Effect of arsenic on tolerance mechanisms of two plant growth-promoting bacteria used as biological inoculants

Bacterial ability to colonize the rhizosphere of plants in arsenic（As） contaminated soils is highly important for symbiotic and free-living plant growth-promoting rhizobacteria（PGPR）used as inoculants, since they can contribute to enhance plant As tolerance and limit metalloid uptake by plants. The aim of this work was to study the effect of As on growth,exopolysaccharide（EPS） production, biofilm formation and motility of two strains used as soybean inoculants, Bradyrhizobium japonicum E109 and Azospirillum brasilense Az39. The metabolism of arsenate（As（V）） and arsenite（As（III）） and their removal and/or possible accumulation were also evaluated. The behavior of both bacteria under As treatment was compared and discussed in relation to their potential for colonizing plant rhizosphere with high content of the metalloid. B. japonicum E109 growth was reduced with As（III）concentration from 10 μM while A. brasilense Az39 showed a reduction of growth with As（III） from 500 μM. EPS and biofilm production increased significantly under 25 μM As（III）for both strains. Moreover, this was more notorious for Azospirillum under 500 μM As（III）,where motility was seriously affected. Both bacterial strains showed a similar ability to reduce As（V）. However, Azospirillum was able to oxidize more As（III）（around 53%） than Bradyrhizobium（17%）. In addition, both strains accumulated As in cell biomass. The behavior of Azospirillum under As treatments suggests that this strain would be able to colonize efficiently As contaminated soils. In this way, inoculation with A. brasilense Az39 would positively contribute to promoting growth of different plant species under As treatment.

The quantitative induction of VIN3 by low temperatures is required for PRc2 repression of FLc and promotion of flowering （vernalization） in Arabidopsis. Histone acetylation, a chromatin modification commonly associated with gene transcription, increased on VIN3 chromatin in two spatially and temporally distinct phases in response to low temperatures. During short-term cold exposure, histone H3 acetylation at the transcription start site rapidly increased, implying that it is required for VlN3 induction. Subsequent changes in histone H3 and H4 acetylation occurred following continued VIN3 transcription during prolonged cold exposure. Members of the SAGA-like transcriptional adaptor complex, including the histone acetyltransferase GcNS, which induces expression of the cold acclimation pathway genes, do not regulate VlN3 induction during cold exposure, indicating that the cold acclimation pathway and the cold-induction of VlN3 are regulated by different transcriptional mechanisms. Mutations in the other 11 histone acetyltransferase genes did not affect VlN3 induction. However, nicotinamide, a histone deacetyiase inhibitor, induced VIN3 and altered histone acetylation at the VIN3 locus. VIN3 induction was proportional to the length of nicotinamide treatment, which was associated with an early-flowering phenotype and repression of FLc. However, unlike vernalization, the repression of FLc was independent of VIN3 activity. Nicotinamide treatment did not cause a change in the expression of any genes in the autonomous pathway or members of the PRc2 complex, the well characterized repressors of FLc. Our data suggest that FLc is repressed via a novel pathway involving the SIR2 class of histone deacetylases.

The purpose of this project was to examine the effects of a 20-week training study, which included varied aerobic exercise intensity and identical resistance and flexibility training, on circumference, body mass, hormonal, health/fitness and psychophysical measures. Methods: Thirty-two of 40 recruited overweight middle-aged women were randomized into two training groups: low-intensity (LOW: -40% VO2peak and high-intensity (HIGH: -80% VO2peak), for a 20-week training program. Participants were weighed, measured, underwent VO2peak graded exercise tests, and had blood drawn pre-training and at 10- and 20-weeks. Results: Both groups experienced significant body mass loss at 10- and 20-weeks (LOW: p circumference loss (10-weeks—LOW: waist, hip, and thigh regions (p cant decrease (p concentrations diminished significantly in both groups (LOW p correlated to the body mass/ anthropometric changes. Discussion: Both low- and high-intensity produced significant loss in body mass and circumference measurements suggesting that it is not the intensity of exercise, but rather overall energy expenditure that should be considered. The advantage of high-intensity aerobic exercise is that it requires less time to produce the desired energy expenditure. The changes in ghrelin and leptin may have contributed to the energy intake during laboratory trials.

