Reptile sex determination is attrac.ing muc. attention bec.use the great diversity of sex-determination and dosage c.mpensation mec.anisms permits us to approac. fundamental questions about mec.anisms of sex c.romosome turnover. Rec.nt studies have made signific.nt progress in better understanding diversity and c.nservation of reptile sex c.romosomes, with however no reptile master sex determination genes identified. Here we desc.ibe an integrated genomic. and c.togenetic. pipeline,c.mbining probes generated from the mic.odissec.ed sex c.romosomes with transc.iptome and genome sequenc.ng to explore the sex c.romosome diversity in non-model Australian reptiles. We tested our pipeline on a turtle, two spec.es of gec.os, and a monitor lizard. Genes identified on sex c.romosomes were c.mpared to the c.ic.en genome to identify homologous regions among the four spec.es. We identified c.ndidate sex determining genes within these regions, inc.uding c.nserved vertebrate sex-determining genes pdgfa,pdgfra amh and wt1, and demonstrated their testis or ovary-spec.fic.expression. All four spec.es showed gene-by-gene rather than c.romosome-wide dosage c.mpensation. Our results imply that reptile sex c.romosomes originated by independent ac.uisition of sex-determining genes on different autosomes,as well as transloc.tions between different anc.stral mac.o-and *** disc.ss the evolutionary drivers of the slow differentiation and turnover of reptile sex c.romosomes.

Toxic.genomic. has played a major role in the past dec.de to unc.ver c.llular stress responses that underlie c.emic.l-induc.d adverse reac.ions at the c.llular as well as organ level and subsequently apply transc.iptomic.-based c.assifiers to predic. adverse *** question remains what func.ional role these stress response pathways as well as the individual genes that underlie these stress responses play in the onset of *** have integrated transc.ip-tomic., phosphoproteomic.,and RNA interferenc. (RNAi) approac.es and used time-resolved live c.ll high c.ntent imaging of c.llular stress responses to identify c.itic.l c.ll signaling c.mponents that determine the breaking point from adaptation to c.ll stress versus maladaptation and onset of c.ll *** pluripotent stem c.lls,DNA damage triggers loss-of-pluripotenc. and apopto-sis as a safeguard to exc.ude damaged DNA from the *** intric.te DNA damage response (DDR) signaling network ensures that the response is proportional to the severity of the damage. We c.mbined an RNAi sc.een targeting all kinases,phosphatases,and transc.iption fac.ors with global transc.iptomic. and phosphopro-teomic. to map the DDR in mouse embryonic.stem c.lls treated with the DNA c.osslinker c.splatin. Integrated networks derived from c.nonic.l pathways shared in all three datasets,were implic.ted in DNA damage repair,c.ll c.c.e and survival, and *** probing of these networks identified,amongst others a novel,p53-independent mode of DNA damage-induc.d Wnt signaling that limits *** findings reveal a balanc. between p53-mediated elimination of stem c.lls,through loss-of-pluripo-tenc. and apoptosis,and Wnt signaling that attenuates this response to tune the outc.me of the *** c.rrently explore several other newly identified signaling networks that modulate the outc.me of the *** further reveal the c.mplexity of dynamic.toxic.ty-related signaling events we have developed systems mic.

Auxin homeostasis is pivotal for normal plant growth and development. The superroot2 （sur2） mutant was initially isolated in a forward genetic.sc.een for auxin overproduc.rs, and SUR2 was suggested to c.ntrol auxin c.nju- gation and thereby regulate auxin homeostasis. However, the phenotype was not uniform and c.uld not be desc.ibed as a pure high auxin phenotype, indic.ting that knoc.out of c.P83B1 has multiple effec.s. Subsequently, SUR2 was identified as c.P83B1, a c.toc.rome P450 positioned at the metabolic.branc. point between auxin and indole gluc.sinolate metabolism. To investigate c.nc.mitant global alterations triggered by knoc.out of c.P83B1 and the c.untermeasures c.osen by the mutant to c.pe with hormonal and metabolic.imbalanc.s, 10-day-old mutant seedlings were c.arac.erized with respec. to their transc.iptome and metabolome profiles. Here, we report a global analysis of the sur2 mutant by the use of a c.mbined transc.iptomic.and metabolomic.approac. revealing pronounc.d effec.s on several metabolic.grids inc.uding the intersec.ion between sec.ndary metabolism, c.ll wall turnover, hormone metabolism, and stress responses. Metabolic.and transc.iptional c.oss-talks in sur2 were found to be regulated by c.mplex interac.ions between both positively and negatively ac.ing transc.iption fac.ors. The c.mplex phenotype of sur2 may thus not only be assigned to elevated levels of auxin, but also to ethylene and absc.sic.ac.d responses as well as drought responses in the absenc. of a water defic.enc.. The delic.te balanc. between these signals explains why minute c.anges in growth c.nditions may result in the non-uniform phenotype. The large phenotypic.variation observed between and within the different surveys may be rec.nc.led by the c.mplex and intric.te hormonal balanc.s in sur2 seedlings dec.ded in this study.

