We isolated naturally occurring actinomycetes with an ability to produce metabolites having antifungal property against, Colletotrichum gloeosporioides, the causal agent of mango anthracnose. One promising strain was strong antifungal activity, was selected for further studies. Based on the physiological and biochemical characteristics, the bacterial strain was identical to Streptomyces aureofaciens. Culture filtrate collected from the exponential and stationary phases inhibited the growth of fungus tested, indicating that growth suppression was due to extracellular antifungal metabolites present in culture filtrate. Isolate highly produced extracellular chitinase and β-1,3-glucanase during the exponential and late exponential phases, respectively. In order to standardize the metabolite production some cultural conditions like different incubation time in hours, pH, carbon sources and concentrations and nitrogen source were determined. During fermentation, growth, pH and hydrolysis enzymes production were monitored .Treatment with bioactive components exhibited a significantly high protective activity against development of anthracnose disease on mango trees and increased fruit yield.

AIM: To evaluate the effect of resveratrol, alone and in combination with fenofibrate, on fructose-induced metabolic genes abnormalities in rats. METHODS: Giving a fructose-enriched diet(FED) to rats for 12 wk was used as a model for inducing hepatic dyslipidemia and insulin resistance. Adult male albino rats(150-200 g) were divided into a control group and a FED group which was subdivided into 4 groups, a control FED, fenofibrate(FENO)(100 mg/kg), resveratrol(RES)(70 mg/kg) and combined treatment( FENO + RES)( half the doses). A l l treatments were given orally from the 9th week till the end of experimental period. Body weight, oral glucose tolerance test(OGTT), liver index, glucose, insulin, insulin resistance(HOMA), serum and liver triglycerides(TGs), oxidative stress(liver MDA, GSH and SOD),serum AST, ALT, AST/ALT ratio and tumor necrosis factor-α(TNF-α) were measured. Additionally, hepatic gene expression of suppressor of cytokine signaling-3(SOCS-3), sterol regulatory element binding protein-1c(SREBP-1c), fatty acid synthase(FAS), malonyl Co A decarboxylase(MCD), transforming growth factor-β1(TGF-β1) and adipose tissue genes expression of leptin and adiponectin were investigated. Liver sections were taken for histopathological examination and steatosis area were ***: Rats fed FED showed damaged liver, impairment of glucose tolerance, insulin resistance, ox ida t ive s t re s s a nd dy s l ipide m ia. As fo r ge ne expression, there was a change in favor of dyslipidemia and nonalcoholic steatohepatitis(NASH) development. All treatment regimens showed some benefit in reversing the described deviations. Fructose caused deterioration in hepatic gene expression of SOCS-3, SREBP-1c, FAS, MDA and TGF-β1 and in adipose tissue gene expression of leptin and adiponectin. Fructose showed also an increase in body weight, insulin resistance(OGTT, HOMA), serum and liver TGs, hepatic MDA, serum AST, AST/ALT ratio and TNF-α compared to control. All treatments im

Staphylococcus aureus represents a public health challenge all over the world. Therefore, this study aims to analyze the prevalence of five genes (sea, seb, sec, see and seg) encoding the staphylococcal enterotoxins in S. aureus isolated from different sources and to evaluate the association of these toxins in comparison to susceptibility towards 12 antimicrobials;antimicrobial susceptibility was conducted by disc diffusion method. Detection of staphylococcal enterotoxins was performed by PCR and the ability to express these genes was assessed among isolates by RT-PCR. The most common enterotoxin gene was sea gene (66%), followed by seb, sec, see and seg (38%, 23%, 19% and 5%) respectively. Expression of sea, seb and seg genes was variable. However, sec and see genes were not expressed by any of the tested isolates. No statistically significant association exists between (seb, sec and see) and isolation sources, while the sea was significantly associated with clinical isolates. High significant correlation was found between elevated sea expression and multidrug-resistance. Our findings indicate that the pathogenic potential of S. aureus may be greater than previously thought. This emphasizes the utmost need to implement proactive measures and more emphasis will be placed on the application of hygiene practices in hospitals to control S. aureus infection and enterotoxins production.