Misfoldedα-synudein is a major component of Lewy bodies,which are a hallmark of Parkinson's disease(PD).A large body of evidence shows thatα-synuclein can aggregate into amyloid fibrils,but the relationship between a-synuclein self-assembly and Lewy body formation remains ***,we show,both in vitro and in a caenorhabditis elegans model of PD,thatα-synuclein undergoes liquid-liquid phase separation by forming a liquid droplet state,which converts into an amyloid-rich hydrogel with Lewy-body-like *** maturation process towards the amyloid state is delayed in the presence of model synaptic vesicles in *** together,these results suggest that the formation of Lewy bodies may be linked to the arrested maturation ofα-synuclein condensates in the presence of lipids and other cellular components.

Sodium hypochlorite (NaOcl) or bleach, found in effluent from wastewater treatment plants, can act as an environmental toxin. The sea urchin Arbacia punctulata is a common subject of embryological toxicity tests due to its sensitivity to environmental pollutants. Using concentrations of NaOcl that mimic those found in treated wastewater (0.11 ppm, 0.06 ppm, and 0.03 ppm) we observed minimal affects on early larval development, though most larvae took longer to develop at higher NaOcl concentration. There was a significant difference in the percentage of non-normal plutei based on concentration (P = 0.038) and significant interaction between the percent of each morphology and NaOcl concentration (P = 0.0027). The most significant change in non-normal plutei was in the retarded (shortened skeletal rods) malformation which increased in frequency with NaOcl concentration (P = 0.001). There was a significant reduction in skeletal length in both normal and retarded plutei (P cl increased.

The adsorption capacity of hydrogels derived from modified xylan hemicellulose has been tested in order to develop new bio-based adsorbent materials useful for removing metal ions pollutants,such as cd(Ⅱ),cu(Ⅱ)and Pb(Ⅱ)from an aqueous *** was extracted from bleached kraft pulp of eucalyptus and subsequently modified with different proportions of functional sulfonic acid groups(HA3-HA7)and sulfonate groups(HS30-HS70)to generate *** results showed that all the synthesized hydrogels were capable of adsorbing metal ions,being the hydrogels with 30%and 50%xylan the ones that presented the highest adsorption *** capacity studies at different initial concentrations revealed that at an initial concentration of 300 mg·L-1,the HA3 hydrogel presented an adsorption capacity of 193mg Pb(Ⅱ),182 mg cd(Ⅱ),and 66 mg cu(Ⅱ)per g *** HA5 hydrogel presented a capacity of 185 mg Pb(Ⅱ),113 mg cd(Ⅱ),and 48 mg cu(Ⅱ)per g *** HS30 hydrogel exhibited an adsorption of 205 mg Pb(Ⅱ),174 mg cd(Ⅱ),and 71 mg cu(Ⅱ)per g hydrogel,and HS50 hydrogel exhibited an adsorption capacity of 273 mg Pb(Ⅱ),143 mg cd(Ⅱ),and 45 mg cu(Ⅱ)per g *** results show that modified Xylan hemicellulose is a promising adsorbent for removal cd(Ⅱ),cu(Ⅱ),and Pb(Ⅱ)ions from aqueous solutions.

Brain injuries in sports drew more and more public attentions in recent years. Brain injuries vary by name, type, and severity in the athletic setting. It should be noted, however, that these injuries are not isolated to only the athletic arena, as non-athletic mechanisms （e.g., motor vehicle accidents） are more common causes of traumatic brain injuries （TBI） among teenagers. Notwithstanding, as many as 1.6 to 3.8 million TBI result from sports and recreation each year in the United States alone. These injuries are extremely costly to the global health care system, and make TBI among the most expensive conditions to treat in children. This article serves to define common brain injuries in sport; describe their prevalence, what happens to the brain following injury, how to recognize and manage these injuries, and what you can expect as the athlete recovers. Some return-to-activity considerations for the brain-injured athlete will also be discussed.
