CD4^(+)T cells can"help"or"license" conventional type 1 dendritic cells(cDC1s)to induce CD8^(+)cytotoxic T lymphocyte(CTL)anticancer responses,as proven in mouse *** recently identified cDC1s with a transcriptomic imprint of CD4^(+)T-cell help,specifically in T-cell-infiltrated human cancers,and these cells were associated with a good prognosis and response to PD-1-targeting ***,we delineate the mechanism of cDC1 licensing by CD4^(+)T cells in *** CD4^(+)T cells produce IFNβvia the STING pathway,which promotes MhC-I antigen(cross-)presentation by cDC1s and thereby improves their ability to induce CTL anticancer *** cooperation with CD40 ligand(L),IFNβalso optimizes the costimulatory and other functions of cDC1s required for CTL response ***-I-producing CD4^(+)T cells are present in diverse T-cell-infiltrated cancers and likely deliver“help”signals to CTLs locally,according to their transcriptomic profile and colocalization with“helped/licensed”cDCs and tumor-reactive CD8^(+)T *** agreement with this scenario,the presence of IFN-I-producing CD4^(+)T cells in the TME is associated with overall survival and the response to PD-1 checkpoint blockade in cancer patients.

The purpose of this study is to examine the feasibility and short-term impact of a fully manualized, telephone-delivered intervention for spouse caregivers, Taking Care of her (TCh). A total of 12 study participants from the Pacific NW were enrolled whose wife was diagnosed with Stage III ovarian cancer within 8 months. Feasibility was confirmed by rates of recruitment and retention;the quality of delivery of the intervention by telephone;and through data obtained on program acceptability during follow up exit interviews. Outcomes from the within-group analysis revealed improvements on standardized measures of spouses’ and patients’ depressed mood and anxiety;marital communication about the cancer;caregivers’ skills and confidence to manage the emotional toll of the illness on themselves and wives;and wives’ positive appraisal of spousal support. Study results suggest that the TCh Program has the potential to positively affect both spouse caregiver and patients’ adjustment to recently diagnosed advanced ovarian cancer. Telephone delivery holds promise for sustainability. A future clinical trial with a larger study sample is warranted.

AIM： hepatitis B virus protein X （hBx） has been shown to be weakly oncogenic in vitro. The transforming activities of hBx have been linked with the inhibition of several functions of the tumor suppressor p53. We have studied whether hBx may have different effects on p53 depending on the cell type. METhODS： We used the human hepatoma cell line hepG2 and the immortalized murine hepatocyte line AML12 and analyzed stably transfected clones which expressed physiological amounts of hBx. P53 was induced by UV irradiation.RESULTS： The p53 induction by UV irradiation was unaffected by stable expression of hBx. however, the expression of the cyclin kinase inhibitor p21^waf/cip/sdi which gets activated by p53 was affected in the hBx transformed cell line AML12-hBx9, but not in hepG2. In AML-hBx9 cells, p21^waf/ciP/sdi -protein expression and p21^waf/dip/sdi transcription were deregulated. Furthermore, the process of apoptosis was affected in opposite ways in the two cell lines investigated. While stable expression of hBx enhanced apoptosis induced by UV irradiation in hepG2-cells, apoptosis was decreased in hBx transformed AML12-hBx9. P53 repressed transcription from the hBV enhancer I, when expressed from expression vectors or after induction of endogenous p53 by UV irradiation. Repression by endogenous p53 was partially reversible by stably expressed hBx in both cell lines.CONCLUSION： Stable expression of hBx leads to deregulation of apoptosis induced by UV irradiation depending on the cell line used. In an immortalized hepatocyte line hBx acted anti-apoptotic whereas expression in a carcinoma derived hepatocyte line hBx enhanced apoptosis.

CD74 is a protein whose initial role in antigen presentation was recognized two decades ago. Recent studies have revealed that it has additional functions as a receptor for macrophage migration inhibitory factor and as a receptor for an important human pathogen, helicobacter pylori （h pylon）. The role of CD74 as a receptor is important because after binding of migration inhibitory factor or h pylori, NF-κB and Erkl/2 activation occurs, along with the induction of proinflammatory cytokine secretion. This review provides an up-to-date account of the functions of CD74 and how it might be involved in inflammation and cancer within the gastrointestinal tract.