Background: Chronic kidney disease (CKD) is associated with cardiovascular morbidity and mortality. Osteopontin (OPN) is a critical factor in developing atherosclerosis and increases the risk for a major adverse cardiovascular event. Aim: To investigate osteopontin serum levels in hemodialysis children and detect the association between the main arteries’ intimal medial thickness (IMT) and peak systolic velocity. Material and Methods: This case-control study included 30 children on regular hemodialysis and 30 children age and sex-matched as controls their age range from 4 to 18 years;we investigated osteopontin serum level in addition to the Doppler ultrasound assessment of intimal medial thickness and peak systolic velocity (PSV) of the main arteries in the same line with the traditional markers of the routine investigations of children on regular hemodialysis. Results: Significantly high osteopontin level in hemodialysis children than the controls was (0.85 ± 0.21 ng/ml) (0.69 ± 0.26 ng/ml), respectively (p = 0.026). A significant increase in the (IMT) of the main arteries, including the carotid and femoral arteries, in the patient’s group than the controls, was (0.51 ± 0.01 mm) (0. 69. ± 0.01 mm) (0.32 ± 0.036 mm) (0.55 ± 0.01), respectively (p = 0.001). There is a strong correlation between (OPN) with the (IMT) of carotid and femoral arteries, a significant positive correlation between (OPN) with urea, creatinine, triglyceride, PTH, ferritin, CRP, and ESR, and a negative correlation with RBCs count and carotid PSV. Conclusion: High osteopontin level is consistent with the increased IMT of the main arteries in hemodialysis children that seem to play a significant role in developing and propagating atherosclerosis in hemodialysis children, evidenced by significant association with inflammatory markers and uremic toxins.

Introduction: Cervical cancer is the most common cause of preventable cancer related deaths;cervical cancer has a long pervasive phase (cervical dysplasia);the prevalence of cervical dysplasia varies according to the socioeconomic characteristics and geographic areas of the population studied. Low-grade lesions regress spontaneously in a significant number of patients, while high grade lesions will progress to an invasive cancer if left untreated. Cervical cancer screening is an important component of the World Health Organization (WHO) strategy for combating cervical cancer. The incidence and prevalence of cervical cancer has reduced remarkably over the last three decades in developed countries where there are effective, well-coordinated screening programs, and treatment of cervical dysplasia, while in developing countries it has been increasing and has constituted major health problems among women where there are no well-coordinated and effective screening programs, also resources are very low and no insurance can cover this programs. Aim of the work: The aim is to assess the prevalence of abnormal cervical cytology in Al Shatby Maternity University Hospital patients using Pap smear. Materials and methods: Inclusion criteria: 1) Married woman from 3 years or more;2) Women age from 21 to age 65 years. Exclusion criteria: 1) Previously known cervical cancer patient;2) Virgin females;3) Woman with active vaginal bleeding. Results: 83% of patients were -ve intraepithelial neoplasia {37.7% was normal cytology and 45.3% was inflammatory}. 17% was +ve intraepithelial neoplasia (abnormal cytology), {11.1% ASCUS, 2.9% LSIL, 1.3% HSIL, 1.1% ASC-H, 0.3% AGS

BACKGROUND Childhood cancer survivors are potentially at a higher risk of infection with hepatitis C virus (HCV).The effects of all-oral direct-acting antiviral therapy (DAA) on both the HCV infection as well as the state of cancer remission have not been well investigated in this *** To test the effects of dual sofosbuvir/daclatasvir (SOF/DCV) therapy in the treatment of chronic HCV in survivors of hematologic malignancy in pediatric age *** We conducted a prospective,uncontrolled,open-label multicenter study.A total of 20 eligible,chronic HCV,genotype-4,infected children who had been in continuous complete remission from hematologic cancer (leukemia/lymphoma) for at least one year were included in the *** patients were treated with combined SOF/DCV for 12 *** were monitored throughout the study till 12 wk after end of treatment for safety and efficacy outcomes including the sustained virologic response 12 (SVR12) rate,hematological indices,liver and kidney *** The intent-to-treat SVR12 rate was 20 of 20 (100%;95%CI: 84%-100%).All patients showed normalized liver enzymes from *** hematological indices,liver and kidney functions were kept normal throughout the *** fatalities or treatment-emergent serious or severe adverse events were reported throughout the *** SOF/DCV combined therapy could be used safely and effectively in the treatment of chronic HCV genotype-4 infection in leukemia/lymphoma treated *** relapses were detected during treatment and throughout the follow up period for either the original malignant disease or the HCV infection.

Indoor Surveillance of Airborne Fungi Contaminating Intensive Care Units and Operation Rooms in Assiut University Hospitals, Egypt

Mycoflora of atmospheric air and dust samples collected from air conditioning systems in 12 of each I. C. U. (intensive care units) and O. R. (operation rooms) were tested using settle and dilution plate methods on four types of agar media and incubated at 25 °C. Forty-five fungal species representing 23 genera were isolated and identified. The most prevalent genera recorded were Cladosporium, Aspergillus, Penicillium and Fusarium. The total colony forming units of airborne fungi recovered in I. C. U. and O. R. ranged between 31.13-49.61 colonies/m 3 on the four types of media used. The fungal total catch of the dust samples collected from the air conditioning system filters in I. C. U. and O. R. were ranged from 65.5-170 colonies/mg dust. Since, the interest to replace synthetic xenobiotics by natural compounds with low environmental persistence and biodegradable to control such airborne fungal contaminants is needed. In this respect, essential oils showed to possess a broad spectrum of antifungal activity. Fungal static ability of six oils was tested on 30 different fungal isolates. Vapors of common thyme oil exhibited the strongest inhibitory effects on the tested isolates, whereas the headspace vapors of blue gum and ginger had no inhibitory effects on the tested fungal isolates. These data revealed that the air conditioning systems may be an important source of contamination in I. C. U. and O. R. of Assiut university hospitals. Thus, patients may be in risk of being exposed to contaminated atmospheric air by opportunistic fungi and the use of essential oils as an alternative option to control hospital wards from fungal contaminants needs further studies.

