Bone blood flow is influenced by muscle contractions

Forces acting on the skeleton could be divided into those originating from gravitational loading and those originating from muscle loading. Flat bones in a non-weight-baring segment of the skeleton probably experience forces mostly generated by muscle contractions. One purpose of muscle contractions is to generate blood flow within skeletal tissues. The present study aimed to investigate the pulsatile patellar bone blood flow after low and high intensity leg extension exercises. Forty-two healthy individuals volunteered for the study. Dynamic isotonic one leg extension/flexion exercises were performed in a leg extension machine. Randomly, the exercises were performed with the left or right leg with either 10 repetition maximum (10 RM) continuously without any resting periods (high intensity muscle work), or 20 RM with a 2 second rest between contractions (low intensity muscle work). The work load, expressed in kilograms totally lifted, was identical in both legs. The pulsatile patellar blood flow was recorded continuously using a photoplethysmographic technique. Blood pressure was measured continuously during muscle work by a non-invasive method (Finapress). The patellar pulsatile bone blood flow increased significantly more after high intensity muscle work (61%) compared to the same work load performed using a lower intensity (22%), p = 0.000073. Systolic blood pressure changed equally during and after both interventions. post-exercise bone hyperaemia appears to be correlated to the intensity of muscle contractions in the muscle compartment attached to the bone.

The Cambrian explosion was coined to describe the geologically sudden appearance of numerous bilaterian body plans(phyla)around the Ediacaran-Cambrian transition,around 565-520million years *** explanations and conjectures have been postulated in order to explain the pattern and duration of this explosive radiation of many different phyla of early ***,we focus on the evolution of a phylum of marine suspension-feeding animals—the brachiopods,as exemplified by the exceptionally preserved taxa from the celebrated Chengjiang Konservat Lagerst(a|¨)tte(Yunnan,China).The abundant soft-bodied preservation at these fossil quarries gives us the only firm insights into what brachiopods looked like and how they functioned and lived when they first appeared on the *** of Chengjiang brachiopods demonstrate that the early animals developed a remarkably varied organization of tissues and organs shortly after the onset of Cambrian *** the marine suspension-feeding brachiopods,most importantly the tentaculate feeding structure of early brachiopods is already differentiated into two shapes of lophophore,anteriorly coiled(spiralled)and posteriorly arching tentacle crowns and the unique latter type was previously not documented from fossil and living *** unlike any known Recent brachiopod,all the known Cambrian brachiopods from Chengjiang have an open digestive tract that was disposed either as a Ushaped gut in linguliform and stem group brachiopods,or straight gut with a posterior anus in some calcareous-shelled ***,in contrast toliving lingulids,all the Cambrian brachiopods have an epibenthic lifestyle either cemented by a ventral valve or attached by variable pedicles to establish complex ecological community encompassing primary tierers and variable secondary *** is therefore assumed that brachiopods were the first benthic metazoan that achieved their success in ecological stratification and tiering complexity by lat

Background： In this study the physiological implications of artificial rearing were investigated. Low（LBW） and normal birth weight（NBW） piglets were compared as they might react differently to stressors caused by artificial rearing. In total, 42 pairs of LBW and NBW piglets from 16 litters suckled the sow until d19 of age or were artificially reared starting at d3 until d19 of age. Blood and tissue samples that were collected after euthanasia at 0, 3, 5, 8 and 19 d of age. Histology, ELISA, and Ussing chamber analysis were used to study proximal and distal small intestine histomorphology, proliferation, apoptosis, tight junction protein expression, and permeability. Furthermore, small intestine,liver and systemic redox parameters（GSH, GSSG, GSH-px and MDA） were investigated using ***： LBW and NBW artificially reared piglets weighed respectively 40 and 33% more than LBW and NBW sowreared piglets at d19（p 〈 0.01）. Transferring piglets to a nursery at d3 resulted in villus atrophy, increased intestinal FD-4 and HRp permeability and elevated GSSG/GSH ratio in the distal small intestine at d5（p 〈 0.05）. GSH concentrations in the proximal small intestine remained stable, while they decreased in the liver（p 〈 0.05）. From d5 until d19, villus width and crypt depth increased, whereas pCNA, caspase-3, occludin and claudin-3 protein expressions were reduced. GSH,GSSG and permeability recovered in artificially reared piglets（p 〈 0.05）.Conclusion： The results suggest that artificial rearing altered the morphology, permeability and redox state without compromising piglet performance. The observed effects were not depending on birth weight.

