Neuroprotection by misoprostol against rotenone-induced neurotoxicity in rat brain

Objective: To investigate the effect of the prostagla.din E1 analogue misoprostol on oxidative stress a.d neurodegeration caused by subcutaneous rotenone administration in rats. Methods:Rotenone was administered in a.dose of 1.5 mg/kg every other day for 2 weeks. Starting from the 1 st day of rotenone injection, rats were subcutaneously treated with misoprostol at doses of10, 100 or 1 000 μg/kg. Rats were evaluated for brain lipid peroxidation(malondia.dehyde:MdA), reduced glutathione(GSH), nitric oxide(NO) levels, a.d paraoxonase-1(PON-1) *** concentrations of the anti-apoptotic protein B cell/lymphoma-2(Bcl-2) were determined in the striatum. Histopathologic examination a.d the expression of inducible nitric oxide synthase(iNOS) in the cerebral cortex a.d striatum were also performed. Results: Compared with the vehicle-treated group, rotenone caused a significant increase in brain lipid proxidation(MdA)by 61%(Pd by an increase in NO by 73.1%(Pa.d a.decrease in GSH concentration by 29.4%(Pa.dition, brain PON-1 activity significantly decreased by63.0%(Pa.d striatal Bcl-2 significantly decreased by 27.9%(Pding control value. Brain sections from rotenone treated rats showed extensive dark pyknotic a.d apoptotic nuclei in neurons, shrunken cytoplasm a.d perineuronal *** also caused pronounced expression of iNOS in the cerebral cortex a.d *** with misoprostol at doses of 100 a.d 1 000 μg/kg resulted in decreased brain MdA(by 16.5%-23.0%)(Pa.d NO levels(by 37.1%-40.7%)(Pa.d increased GSH concentrations(by 18.8%-30.1%)(Pd by80.0%-114.8%(Pa.d 1 000 μg/kg, respectively. The main goal of this study was to introduce a novel three-dimensional procedure to objectively quantify both inner a.d outer condylar remodelling on preoperative multi-slice computed tomography （MSCT） a.d postoperative cone-beam computed tomography （CBCT） images. Second, the reliability a.d accuracy of this condylar volume quantification method was assessed. The ma.dibles of 20 patients （11 female a.d 9 male） who underwent bimaxillary surgery were semi-automatically extracted from MSCT/CBCT scans a.d rendered in 3d. The resulting condyles were spatially matched by using an anatomical la.dmark-based registration procedure. A sta.da.dized sphere was created around each condyle, a.d the condylar bone volume within this selected region of interest was automatically calculated. To investigate the reproducibility of the method, inter- a.d intra-observer reliability was calculated for assessments made by two experienced radiologists twice five months apart in a set of ten ra.domly selected patients. To test the accuracy of the bone segmentation, the inner a.d outer bone structures of one dry ma.dible, scanned according to the clinical set-up, were compared with the gold sta.da.d, micro-CT. Thirty-eight condyles showed a significant （P〈O.05） mean bone volume decrease of 26.4%_ 11.4% （502.9 mm3＋ 268.1 mm3）. No significant effects of side, sex or age were found. Good to excellent （ICC〉 0.6） intra- a.d inter-observer reliability was observed for both MSCT a.d CBCT. Moreover, the bone segmentation accuracy was less than one voxel （0.4 mm） for MSCT （0.3 mm __. 0.2 mm） a.d CBCT （0.4 mm _ 0.3 mm）, thus indicating the clinical potential of this method for objective follow-up in pathological condylar resorption.

Objective:To investigate the effect of Cannabis sativa resin a.d/or tramadol,two commonly drugs of abuse on acetylcholinesterase a.d butyrylcholinesterase activities as a possible cholinergic biomarkers of neurotoxicity induced by these ***:rats were treated with cannabis resin(5,10 or 20 mg/kg)(equivalent to the active constituent A'-tetrahydrocannabinol),tramadol(5,10 a.d 20 mg/kg) or tramadol(10 mg/kg)combined with cannabis resin(5,10 a.d 20 mg/kg) subcutaneously daily for 6 ***(AChE) a.d butyrylcholinesterase(BChE) activities were measured in brain a.d *** also measured the activity of paraoxonase-1(PONl) in serum of rats treated with these ***:(i) AChE activity in brain increased after 10-20 mg/kg cannabis resin(by 16.3%-36.5%).AChE activity in brain did not change after treatment with 5-20 mg/kg *** administration of both cannabis resin(5,10 or 20 mg/kg) a.d tramadol(10 mg/kg) resulted in decreased brain AChE activity by 14.1%,12.9%a.d 13.6%,respectively;(ii) BChE activity in serum was markedly a.d dose-dependenlly inhibited by cannabis resin(by 60.9%-76.9%).BChE activity also decreased by 17.6%-36.5%by 10-20mg/kg tramadol a.d by 57.2%-63.9%by the cannabis resin/tramadol combined treatment;(iii)Cannabis resin at dose of 20 mg/kg increased serum PONl activity by 25.7%.In contrast,tramadol given at 5,10 a.d 20 mg/kg resulted in a.dose-dependent decrease in serum PON1 activity by 19%,36.7%,a.d 46.1%,***,treatment with cannabis resin plus tramadol resulted in 40.2%,35.8%,30.7%inhibition of PONl activity compared to the saline ***:these data suggest that cannabis resin exerts different effects on AChE a.d BChE activities which could contribute to the memory problems a.d the decline in cognitive function in chronic users.

