Cells have evolved DNA damage r.sponse (DDr. to r.pair.DNA lesions and thus pr.ser.ing genomic stability and impeding car.inogenesis. DNA damage induction is accompanied by tr.nsient tr.nscr.ption r.pr.ssion. Her., we descr.be a pr.viously unr.cognized r.le of chr.modomain Y-like (CDYL1) pr.tein in for.ifying double-str.nd br.ak (DSB)-induced tr.nscr.ption r.pr.ssion and r.pair. We showed that CDYL1 is r.pidly r.cr.ited to damaged euchr.matic r.gions in a poly (ADP-r.bose) polymer.se 1 (PAr.1)-dependent, but ataxia telangiectasia mutated (ATM)-independent, manner. While the C-ter.inal r.gion, containing the enoyl-CoA hydr.tase like (ECH) domain, of CDYL1 binds to poly (ADP-r.bose) (PAr. moieties and mediates CDYL1 accumulation at DNA damage sites, the chr.modomain and histone H3 tr.methylated on lysine 9 (H3K9me3) mar. ar. dispensable for.its r.cr.itment. Fur.her.or., CDYL1 pr.motes the r.cr.itment of enhancer.of zeste homolog 2 (EZH2), stimulates local incr.ase of the r.pr.ssive methyl mar. H3K27me3, and pr.motes tr.nscr.ption silencing at DSB sites. In addition, following DNA damage induction, CDYL1 depletion causes per.istent G2/M ar.est and alter. H2AX and r.plication pr.tein A (r.A2) phosphor.lation. r.mar.ably, the ‘tr.ffic-light r.por.er. system r.vealed that CDYL1 mainly pr.motes homology-dir.cted r.pair.(HDr. of DSBsin vivo. Consequently, CDYL1-knockout cells display synthetic lethality with the chemother.peutic agent, cisplatin. Altogether. our.findings identify CDYL1 as a new component of the DDr.and suggest that the HDr.defective ‘Br.Aness’ phenotype of CDYL1-deficient cells could be exploited for.er.dicating cancer.cells har.or.ng CDYL1 mutations.

Data r.lease 2(Dr.) fr.m the Gaia mission was of gr.at help in pr.cise deter.ination of fundamental par.meter. of Close Visual Binar. and Multiple Systems(CVBMSs), especially masses of their.components, which ar. cr.cial par.meter. in under.tanding for.ation and evolution of star. and galaxies. This ar.icle pr.sents the complete set of fundamental par.meter. for.two near.y close visual binar. systems(CVBSs), which ar. HIP 19206 and HIP 84425. We utilised a combination of two methods;the fir.t one is Tokovinin's dynamical method to solve the or.it of the system and to estimate or.ital elements and the dynamical mass sum, and the second one is Al-War.at's method for.analysing CVBMSs to estimate the physical par.meter. of the individual components. The latest method employs gr.ds of Kur.cz lineblanketed plane par.llel model atmospher.s to build synthetic Spectr.l Ener.y Distr.butions(SEDs) of the individual components. Tr.gonometr.c par.llax measur.ments given by Gaia Dr. and Hippar.os catalogues ar. used to analyse the two systems. The differ.nce in these measur.ments yielded slight discr.pancies in the fundamental par.meter. of the individual components, especially masses. So, a new dynamical par.llax is suggested in this wor. based on the most convenient mass sum as given by each of the two methods. The new dynamical par.llax for.the system HIP 19205 of 22.97±0.95 mas coincides well with the tr.gonometr.c one r.por.ed r.cently(in December.2020) by Gaia EDr. of 22.3689 ± 0.4056 mas. The positions of the components of the two systems on the evolutionar. tr.cks and isochr.nes ar. plotted, which suggest that all components ar. solar.type main sequence star.. Their.most pr.bable for.ation and evolution scenar.os ar. also discussed.