Gene therapy using small interfering RNA(siRNA)is emerging as a novel therapeutic approach to treat various ***,safe and efficient siRNA delivery still constitutes the major obstacle for clinical implementation of siRNA *** we report an ionizable supramolecular dendrimer vector,formed via self-assembly of a small amphiphilic dendrimer,as an effective siRNA delivery system with a favorable safety *** virtue of the ionizable tertiary amine terminals,the supramolecular dendrimer has a low positively charged surface potential and no notable cytotoxicity at physiological ***,this ionizable feature imparted sufficient surface charge to the supramolecular dendrimer to enable formation of a stable complex with siRNA via electrostatic *** resulting siRNA/dendrimer delivery system had a surface charge that was neither neutral,thus avoiding aggregation,nor too high,thus avoiding cytotoxicity,but was sufficient for favorable cellular uptake and endosomal release of the *** tested in different cancer cell lines and patient-derived cancer organoids,this dendrimer-mediated siRNA delivery system effectively silenced the oncogenes Myc and Akt2 with a potent antiproliferative effect,outperforming the gold standard vector,Lipofectamine ***,this ionizable supramolecular dendrimer represents a promising vector for siRNA *** concept of supramolecular dendrimer nanovectors via self-assembly is new,yet easy to implement in practice,offering a new perspective for supramolecular chemistry in biomedical applications.

Characteristics of Clostridium difficile infection in patients hospitalized with myelodysplastic syndrome or acute myelogenous leukemia

AIM To evaluate factors associated with Clostridium difficile infection (CDI) and outcomes of CDI in the myelodysplastic syndrome(MDS) and acute myeloid leukemia (AML) *** After IRB approval,all MDS/AML patients hospitalized at the University of Maryland Greenebaum Comprehensive Cancer Center between August 2011 and December 2013 were *** charts were reviewed for demographics,clinical information,development of CDI,complications of CDI,and *** with CDI,defined as having a positive stool pCR done for clinical suspicion of CDI,were compared to those without CDI in order to identify predictors of disease.A t-test was used for comparison of continuous variables and chisquare or Fisher's exact tests were used for categorical variables,as *** Two hundred and twenty-three patients (60.1% male,mean age 61.3 years,13% MDS,87% AML) had 594 unique hospitalizations during the study *** patients (15.2%) were diagnosed with *** significantly associated with CDI included lower albumin at time of hospitalization (p prior diagnosis of CDI (p pt of cytarabine-based chemotherapy (p = 0.015),total days of neutropenia (p = 0.014),and total days of hospitalization (p = 0.005).Gender (p = 0.10),age (p = 0.77),proton-pump inhibitor use (p = 0.73),receipt of antibiotics (p = 0.66),and receipt of DNA hypomethylating agent-based chemotherapy (p = 0.92) were not significantly associated with *** CDI is common in the MDS/AML *** significantly associated with CDI in this population include low albumin,prior CDI,use of cytarabine-based chemotherapy,and prolonged *** this study,we have identified a subset of patients in which prophylaxis studies could be targeted.

