from increased rates of liver fi brosis. MeTHODS: We analyzed DNA samples of 168 HCV patients for three common hypercoagulable gene mutations: prothrombin 20210 (PT20210), factor V Leiden (FV Leiden) and methylene tetrahydrofolate reductase (MTHFR). The patients were consecutively recruited as part of the prospective "Fibroscore Study" in France. The effect of the various mutations on the rate of fi-brosis was analyzed statistically and was correlated with epidemiological, clinical and biochemical data such as grade and stage of liver biopsies, patients' risk factors for liver cirrhosis, and timing of infection. ReSULTS: Fifty two of the patients were categorized as "fast fi brosers" and 116 as "slow fi brosers"; 13% of the "fast fi brosers" carried the PT20210 mutation as compared with 5.5% of the "slow fi brosers", with an odds ratio of 4.76 (P = 0.033; 95% CI: 1.13-19.99) for "fast" liver fibrosis. Carriage of MTHFR or FV Leiden mutations was not associated with enhanced liver fi brosis. CONCLUSION: Carriage of the PT20210 mutation is related to an increased rate of liver fi brosis in HCV patients.

Changes in Body Mass Index after Arthroscopic Surgery and Its effect on Functional Outcomes

Objective: Arthroscopic surgery is one of the most common surgical procedures in orthopedic surgery. Post-surgery weight gain is one of the most important factors that affects the functional outcomes of arthroscopic surgery. This study investigated the effect of changes in the body mass index (BMI) in the period after arthroscopic surgery and the potential impact of these changes on functional outcomes. Methods: One hundred-sixty four patients (knee and hip arthroscopy patients) were evaluated retrospectively. Changes in the BMI and functional scores of the patients between the preoperative period and first, third, and sixth months postoperatively were compared. Results: The mean age of the patients was 36.03 (15 - 64) years, and the mean follow-up was 14.2 months (6 - 29) months. Postoperative increases in the BMI values in the first, third, and sixth months were statistically significant in the three groups of patients (i.e., meniscal pathology, ligament pathology, and intra-articular hip pathology) (p p = 0.564), meniscal pathology (p = 0.854), and ligament pathology groups (p = 0.908) were not statistically significant. Conclusion: Patients may gain weight following arthroscopic surgery of the knee and hip. Orthopedic surgeons should keep this issue in mind and educate patients about how to prevent potential weight gain to ensure it does not adversely affect functional outcomes.

In assessing the relationship between calcium supplementation and maintaining bone mass or reducing the risk of fracture, the effectiveness of calcium supplementation has never been decisive. Freshwater crayfish rely on amorphous calcium carbonate (ACC), an instable polymorph of calcium carbonate, as the main mineral in the exoskeleton and in the temporary storage organ, the gastrolith. Inspired by the crayfish model, we have previously shown an increase in calcium bioavailability in rats administered with synthetic stable ACC vs. crystalline calcium carbonate (CCC). The current study compared the effects of amorphous calcium derived from either gastrolith or synthetic ACC with those of crystalline calcium, found in commercial CCC or calciumcitrate supplements, in a bone loss prevention model. Rats were subjected to either sham or ovariectomy (OVX) operation (n^20/ group) followed by administration of food pellets supplemented with 0.5% calcium from either source over 12 weeks. Micro-computed tomography (μCT) and histomorphometric analyses revealed bone loss prevention by both gastrolith and ACC treatments, manifested by an increase in morphometric bone parameters, compared to both CCC-?and calcium citrate-treated groups. Both gastrolith and ACC treatments resulted in bone formation in the tibia cancellous bone, indicated by dynamic histomorphometry parameters, compared to either the CCC or calcium citrate treatments. Levels of urine deoxypyridinoline (DPD), suggested an anti-resorptive effect of ACC, which was also the only treatment that led to a significant increase in vertebral mechanical strength, as supported by μCT analysis of topology and orientation parameters of the vertebral trabeculae. To our knowledge, such levels of bone loss prevention by calcium supplements have never been reported.