Multiple Sclerosis (MS) is a chronic demyelinating disease of the CNS with assumed autoimmune etiology. Human herpes viruses have probable effects on relapsing-remitting MS pathogenesis presumably through molecular mimicry and/or bystander mechanisms. In this study we probed the possible contribution of the two herpes viruses, human herpesvirus 6 (HHV-6) and human herpesvirus 8 (HHV-8), in clinically definite multiple sclerosis (CDMS) pa-tients-relapsing remitting type (RRMS) during clinical exacerbations. All patients had no history of immune modulating or suppressing drugs intake in the last 6 months. The peripheral blood samples, from CDMS patients (n = 20) (13F/7M, age (y) = 30.3 ± 3.21) and other immune mediated neurological disorders (OIND) (11F/9M, age = 25.2 ± 12.1), (My-asthenia Gravis, Guillain Barré Syndrome, ischemic stroke in adolescent and young adult with no clear risk factors), as a control group, had been enrolled within 15 months (January-2007-- March -2008). We investigated the existence of specific deoxyribonucleic acid (DNA) sequences belonging to HHV-6 and HHV-8, using polymerase chain reaction (PCR) in the isolated peripheral blood mononuclear cells (PBMCs) and in plasma. PCR demonstrated HHV-6 DNA in 7 cases (35%), HHV-8 sequences in only one cases (5%) in PBMCs from 20 relapsed CDMS patients;all HHV-6 posi-tive cases showed positive plasma results, while the blood samples from 20 OIND patients showed negative results ex-cept one case (5%) out of 9 cases of GBS was positive for HHV-8 in PBMCs. We consequently concluded that there is considerable evidence in this study that proposed the roles of HHV-6 and HHV-8 in MS pathogenesis and clinical ex-acerbation.

Cost Effective Adsorption of Aluminium and Iron from Synthetic and Real Wastewater by Rice Hull Activated Carbon (RHAC)

The adsorption of aluminium(III) and iron(III) ions from their single and binary systems, by RHAC was investigated in a batch system. The activated carbon prepared from rice hulls was characterized by scanning electron microscopy and Fourier transformation infrared techniques. Batch adsorption experiments were performed under different operating conditions including pH (2 - 5), adsorbent dosage (0.5 - 2.0 g/l), initial ion concentration (5 - 100 mg/l), and contact time (30 - 240 min). The equilibrium time for maximum ions removal was found to be 180 min in single and binary ions systems. The kinetics of adsorption was evaluated using the pseudo-first order, pseudo-second order and Elovich kinetic models. The Langmuir, Freundlich and Temkin equilibrium models were applied to the adsorption experimental data. Real wastewater samples were collected from different locations to investigate the efficiency of rice hull activated carbon in treating real samples. The real wastewater samples were treated with the activated carbon prepared from rice hulls and a commercial activated carbon. The results showed that the activated carbon prepared in the present work was more efficient in the removal of aluminium and iron from real wastewater as compared to the commercial activated carbon which is more advantageous considering both economics and environmental parameters.

Formulation and evaluation of anti-asthmatic drug montelukast in mucoadhesive buccal patches

Objective: To formulate and evaluate anti-asthmatic drug montelukast in mucoadhesive buccal patches. Methods:Buccal patches were formulated by using different hydrophilic polymers by solvent casting technique. Buccal patches were evaluated by seven physical appearances, in addition toin vitro drug release study. Results: All patches were uniform and translucent, and had smooth surface. In vitro release studies were conducted for montelukast buccal patches proved that release in the range of 75.26%-92.30% in 8 h. Emission of montelukast from all patches simulated zero order and diffusion mechanism. Finally it can be concluded that F3, F15 and F16 are the best formulation. Conclusions: The investigation concluded that patch of 5 mg of montelukast sodium were formulated by using sodium alginate with sodium carboxy methyl cellulose, hydroxy propyl methyl cellulose K100M with sodium carboxy methyl cellulose, and hydroxy propyl methyl cellulose K100M with sodium alginate (F3, F15 and F16 formulations) were the best formulations.