BACKGROUND Timely differentiation of biliary atresia(BA)from other infantile cholestatic diseases can impact patient ***,non-invasive staging of fibrosis after Kasai hepatoportoenterostomy has not been widely *** wave elastography is an ultrasound modality that detects changes in tissue *** authors propose that the utility of elastography in BA can be elucidated through meta-analysis of existing *** To assess the utility of elastography in:(1)BA diagnosis,and(2)post-Kasai fibrosis *** A literature search identified articles that evaluated elastography for BA diagnosis and for post-Kasai *** studies met criteria for meta-analysis:Eleven for diagnosis and nine for follow-up *** diagnostic odds ratio(DOR),sensitivity,and specificity of elastography were calculated through a random-effects model using Meta-DiSc *** Mean liver stiffness in BA infants at diagnosis was significantly higher than in non-BA,with overall DOR 24.61,sensitivity 83%,and specificity 79%.Post-Kasai,mean liver stiffness was significantly higher in BA patients with varices than in patients without,with DOR 16.36,sensitivity 85%,and specificity 76%.Elastography differentiated stage F4 fibrosis from F0-F3 with DOR of 70.03,sensitivity 96%,and specificity 89%.Elastography also differentiated F3-F4 fibrosis from F0-F2 with DOR of 24.68,sensitivity 85%,and specificity 81%.CONCLUSION Elastography has potential as a non-invasive modality for BA diagnosis and surveillance *** paper’s limitations include inter-study method heterogeneity and small sample ***,standardized,multi-center studies are recommended.

Modeling regional landslide susceptibility using dynamic soil moisture profiles

A landslide susceptibility mapping study was performed using dynamic hillslope hydrology. The modified infinite slope stability model that directly includes vadose zone soil moisture(SM) was applied at Cleveland Corral, California, US and Krishnabhir, Dhading, Nepal. The variable infiltration capacity(VIC-3L) model simulated vadose zone soil moisture and the wetness index hydrologic model simulated groundwater(GW). The GW model predictions had a 75% NASh-Sutcliffe efficiency when compared to California's in-situ GW measurements. The model performed best during the wet season. Using predicted GW and VIC-3L vadose zone SM, the developed landslide susceptibility maps showed very good agreement with mapped landslides at each study region. Previous quasi-dynamic model predictions of Nepal's hazardous areas during extreme rainfall events were enhanced to improve the spatial characterization and provide the timing of hazardous conditions.

h py/ori is probably the most prevalent human pathogen worldwide. Since it was initially suggested in 1983 by Marshall and Warren to be implicated in gastritis and peptic ulcer disease, h pylori has also been implicated in gastric carcinoma and was classified as a class I carcinogen. In the last two decades, a noteworthy body of research has revealed the multiple processes that this gram negative bacterium activates to cause gastroduodenal disease in humans. Most infections are acquired early in life and may persist for the life of the individual. While infected individuals mount an inflammatory response that becomes chronic, along with a detectable adaptive immune response, these responses are ineffective in clearing the infection. hpylori has unique features that allow it to reside within the harsh conditions of the gastric environment, and also to evade the host immune response. In this review, we discuss the various virulence factors expressed by this bacterium and how they interact with the host epithelium to influence pathogenesis.

The gastric mucosa separates the underlying tissue from the vast array of antigens that traffic through the stomach lumen. While the extreme ph of this environment is essential in aiding the activation of enzymes and food digestion, it also renders the gastric epithelium free from bacterial colonization, with the exception of one important human pathogen, hpylori. This bacterium has developed mechanisms to survive the harsh environment of the stomach, actively move through the mucosal layer, attach to the epithelium, evade immune responses, and achieve persistent colonization. While a hallmark of this infection is a marked inflammatory response with the infiltration of various immune cells into the infected gastric mucosa, the host immune response is unable to clear the infection and may actually contribute to the associated pathogenesis. here, we review the host responses involved during infection with hpylori and how they are influenced by this bacterium.
