Investigation of HCAR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving the herd. In vitro administration of beta-hydroxybutyric acid(BHBA;the primary blood ketone) has inhibitory effects on the function of bovine leukocytes. BHBA is a ligand of HCAR2 and the activation of these receptors promotes an anti-inflammatory response which may be related with immunosuppression observed in transition dairy cattle. The obj.ctive of this study was to identify and test antagonists for HCAR2 in bovine immune cells cultured with *** We observed expression of HCAR2 at the protein level within lymphocytes, monocytes, and granulocytes. The proportion of cells expressing HCAR2 tended to be greater in mid-lactation compared to early lactation cows;the increase was a result of increased proportion of T and B cells expressing HCAR2. Stimulation of HCAR2 with niacin or BHBA promoted Ca^(2+) mobilization in neutrophils and mononuclear cells. Mononuclear cells treated with BHBA had diminished intracellular Ca^(2+) responses when HCAR2 was knocked down by si RNA silencing, indicating Ca^(2+) mobilization was mediated by HCAR2 signaling. Two candidate antagonists for HCAR2, synthesized from niacin(NA-1 and NA-5), were tested;monocytes and neutrophils pre-treated with NA-1 and NA-5 had reduced Ca^(2+) mobilization after incubation with BHBA. Furthermore, NA-5 but not NA-1 prevented BHBA-associated reductions in cyclic *** We demonstrated that HCAR2 is present on bovine leukocytes and has greater expression later in lactation. We confirmed that BHBA and niacin derived HCAR2 antagonists alter bovine leukocyte activity. Our results demonstrate that both BHBA and niacin affect bovine leukocyte Ca^(2+) mobilization in a HCAR2-dependent manner.

Characterizing foliar trait variation in sun and shade leaves can provide insights into inter-and intra-species resource use strategies and plant response to environmental ***,datasets with records of multiple foliar traits from the same individual and including shade leaves are sparse,which limits our ability to investigate trait-trait,trait-environment relationships and trait coordination in both sun and shade *** presented a comprehensive dataset of 15 foliar traits from sun and shade leaves sampled with leaf spectroscopy,including 424 individuals of 110 plant species from 19 sites across eastern North *** investigated trait variation,covariation,scaling relationships with leaf mass,and the effects of environment,canopy position,and taxonomy on trait ***,sun leaves had higher leaf mass per area,nonstructural carbohydrates and total phenolics,lower mass-based chlorophyll a+b,carotenoids,phosphorus,and potassium,but exhibited species-specific *** between sun and shade leaf traits,and trait-environment relationships were overall consistent across *** main dimensions of foliar trait variation in seed plants were revealed including leaf economics traits,photosynthetic pigments,defense,and structural *** and canopy position collectively explained most of the foliar trait *** study highlights the importance of including intra-individual and intra-specific trait variation to improve our understanding of ecosystem *** findings have implications for efficient field sampling,and trait mapping with remote sensing.

Hydrological models are developed to simulate river flows over a watershed for many practical applications in the field of water resource management. The present paper compares the performance of two recurrent neural networks for rainfall-runoff modeling in the Zou River basin at Atchérigbé outlet. To this end, we used daily precipitation data over the period 1988-2010 as input of the models, such as the Long Short-Term Memory (LSTM) and Recurrent Gate Networks (GRU) to simulate river discharge in the study area. The investigated models give good results in calibration (R2 = 0.888, NSE = 0.886, and RMSE = 0.42 for LSTM;R2 = 0.9, NSE = 0.9 and RMSE = 0.397 for GRU) and in validation (R2 = 0.865, NSE = 0.851, and RMSE = 0.329 for LSTM;R2 = 0.9, NSE = 0.865 and RMSE = 0.301 for GRU). This good performance of LSTM and GRU models confirms the importance of models based on machine learning in modeling hydrological phenomena for better decision-making.

Some refractory linings that protect metallic vessels from the hot temperature of the products they contain are made of masonries with or without *** j.ints play an important role,reducing the stresses in the masonries during ***,the presence of these j.ints makes the behaviour of the masonry nonlinear and *** perform a thermomechanical simulation using a finite element method of an industrial vessel that contains hundreds or thousands of bricks and j.ints,microscopic models are not suitable due to the high computational time and the management of the behaviour of the j.ints(opening/closing)which affects the *** overcome these problems,it is proposed to replace the masonry by a homogeneous material that has a behaviour equivalent to the set of bricks and j.ints,using a periodic homogenization *** the j.ints can be closed or open,the equivalent material will have a different behaviour according to the j.int ***,refractory materials have an elastic-viscoplastic behaviour at high ***,the equivalent material will have an orthotropic elastic-viscoplastic behaviour,requiring a nonlinear homogenisation *** overview of this approach developed at University of Orléans is presented with two industrial applications(blast-furnace and steel ladle).