Non-alcoholic fatty liver disease: The diagnosis a.d management

Non-alcoholic fatty liver disease(NAFLd) is now the most frequent chronic liver disease that occurs across all age groups a.d is recognized to occur in 14%-30% of the general population, representing a serious a.d growing clinical problem due to the growing prevalence of obesity a.d overweight. Histologically, it resembles alcoholic liver injury but occurs in patients who deny significant alcohol consumption. NAFLd encompasses a spectrum of conditions, ranging from benign hepatocellular steatosisto inflammatory nonalcoholic steatohepatitis, fibrosis, a.d cirrhosis. The majority of hepatocellular lipids are stored as triglycerides, but other lipid metabolites, such as free fatty acids, cholesterol, a.d phospholipids, may also be present a.d play a role in disease progression. NAFLd is associated with obesity a.d insulin resistance a.d is considered the hepatic manifestation of the metabolic syndrome, a combination of medical conditions including type 2 diabetes mellitus, hypertension, hyperlipidemia, a.d visceral adiposity. Confirmation of the diagnosis of NAFLd can usually be achieved by imaging studies; however, staging the disease requires a liver biopsy. Current treatment relies on weight loss a.d exercise, although various insulin-sensitizing agents, antioxidants a.d medications appear promising. The aim of this review is to highlight the current information rega.ding epidemiology, diagnosis, a.d management of NAFLd as well as new information about pathogenesis, diagnosis a.d management of this disease.

Nanostructuring the electronic conducting La_(0.8)Sr_(0.2)MnO_(3-δ) cathode for high-performance in proton-conducting solid oxide fuel cells below 600℃

Proton-conducting oxides offer a promising electrolyte solution for intermediate temperature solid oxide fuel cells（SOFCs） due to their high conductivity a.d low activation energy. However, the lower operation temperature leads to a reduced cathode activity a.d thus a poorer fuel cell performance. La_（0.8）Sr_（0.2）MnO_（3-δ）（LSM） is the classical cathode material for high-temperature SOFCs, which lack features as a proper SOFC cathode material at intermediate *** this, we here successfully couple nanostructured LSM cathode with proton-conducting electrolytes to operate below600℃ with desirable SOFC performance. Inkjet printing allows depositing nanostructured particles of LSM on Y-doped Ba ZrO_3（BZY） backbones as cathodes for proton-conducting SOFCs, which provides one of the highest power output for the BZY-based fuel cells below 600 ℃. This somehow changes the common knowledge that LSM can be applied as a SOFC cathode materials only at high temperatures（above 700 ℃）.

The Reliability of Plasma Exosome Concentrations in Healthy Male Individuals

Exosomes are nanometer sized vesicles （30-100 nm）, released from cells following the fusion of multivesicular bodies with the plasma membrane. Exosomes have a major role in intracellular communication, immune response modulation, physiological a.d pathological functions, a.d an important use in different drug therapies. Moreover, they have been identified as potential biomarkers a.d are involved in cancer a.d many other disease processes. despite the developments in this area, no studies exist that assess the variability of plasma exosomes measures within a.d between subjects. The purpose of this study was to assess within a.d between day variability of plasma exosomes concentration in healthy individuals. Also, the stability of exosomes during freeze/thawing cycles was measured in this study. Eleven healthy men were assessed for reliability of exosome concentrations taken over the same sampling period, over 10 hours of a.day a.d over five days. Exosomes were isolated by differential ultracentrifugation a.d characterized by their size distribution a.d morphology by electron microscopy a.d using the Nanosight tracking analysis respectively. Mean plasma exosome values werel.505-2.245 x 10^8/mL）. Within day variability was not significantly different （P = 0.95）, a.d between day variability was not significantly different （P = 0.42）. The interclass correlation coefficients （ICC） of 0.80 for within sample showed good reliability; ICC of 0.84 for between day plasma exosomes concentrations showed good reliability, a.d the within day ICC of 0.70 showed a moderate reliability. There was no significant difference observed in using fresh or frozen plasma in exosome quantification or exosome concentration. The statistics show that there is little significant variability in plasma exosome concentrations taken over a single sampling period, over a.day or between days, which is vital for plasma exosome studies.

Cyclooxygenase-2 expression as a predictor of outcome in colorectal carcinoma

AIM:To correlate cyclooxygenase-2(COX-2)expression profile with clinical a.d pathological variables to assess their prognostic/predictive value in colorectal carcinoma(CRC).METHOdS:Archival tumor samples were analyzed using immunohistochemistry for COX-2 expression in 94 patients with *** were diagnosed a.d treatedat the departments of Surgery a.d Oncology,King a.dulaziz University Hospital,Sa.di ***:Fifty-six percent of the tumors showed positive cytoplasmic COX-2 expression,whereas 44%of cases were completely *** were no significant correlations between COX-2 expression a.d sex,age,grade or tumor ***,COX-2 expression revealed a significant correlation with tumor stage(P=0.01)a.d distant metastasis(P=0.02),a.d a borderline association with lymph node involvement(P =0.07).Tumors with high COX-2 expression showed a higher recurrence rate than tumors with no expression(Pdifference in disease-free survival between COX-2-positive a.d negative tumors in favor of the ***-2 expression did not significantly predict disease-specific survival,which was much shorter for COX-2-positive *** multivariate(COX)models,COX-2 did not appear among the independent predictors of disease-free survival or disease-specific ***:COX-2 expression seems to provide useful prognostic information in CRC,while predicting the patients at high risk for recurrent disease.
