Disruptions in AUXl-Dependent Auxin Influx Alter Hypocotyl Phototropism in Arabidopsis

Phototropism represents a differential growth response by which plant organs can respond adaptively to changes in the direction of incident light to optimize leaf/stem positioning for photosynthetic light capture and root growth orientation for water/nutrient acquisition. Studies over the past few years have identified a number of compo- nents in the signaling pathway（s） leading to development of phototropic curvatures in hypocotyls. These include the pho- totropin photoreceptors （photl and phot2） that perceive directional blue-light （BL） cues and then stimulate signaling, leading to relocalization of the plant hormone auxin, as well as the auxin response factor NPH4/ARF7 that responds to changes in local auxin concentrations to directly mediate expression of genes likely encoding proteins necessary for development of phototropic curvatures, While null mutations in NPH4/ARF7 condition an aphototropic response to unidirectional BL, seedlings carrying the same mutations recover BL-dependent phototropic responsiveness if coirradiated with red light （RL） or pre-treated with either ethylene. In the present study, we identify second-site enhancer mutations in the nph4 background that abrogate these recovery responses. One of these mutations-mapl （modifier of arf7 phenotypes 1）-was found to represent a missense allele of AUX1-a gene encoding a high-affinity auxin influx carrier previously associated with a number of root responses. Pharmocological studies and analyses of additional auxl mutants confirmed that AUX1 functions as a modulator of hypocotyl phototropism. moreover, we have found that the strength of dependence of hypocotyl phototropism on AUX1-mediated auxin influx is directly related to the auxin responsiveness of the seedling in question.

The pig as a preclinical traumatic brain injury model:current models,functional outcome measures,and translational detection strategies

Traumatic brain injury(TBI) is a major contributor of long-term disability and a leading cause of death worldwide. A series of secondary injury cascades can contribute to cell death, tissue loss, and ultimately to the development of functional impairments. However, there are currently no effective therapeutic interventions that improve brain outcomes following TBI. As a result, a number of experimental TBI models have been developed to recapitulate TBI injury mechanisms and to test the efficacy of potential therapeutics. The pig model has recently come to the forefront as the pig brain is closer in size, structure, and composition to the human brain compared to traditional rodent models, making it an ideal large animal model to study TBI pathophysiology and functional outcomes. This review will focus on the shared characteristics between humans and pigs that make them ideal for modeling TBI and will review the three most common pig TBI models–the diffuse axonal injury, the controlled cortical impact, and the fluid percussion models. It will also review current advances in functional outcome assessment measures and other non-invasive, translational TBI detection and measurement tools like biomarker analysis and magnetic resonance imaging. The use of pigs as TBI models and the continued development and improvement of translational assessment modalities have made significant contributions to unraveling the complex cascade of TBI sequela and provide an important means to study potential clinically relevant therapeutic interventions.

Lymphocyte phenotypes in wild-caught rats suggest potential mechanisms underlying increased immune sensitivity in post-industrial environments

The immune systems of wild rats and of laboratory rats can been utilized as models of the human immune system in pre-industrial and post-industrial societies, respectively. In this study, lymphocyte phenotypes in wild rats were broadly characterized, and the results were compared to those obtained by us and by others using cells derived from various strains of laboratory rats. Although not expected, the production of regulatory T cells was not apparently different in wild rats compared to laboratory rats. On the other hand, differences in expression of markers involved in complement regulation, adhesion, signaling and maturation suggest increased complement regulation and decreased sensitivity in wild-caught rats compared to laboratory rats, and point toward complex differences between the maturation of T cells. The results potentially lend insight into the pathogenesis of post-industrial epidemics of allergy and autoimmune disease.

modeling neurodegenerative disorders in adult somatic cells： A critical review

Development of new therapeutic targets for neurodegenerative disorders has been hampered by a reliance onpost mortem tissue that is representative of end-stage disease, or on animal models that fail to provide faithful analogs. However, rapid advances in cellular genetic reprogramming, in particular the induction of somatic cells into stem cells, or directly into neurons, has led to intense interest in modeling of human neurodegeneration in vitro. Here, we critically review current methods and recent progress in cellular models of Alzheimer＇s disease, Parkinson＇s disease and amyotrophic lateral sclerosis. Several challenges are identified, including technical variability, lack of degenerative phenotypes, neurodevelopmental age and establishing ground truths for models of sporadic disease. Recommendations for evaluating neurodegenerative cellular models are proposed along with suggestions for future research.

Efficacy of an esfenvalerate plus methoprene aerosol for the control of eggs and fifth instars of Plodia interpunctella （Lepidoptera： Pyralidae）

Aerosol insecticides may provide an alternative to fumigants for control of the Indianmeal moth, Plodia interpunctella （Hübner）, the Indianmeal moth, a major insect pest of stored processed food. In this study, eggs and larvae （5th instars） ofP. interpunctella were exposed to aerosol applications of the pyrethroid esfenvalerate and insect growth regulator methoprene, alone and in combination, in open and obstructed positions inside small sheds. When larvae were exposed to methoprene alone, adult emergence from those exposed larvae was 7.1% ± 1.5%. In contrast, adult emergence was 92.5% ± 3.5% when larvae were exposed to esfenvalerate alone. When eggs were exposed to methoprene, adult emergence of those exposed eggs was approximately 75%; however, when eggs were exposed to esfenvalerate, adult emergence was approximately 35%. In the combination treatment of methoprene plus esfenvalerate at their respective label rates, adult emergence following larval exposure was 0.91% ±0.61% compared to 16.3% ± 9.6% when eggs were exposed. Based on our results, methoprene alone is highly effective in reducing adult emergence after larval exposure. However, it is not as effective on eggs as esfenvalerate. A combination treatment of esfenvalerate plus methoprene could be used to control eggs and the wandering-phase larval stages ofP. interpunctella. An economic risk analysis also supports a strategy of combining methoprene and esfenvalerate.