Mobile Ad Hoc Networks （MANETs） are receiving significant interest and are becoming very popular in the world of wireless networks and telecommunication. MANETs consist of mobile nodes which can communicate with each other without any infrastructure or centralized administration. In MANETs, the movement of nodes is unpredictable and complex;thus making the routing of the packets challenging. Most of the work done on the performance evaluation of routing protocols is done using the Constant Bit Rate （CBR） traffic. This paper involves the modeling and simulation of Mobile Ad hoc Networks （MANETs）. The performance analysis of the MANET routing protocols such as Ad hoc on Demand Distance Vector （AODV）, Dynamic Source Routing （DSR）, Temporary Ordered Routing Algorithm （TORA）, and Optimized Link State Routing （OLSR） of MANET routing protocols are evaluated under different scenarios using hypertext transfer protocol （http） traffic. The performance metrics used for the evaluation of these routing protocols are delay and throughput as function of the load;that is under light browsing and heavy browsing. The overall results show that the proactive routing protocol （OLSR） performs better in terms of delay and throughput than the reactive routing protocols AODV, DSR and TORA for a medium size MANETs

Finding the Asymptotically Optimal Baire Distance for Multi-Channel Data

A novel permutation-dependent Baire distance is introduced for multi-channel data. The optimal permutation is given by minimizing the sum of these pairwise distances. It is shown that for most practical cases the minimum is attained by a new gradient descent algorithm introduced in this article. It is of biquadratic time complexity: Both quadratic in number of channels and in size of data. The optimal permutation allows us to introduce a novel Baire-distance kernel Support Vector Machine (SVM). Applied to benchmark hyperspectral remote sensing data, this new SVM produces results which are comparable with the classical linear SVM, but with higher kernel target alignment.

Auxin homeostasis is pivotal for normal plant growth and development. The superroot2 （sur2） mutant was initially isolated in a forward genetic screen for auxin overproducers, and SUR2 was suggested to control auxin conju- gation and thereby regulate auxin homeostasis. However, the phenotype was not uniform and could not be described as a pure high auxin phenotype, indicating that knockout of CYp83B1 has multiple effects. Subsequently, SUR2 was identified as CYp83B1, a cytochrome p450 positioned at the metabolic branch point between auxin and indole glucosinolate metabolism. To investigate concomitant global alterations triggered by knockout of CYp83B1 and the countermeasures chosen by the mutant to cope with hormonal and metabolic imbalances, 10-day-old mutant seedlings were characterized with respect to their transcriptome and metabolome profiles. Here, we report a global analysis of the sur2 mutant by the use of a combined transcriptomic and metabolomic approach revealing pronounced effects on several metabolic grids including the intersection between secondary metabolism, cell wall turnover, hormone metabolism, and stress responses. Metabolic and transcriptional cross-talks in sur2 were found to be regulated by complex interactions between both positively and negatively acting transcription factors. The complex phenotype of sur2 may thus not only be assigned to elevated levels of auxin, but also to ethylene and abscisic acid responses as well as drought responses in the absence of a water deficiency. The delicate balance between these signals explains why minute changes in growth conditions may result in the non-uniform phenotype. The large phenotypic variation observed between and within the different surveys may be reconciled by the complex and intricate hormonal balances in sur2 seedlings decoded in this study.

Toxicogenomics has played a major role in the past decade to uncover cellular stress responses that underlie chemical-induced adverse reactions at the cellular as well as organ level and subsequently apply transcriptomics-based classifiers to predict adverse *** question remains what functional role these stress response pathways as well as the individual genes that underlie these stress responses play in the onset of *** have integrated transcrip-tomics, phosphoproteomics,and RNA interference (RNAi) approaches and used time-resolved live cell high content imaging of cellular stress responses to identify critical cell signaling components that determine the breaking point from adaptation to cell stress versus maladaptation and onset of cell *** pluripotent stem cells,DNA damage triggers loss-of-pluripotency and apopto-sis as a safeguard to exclude damaged DNA from the *** intricate DNA damage response (DDR) signaling network ensures that the response is proportional to the severity of the damage. We combined an RNAi screen targeting all kinases,phosphatases,and transcription factors with global transcriptomics and phosphopro-teomics to map the DDR in mouse embryonic stem cells treated with the DNA crosslinker cisplatin.