Geodynamic.proc.ss as advec.ion-c.nvec.ion of the Mid-Atlantic.Oc.an Ridge (MAR), that is exposed on land in Ic.land is investigated. Advec.ion is c.nsidered for the plate spreading veloc.ty. Geodetic.GPS data during 2000-2010 is used to estimate plate spreading veloc.ty along a profile in the Eastern Volc.nic.Zone (EVZ), Ic.land striking N102。E, approximately parallel to the NUVEL-1A spreading direc.ion between the Eurasian and North Americ.n plates. To predic. subsurfac. mass flow patterns, temperature-dependent Newtonian rheology is c.nsidered in the finite-element models (FEM). All models are c.nsidered 2-D with steady-state, inc.mpressible rheology whose visc.sity depends on the subsurfac. temperature distribution. The thic.ness of lithosphere along the profile in the EVZ is identified by 700。c.isotherm and 1022 Pa s iso-visc.sity, those reac. 50 ± 3 km depth at distanc. of 100 km from rift axis. Geodetic.observation and model predic.ion results show the ~90% of spreading is ac.ommodated within ~45 km of the rift axis in eac. direc.ion. Model predic.s ~8.5 ***-1 subsidenc. at the surfac. of rift c.nter when magmatic.plumbing is inac.ive. The rift c.nter (the highest magmatic.influx is ~11 ***-1) in model shifts ~10 - 20 km west to generate observed style surfac. deformation. The spreading veloc.ty, isotherm and depth of isotherm are the driving forc.s resulting in the surfac. deformation. These three parameters have more or less equal weight. However, as the c.nter of deformation in the EVZ shifts and most of the subsidenc. takes plac. in the volc.nic.system that is c.rrently the ac.ive whic. is the loc.ted of plate axis.

In this report,a series of novel piperidine-substituted thiophene[3,2-d]pyrimidine derivatives were designed to explore the hydrophobic.c.annel of the non-nuc.eoside reverse transc.iptase inhibitors binding poc.et(NNIBP)by inc.rporating an aromatic.moiety to the left wing of the lead K-5 *** newly synthesized c.mpounds were evaluated for anti-HIV potenc. in MT-4 c.lls and inhibitory ac.ivity to HIV-1 reverse transc.iptase(RT).Most of the synthesized c.mpounds exhibited broad-spec.rum ac.ivity toward wild-type and a wide range of HIV-1 strains c.rrying single non-nuc.eoside reverse transc.iptase inhibitors(NNRTI)-resistant ***,c.mpound 26 exhibited the most potent ac.ivity against wild-type and a panel of single mutations(L1001,K103 N,Y181 c.Y188 L and E138 K)with an Ec.0 ranging from 6.02 to 23.9 nmol/L,whic. were c.mparable to those of etravirine(ETR).Moreover,the RT inhibition ac.ivity,preliminary struc.ure-ac.ivity relationship and molec.lar doc.ing were also ***,26 exhibited favorable pharmac.kinetic.(PK)profiles and with a bioavailability of 33.8%.Taken together,the results c.uld provide valuable insights for further optimization and c.mpound 26 holds great promise as a potential drug c.ndidate for the treatment of HIV-1 infec.ion.

Purpose The utility of positron emitter nuc.ei generated by photonuc.ear reac.ions from MM50 is verified for dose delivery and loc.tion monitoring in a phantom and patient after 50 MV high energy photon in radiation therapy. Materials and methods The tec.nique is based on the ac.ivation of body tissues by high energy bremsstrahlung beams,energies well above 20 MeV,resulting primarily in c.and O but also N,all positron-emitting radionuc.ides produc.d by photoneutron reac.ions in the nuc.ei of c.O and *** relations of c.oss sec.ion for different photonuc.ear reac.ions with energy are *** the c.rbon and oxygen radionuc.ides are the most abundant elements in soft tissue photonuc.ear reac.ions we study these two radionuc.ides.A solid water phantom and a frozen animal tissue were irradiated with an X-ray beam energy of 50 MV from *** 3D radiation treatment planning system,Nuc.etron-TPP,was used to c.lc.late the delivered dose *** phantom or tissue was direc.ly simulated in the Nuc.etron-TPP after c. sc.n. This paper presents equipment for early detec.ion of failures in the insulation of power transformers, c.ec.ing existing partial disc.arges inside. The equipment involves hardware, c.ntrol and signal ac.uisition software, and signal analysis software. This equipment has a set of algorithms that were made with intelligent extrac.ion tec.niques and interpretation of data. The degradation diagnosis of the equipment insulation is based on digital signal proc.ssing algorithms for extrac.ion of features and also in artific.al intelligenc. algorithms that allies a mining involving all the data linked to the equipment throughout its operating life c.n make an assessment of the operating c.nditions of the equipment and suggest interventions and provide an estimated time so that they have to be made.
